The survival of Leptosphaeria maculans , which causes phoma stem canker (blackleg), on oilseed rape residues ( Brassica napus ) in South Australia was investigated. Using a quantitative polymerase chain reaction (PCR) assay for L. maculans DNA, the pathogen was mainly detected in the upper 5 cm of the soil profile, including residues on the soil surface. As the size of organic matter particles in the soil decreased, so did the quantity of L. maculans detected in them. To obtain representative data for a field, at least 30 subsamples needed to be collected over the 0·81 ha area studied. In a survey of 49 commercial fields in South Australia, most L. maculans was detected in fields 1 year after oilseed rape had been grown, with less detected after 2 years and negligible amounts 3 years or more after cropping. The diagnostic DNA-based assay for L. maculans reduced the time and cost of studying L. maculans survival in soil and increased the sensitivity and accuracy of results compared with estimates of propagule number of colony-forming units on a semiselective medium.  相似文献

As a first step in the development of a mycoherbicide for biological control of Cirsium arvense indigenous fungal pathogens that had been isolated from diseased hosts were tested both singly and in combination on potted plants under environmental conditions. Disease symptoms (necroses, chloroses and macerations) and parameters of growth and development were evaluated to determine the potential for weed control. During 1998, Phoma destructiva, Phoma hedericola, a Mycelia sterilia and Puccinia punctiformis were inoculated singly. With the exception of the obligate biotroph P. punctiformis (local infections), the single inoculations reduced the measured parameters. A combined inoculum of P. punctiformis and Ph. hedericola was less effective than Ph. hedericola alone. In 1999, Ph. hedericola, Ph. destructiva, Ph. nebulosa and the Mycelia sterila were applied both singly and as a combined inoculum. Of all the tested inocula, the mixture of four pathogens had the greatest potential for biocontrol, significantly reducing reproductive capacity of the plants. These results demonstrate the importance of taking advantage of synergisms in developing a mycoherbicide.  相似文献

【目的】明确2018年起在重庆万州柑橘种植区新发生病害的病原菌,筛选有效防控药剂,为田间防治该病害和后续深入研究提供理论参考。【方法】通过组织分离法从患病样品中获得菌株,进行病原菌形态学、分子生物学和致病性鉴定,采用生长速率法测定不同药剂对目标菌株的毒力。【结果】此病害的病原菌为柑橘轮斑病菌Pseudofabraea citricarpa。病原菌对万州地区主栽的8个品种柑橘的叶、枝、果和昭通苹果及砀山酥梨果实具有致病性。苯醚甲环唑、咪酰胺、甲基硫菌灵等17种杀菌剂的EC50值小于3 mg/kg,代森锰锌与啶酰菌胺1:64比列复配、克菌丹与苯醚甲环唑1:1比例复配、代森锰锌和矿物油1:50比例复配对菌株生长抑制效果良好。【结论】重庆万州柑橘种植园区发生柑橘轮斑病,病原菌对柑橘枝、叶、果和其它果树果实都具有致病性。17种药剂单用或复配对病原菌生长有抑制作用,可进一步用于田间实验。  相似文献

【目的】 柑橘轮斑病(citrus target spot)作为一种新发柑橘病害,造成发病果园严重的经济损失。本研究针对该病害进行适生区预测及风险分析,以便对该病采取及时、有效的管控措施,最终达到降低其流行风险等级,防止病害传播扩展的目的。【方法】 基于环境变量数据和柑橘轮斑病发生分布数据,运用MaxEnt生态位模型模拟预测柑橘轮斑病菌(Pseudofabraea citricarpa)在中国的潜在适生区分布。并通过ROC(receiver operating characteristic)曲线下面积(area under the curve,AUC)评估预测模型的精度,运用正规化训练增益刀切法(regularized training gain)获取气候因子与分布概率间的关系。同时采用有害生物风险分析理论,以有害生物风险分析的规定程序为依据探索柑橘轮斑病病害的风险分析体系和评价值的计算方法,对评价指标进行定性分析,进而量化评价值。在建立综合评价模型的基础上,计算柑橘轮斑病风险性危害值,最后对病害的风险性危害值进行评价。【结果】 柑橘轮斑病菌MaxEnt模型预测结果的平均AUC值为0.998,表明预测结果精度高。柑橘轮斑病菌的潜在适生区面积约占全国面积12.19%,高适生区、中适生区、低适生区各占全国面积约2.85%、3.99%、5.35%。高、中适生区主要集中于长江中上游柑橘优势区及其周边。其中,高适生区主要集中在四川、重庆、陕西南部,以及贵州、湖北等少量地区。中、低适生区是高适生区的外围扩展。通过MaxEnt模型正规化训练增益刀切法获取的环境变量重要性分析结果表明,最冷季度平均温度(Bio11)、最干季度平均温度(Bio9)、最冷月最低温(Bio6)是影响柑橘轮斑病菌分布的3个关键环境因子,这意味着低温、干冷季节柑橘轮斑病发生可能性大。风险分析最终创建出5个准则层、13个指标层的多指标综合评价体系,并对各指标层定量与定性分析,柑橘轮斑病在我国的风险性危害值(R值)为2.08,处于高度风险等级,对长江中上游及湖北西部-湖南西部两大柑橘产区的潜在危害最大。【结论】 柑橘轮斑病风险性较高,需要尽快建立监测体系,针对病害采取有效控制措施,阻止病害在长江中上游柑橘优势区及相邻柑橘产区传播。  相似文献

为了解我国油菜黑胫病病原菌(Leptosphaeria biglobosa)的生物学特性,在实验室里研究了温度、pH值、光照、碳源及氮源等条件对该病菌营养生长及分生孢子产生的影响。结果表明:病原菌在5℃~35℃均可进行营养生长并产孢,最适生长和产孢温度为25℃;在pH4~10的范围内该菌均能生长并产孢,在pH7的条件下生长最快,产孢最多。光暗交替有利于菌丝生长和产孢,而持续光照不利于菌丝生长,尤其不利于产孢。最适的碳源、氮源分别是可溶性淀粉、牛肉膏。分生孢子的致死温度为52℃。  相似文献

Potato seed tubers are imported annually to Israel from northern Europe. Although the seed is registered as certified, a survey carried out over a 9-year period indicated that most lots were affected by latent or active bacterial and fungal infections. Latent infection byErwinia carotovora subsp.atroseptica, the causal agent of blackleg, at a level of 103 cells/g peel, was present in 30% of the lots in most years. Black scurf caused byRhizoctonia solani was present in 20–70% of the imported lots, with a moderate to high level of infection in all years except 1985. In contrast, although many lots were affected by powdery scab, common scab, and Fusarium dry rot in most years, disease incidence within lots was generally low. The gangrene pathogen (Phoma exigua) was rarely detected. The survey findings are of marked importance, due to the extensive use of soil fumigation in Israeli agriculture.  相似文献

Resistance of Brassica napus (oilseed rape, canola) conferred by three different major resistance genes has been overcome by changes in virulence of Leptosphaeria maculans populations in France and Australia. In South Australia where B. napus cultivars with major gene resistance derived from Brassica rapa ssp. sylvestris were grown extensively, resistance was rendered ineffective within 3 years of commercial release of the cultivar. Disease severity was higher on cultivars with sylvestris-derived resistance than cultivars with polygenic resistance. This Australian situation is compared to that in France, where resistance conferred by the Rlm1 gene was overcome nation-wide in 5 years under commercial cropping practices, and also where a source of resistance introgressed into B. napus from B. juncea was rendered inefficient in 3 years in experimental field plots near Rennes.  相似文献

The aim of this study was to isolate, identify and characterize ascochyta blight pathogens from Cicer judaicum , a wild annual Cicer species which grows in Israel and other Mediterranean countries in sympatric distribution with legume crops, and determine their virulence and aggressiveness to other wild and domesticated legumes. Native C. judaicum plants exhibited symptoms resembling ascochyta diseases of grain legume crops. Two distinct pathogens were isolated and identified as Phoma pinodella and Didymella rabiei using morphological and molecular tools; their infectivity was verified using Koch's postulates. The virulence of these pathogens was examined on 13 legume species, of which P. pinodella was virulent to Pisum sativum , P. fulvum , C. judaicum , C. arietinum , C. reticulatum , C. pinnatifidum and C. bijugum . Didymella rabiei infected all these Cicer species, but not the other legume species tested. Aggressiveness of the pathogens was tested on wild and domesticated chickpea and pea. Didymella rabiei isolated from C. judaicum had significantly higher ( P  < 0·001) aggressiveness than P. pinodella from C. judaicum on both wild and domesticated chickpea. Disease severity on the former species ranged from 62·5% to 70% and on the latter from 41% to 56%. Phoma pinodella isolates from C. judaicum were more aggressive on C. arietinum and P. sativum than on C. judaicum and P. fulvum . Results of the current study suggest that C. judaicum may serve as an alternative host to ascochyta pathogens that endanger chickpea and possibly other crops and wild species growing in close proximity.  相似文献
