用高效液相色谱法测定油茶肉质果、肉质叶维生素C的含量.采用YWG-C18(10μm,4.6mm×250mm i.d.)色谱柱,以0.02 M醋酸钠缓冲液-甲醇为流动相,在254 nm波长下检测.回收率为:94.8%~101.2%.相对标准偏差小于2%.  相似文献

[目的]研究毕赤酵母菌株(Pichia GS115)表达人C型溶菌酶(H-LYZ)的发酵与分离纯化的工艺.[方法]探讨酵母生长规律,研究不同菌体浓度、不同诱导时间对蛋白产量的影响,并且探讨不同pH条件对DEAE-SepharseFF和CM-Sepharose FF层析分离纯化目标蛋白的影响.[结果]毕赤酵母菌株表达H-LYZ的最佳发酵条件为:发酵24h用浓度1%甲醇诱导,以后每12 h用浓度1%甲醇诱导,共6次,在此条件下可获得高产量目标蛋白.DEAE-Sepharose FF和CM-Sepharose FF分离纯化目标蛋白的最佳pH分别为6.65和6.50.[结论]该研究可为人C型溶菌酶的工艺化生产提供理论指导.  相似文献

Shuang-Huang-Lian (SHL), a traditional Chinese formula containing Lonicerae japonicae flos (LJF), Scutellariae radix (SR) and Forsythiae fructus (FF), is commonly used to treat acute upper respiratory tract infection, acute bronchitis and light pneumonia. Forsythoside A is one of the main active ingredients in Forsythiae fructus, a key herb in SHL. In the present study, effects of different compositions in SHL on the intestinal absorption of forsythoside A were investigated. The observations from in situ intestinal circulation model showed that A/%(h− 1) of forsythoside A in FF + LSF, FF + SR and SHL were all reduced greatly compared with that in FF. However, in pharmacokinetics study, Cmax and AUC0 → 1440 of forsythoside A all increased and T1/2 prolonged in SHL, FF + LJF and FF + SR compared with FF. The results indicated that the different compositions of SHL decreased absorption but increased bioavailability of forsythoside A, which may be related to its metabolism inhibited in intestine or liver.  相似文献

采用乙酸乙酯提取目标物,正己烷脱脂,微波辅助衍生化,建立了能同时测定水产品中氯霉素、甲砜霉素和氟甲砜霉素残留量的毛细管电子捕获气相色谱法(GC—ECD)。加标回收实验结果显示,氯霉素在1.0-200.0μg/L范围内、甲砜霉素和氟甲砜霉素在5.0~200.0μg/L范围内成良好的线性,相关系数均大于0.996;氯霉素、甲砜霉素和氟甲砜霉素的检出限分别为0.1、0.2和0.2μg/kg,回收率在79.2%-88.0%之间,相对标准偏差在2.0%-4.1%之间。表明该方法定性定量准确、检出限低、重现性好、省时省力,可以满足水产品中3种氯霉素类药物的多残留安全检测。  相似文献

The effect of thermal processing on the degradation of the phytochemicals in black rice flour by means of fluorescence spectroscopy and degradation kinetics was investigated. In order to investigate the influence of food matrices, a comparative analysis between integral rice flour and different fractions was performed. The preliminary compositional results suggested a higher content in phytochemicals in fraction four of the seven fractions of black rice flour, which was sifting through a sieve with a diameter of 180 μm. The compositional complexity was highlighted by fluorescence spectroscopy. The heat-treatment caused structural changes that led to red- or blue-shifts in maximum emission. The first-order kinetic model was used to describe the mechanism of degradation. The activation energies were 10.07 ± 1.04 kJ/mol for total polyphenolic, 7.26 ± 0.58 kJ/mol for total monomeric anthocyanins and 6.71 ± 1.12 kJ/mol for antioxidant activity in case of integral flour extract. For fraction four extract obtained by, the Ea values were: 3.51 ± 0.53 kJ/mol, 11.49 ± 1.47 kJ/mol, 15.80 ± 1.50 kJ/mol and 19.91 ± 3.27 kJ/mol, respectively. The calculated values of the activation energy revealed higher temperature dependence of total polyphenols in integral flour and of antioxidant activity in fraction four, respectively.  相似文献

建立了高效液相色谱荧光检测法同时检测鸡蛋中氟苯尼考及其代谢物氟苯尼考胺残留的方法.鸡蛋经碱性乙酸乙酯提取,饱和正己烷脱脂,氮吹仪吹干浓缩后用流动相溶解.用Lichrospher C1s柱,以乙腈-磷酸二氢钠溶液(0.01 mol/L,含0.005 mol/L十二烷基硫酸钠和0.1%三乙胺)(35:65,v/v)为流动相,在激发波长224 nm、发射波长285 nm处用荧光检测器检测.氟苯尼考在0.01~10.0 mg/L、氟苯尼考胺在0.002 5~2.5 mg/L浓度范围内,本方法线性关系良好,相关系数分别为0.99 97和0.999 8.当添加水平氟苯尼考为15~500μg/kg、氟苯尼考胺为5~500 μg/kg时,该方法平均回收率分别为87.41%~92.29%和89.01%~95.15%,相对标准偏差分别为3.51%~5.36%和4.28%~5.72%;检测限分别为1.5 μg/kg和0.5 μg/kg(S/N=3);定量限分别为5μg/kg和2 μg/kg(S/N=10).该方法简便、快速、灵敏度高,可同时检测鸡蛋中氟苯尼考和氟苯尼考胺的残留.  相似文献

通过田间小区试验,采用Biolog FF微平板培养方法,研究小麦与蚕豆间作对小麦和蚕豆根际真菌代谢功能多样性的影响。结果表明:与单作相比,间作明显提高了小麦和蚕豆根际真菌的碳源平均利用率(AWCD),且间作提高蚕豆根际真菌活性的幅度大于小麦。与单作相比,间作显著提高了蚕豆和小麦根际真菌对Biolog FF板中聚合物、糖类、羧酸类、氨基酸类和胺类碳源的利用;同时间作使小麦和蚕豆根际真菌对碳源的总利用强度比单作分别显著提高63.30%和52.02%,小麦和蚕豆根际真菌对Biolog FF板中6类碳源的利用强度百分比以糖类、羧酸类和氨基酸最高,分别为36.66%~45.99%、25.65%~27.70%和16.37%~20.67%。小麦与蚕豆间作显著提高了小麦和蚕豆根际真菌的香农多样性指数(H)和丰富度指数(S)。主成分分析表明,间作明显改变了小麦和蚕豆根际真菌的群落结构。小麦与间作蚕豆明显促进了小麦和蚕豆根际土壤真菌的碳源代谢强度,显著提高了根际真菌的多样性和丰富度,改变了根际真菌的群落结构,因而是一种有利于改善蚕豆连作栽培根际微生态环境的有效措施。  相似文献

In the present study, effects of different compositions in SHL on the in vitro metabolism in Sprague–Dawley rat liver microsomes of forsythoside A were investigated. The observations from Sprague–Dawley rat liver microsomes in the presence of β-NADPH or UDPGA that forsythoside A may be the substrates of CYP3A4, CYP2C9, CYP1A2, UGT1A6, UGT1A3, UGT1A1 and UGT1A9; Chlorogenic acid may be the substrates of CYP3A4, CYP2C9, CYP1A2, CYP2C19, UGT1A6, UGT1A3 and UGT1A1; Baicalin may be the substrates of CYP3A4, CYP2C19, CYP1A2, UGT1A9, UGT1A1 and UGT1A3; Baicalein may be the substrates of CYP3A4, CYP2E1 and UGT1A6. It was also found that the residue of forsythoside A in SHL, FF + LJF and FF + SR was greatly increased compared with that in FF in Sprague–Dawley rat liver microsomes in the presence of β-NADPH or UDPGA, which indicated that the metabolism of forsythoside A in SHL may be influenced by chlorogenic acid in LJF acting on the CYP3A4, CYP2C9, CYP1A2, UGT1A6, UGT1A3 and UGT1A1; baicalin in SR acting on the CYP3A4, CYP1A2, UGT1A9, UGT1A1 and UGT1A3; baicalein acting on the CYP3A4 and UGT1A6 respectively.  相似文献

研究了亚麻纤维(flaxfiber,FF)的含量和马来酸酐接枝聚丙烯(MAH-g-PP)对聚丙烯/亚麻纤维(PP/FF)复合材料各项性能的影响,通过扫描电镜研究了复合材料的界面结构.结果表明:随着FF含量增大,复合材料拉伸强度、硬度和热变形温度提高;添加MAH-g-PP提高了复合材料力学性能和热稳定性,降低了复合材料吸水性.  相似文献
