Meats need to be heated to inactivate foodborne pathogens such as Escherichia coli O157:H7. High-temperature treatment used to prepare well-done meats increases the formation of carcinogenic heterocyclic amines (HCAs). We evaluated the ability of plant extracts, spices, and essential oils to simultaneously inactivate E. coli O157:H7 and suppress HCA formation in heated hamburger patties. Ground beef with added antimicrobials was inoculated with E. coli O157:H7 (10(7) CFU/g). Patties were cooked to reach 45 °C at the geometric center, flipped, and cooked for 5 min. Samples were then taken for microbiological and mass spectrometry analysis of HCAs. Some compounds were inhibitory only against E. coli or HCA formation, while some others inhibited both. Addition of 5% olive or apple skin extracts reduced E. coli O157:H7 populations to below the detection limit and by 1.6 log CFU/g, respectively. Similarly, 1% lemongrass oil reduced E. coli O157:H7 to below detection limits, while clove bud oil reduced the pathogen by 1.6 log CFU/g. The major heterocyclic amines 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) were concurrently reduced with the addition of olive extract by 79.5% and 84.3% and with apple extract by 76.1% and 82.1%, respectively. Similar results were observed with clove bud oil: MeIQx and PhIP were reduced by 35% and 52.1%, respectively. Addition of onion powder decreased formation of PhIP by 94.3%. These results suggest that edible natural plant compounds have the potential to prevent foodborne infections as well as carcinogenesis in humans consuming heat-processed meat products.  相似文献

Two trials were conducted to determine the effects of dietary enrichments with the microalga Parietochloris incisa , rich in arachidonic acid (ARA), on stress resistance in guppies Poecilia reticulata . The microalga was added to commercial diets as a neutral lipid (NL) extract and its fractions or as broken cells. Experimental diets were applied for a period of 14 days. In trial 1, commercial diets were supplemented with NL (containing 25 mg ARA and 0.11 mg β-carotene g−1 feed), its triacylglycerol (TAG) fraction (containing 25 mg ARA g−1 feed and no β-carotene) and the β-carotene fraction (containing 0.11 mg carotenoid g−1 feed and minute amounts of ARA). Neutral lipid-fed fish demonstrated the highest resistance ( P <0.05) to osmotic stress (32-ppt NaCl), followed by fish fed with diets supplemented with TAG and β-carotene alone, which were more resistant than control ( P <0.05). In trial 2, fish fed diets supplemented with higher levels of broken alga (26.1 mg ARA g−1 feed) were more resistant ( P <0.05) to stress as compared with fish fed lower ARA (16.3 mg g g−1) or an unsupplemented control diet. We suggest a dietary supplementation with broken P. incisa cells to enhance stress resistance in guppies before a stressful event.  相似文献

The occurrence of nests of the pine processionary caterpillar (PPC),Thaumetopoea wilkinsoni Tarns (Lepidoptera: Thaumetopoeidae), was recorded in two 9-year-old plots planted withPinus halepensis, P. brutia andP. eldarica in a randomized block design. Nest density was highest inP. eldarica and lowest inP. halepensis. The mean number of nests per tree and their frequency distribution indicate that the preference of the PPC is in the order P.eldarica >P brutia >P. halepensis;it is believed to be related to shape of the tree. Planting ofP. eldarica in camping sites is not recommended, due to its high susceptibility to colonization by the PPC.  相似文献

International provenance trials of Pinus halepensis and related species, P. brutia and P. eldarica, allowed study of the relationship between seed origin of P. halepensis and susceptibility to the Israeli pine bast scale (Matsucoccus josephi, Homoptera: Margarodidae), and the infestation of P. brutia and P. eldarica by this pest. The Greek and Israeli provenances displayed a lower susceptibility to attacks by the pest than did the North African and Spanish provenances. The lower susceptibility is expressed by a higher percentage of unaffected trees, low rate of injury and low density of Matsucoccus ovisacs. Two different mechanisms may be responsible for the relative resistance of P. halepensis of the Greek and the Israeli populations. It is possible that resistance to the pest was transferred from P. brutia to P. halepensis in Greece, where the natural distribution of these pine species overlaps or overlapped. On the other hand, the relative resistance of the Israeli provenances may be due to long coexistence in Israel of M. josephi and P. halepensis.  相似文献

The Eucalyptus gall waspOphelimus maskelli (Ashmead) (Hymenoptera: Chalcidoidea: Eulophidae: Ophelimini) and its introduced parasitoidClosterocerus chamaeleon (Hymenoptera: Chalcidoidea: Eulophidae: Entedoninae) are recorded for the first time in the Mediterranean and Aegean Regions of Turkey. Some morphological characters of adult and larva ofO. maskelli and larva ofC. chamaeleon are described and illustrated. Distributions of the two wasps collected in Turkey are given. Results indicate that the biological control agent,C. chamaeleon, has spread over 1,300 km in the 16 months since its initial release.  相似文献

We investigated antitumor effects of the following four extracts of freeze-dried Hericium erinaceus mushrooms in Balb/c mice intracutaneously transplanted on the backs with CT-26 colon cancer cells: HWE, hot water extraction by boiling in water for 3 h; MWE, microwaving in 50% ethanol/water at 60 W for 3 min; and ACE and AKE, boiling in 1% HCl or 3% NaOH for 2 h. HWE and MWE with a higher content of β-glucans, determined by an assay kit, than ACE and MKE were active in all bioassays. Gas chromatography/mass spectrometry analyses showed the presence of 40, 27, 16, and 13 compounds, respectively, in the four extracts. Daily intraperitoneal (ip) injections of HWE and MWE for 2 weeks significantly reduced tumor weights by 38 and 41%. Tumor regressions were associated with changes in the following cancer biomarkers as compared to phosphate buffer (PBS)-treated control mice: 2.7- and 2.4-fold increases in cytolytic activity of splenic natural killer (NK) cells; restored nitric oxide production and phagocytosis in peritoneal macrophages to 95-98% of normal levels; ~2-fold increase in released pro-inflammatory cytokines tumor necrosis factor-α, interleukin-1β, and interleukin-6 from macrophages; and ~56 and ~60% reductions in the number of blood vessels inside the tumor. The pro-angiogenic factors vascular endothelial growth factor (VEGF), cyclooxygenase 2 (COX-2), and 5-lipoxygenase (5-LOX) were also significantly reduced in mRNA and protein expression by tumor genes. Enzyme-linked immunosorbent assay of tumor cells confirmed reduced expression of COX-2 and 5-LOX (32 and 31%). Reduced COX-2 and 5-LOX expression down-regulated VEGF expression, resulting in inhibition of neo-angiogenesis inside the tumors. The results indicate that induction of NK activity, activation of macrophages, and inhibition of angiogenesis all contribute to the mechanism of reduction of tumor size.  相似文献

Molecular dynamics simulations were performed to study the interactions of bioactive catechins (flavonoids) commonly found in green tea with lipid bilayers, as a model for cell membranes. Previously, multiple experimental studies rationalized catechin's anticarcinogenic, antibacterial, and other beneficial effects in terms of physicochemical molecular interactions with the cell membranes. To contribute toward understanding the molecular role of catechins on the structure of cell membranes, we present simulation results for seven green tea catechins in lipid bilayer systems representative of HepG2 cancer cells. Our simulations show that the seven tea catechins evaluated have a strong affinity for the lipid bilayer via hydrogen bonding to the bilayer surface, with some of the smaller catechins able to penetrate underneath the surface. Epigallocatechin-gallate (EGCG) showed the strongest interaction with the lipid bilayer based on the number of hydrogen bonds formed with lipid headgroups. The simulations also provide insight into the functional characteristics of the catechins that distinguish them as effective compounds to potentially alter the lipid bilayer properties. The results on the hydrogen-bonding effects, described here for the first time, may contribute to a better understanding of proposed multiple molecular mechanisms of the action of catechins in microorganisms, cancer cells, and tissues.  相似文献

As part of an effort to improve plant-derived foods such as potatoes, eggplants, and tomatoes, the antiproliferative activities against human colon (HT29) and liver (HepG2) cancer cells of a series of structurally related individual compounds were examined using a microculture tetrazolium (MTT) assay. The objective was to assess the roles of the carbohydrate side chain and aglycon part of Solanum glycosides in influencing inhibitory activities of these compounds. Evaluations were carried out with four concentrations each (0.1, 1, 10, and 100 microg/mL) of the the potato trisaccharide glycoalkaloids alpha-chaconine and alpha-solanine; the disaccharides beta(1)-chaconine, beta(2)-chaconine, and beta(2)-solanine; the monosaccharide gamma-chaconine and their common aglycon solanidine; the tetrasaccharide potato glycoalkaloid dehydrocommersonine; the potato aglycon demissidine; the tetrasaccharide tomato glycoalkaloid alpha-tomatine, the trisaccharide beta(1)-tomatine, the disaccharide gamma-tomatine, the monosaccharide delta-tomatine, and their common aglycon tomatidine; the eggplant glycoalkaloids solamargine and solasonine and their common aglycon solasodine; and the nonsteroidal alkaloid jervine. All compounds were active in the assay, with the glycoalkaloids being the most active and the hydrolysis products less so. The effectiveness against the liver cells was greater than against the colon cells. Potencies of alpha-tomatine and alpha-chaconine at a concentration of 1 microg/mL against the liver carcinoma cells were higher than those observed with the anticancer drugs doxorubicin and camptothecin. Because alpha-chaconine, alpha-solanine, and alpha-tomatine also inhibited normal human liver HeLa (Chang) cells, safety considerations should guide the use of these compounds as preventative or therapeutic treatments against carcinomas.  相似文献
