Classical studies of mammalian movement control define a prominent role for the primary motor cortex. Investigating the mouse whisker system, we found an additional and equally direct pathway for cortical motor control driven by the primary somatosensory cortex. Whereas activity in primary motor cortex directly evokes exploratory whisker protraction, primary somatosensory cortex directly drives whisker retraction, providing a rapid negative feedback signal for sensorimotor integration. Motor control by sensory cortex suggests the need to reevaluate the functional organization of cortical maps.  相似文献

In this study we present the first report on partial amplification, sequencing and phylogenetic relationship of VP2 of the Indian isolate BTV-2. A PCR product of 1135 bp was amplified, cloned and sequenced. About 1063 bp of partial VP2 gene (1792-2854 bp region) of the Indian isolate was subjected to sequence analysis with already published sequences available in the genome database. The percent similarity of 85.2 was observed with Taiwan isolate and 59% with other isolates of BTV-2. However, 46.2% similarity with Australian BTV-1 and no significant similarity were noted with other serotypes. In-silico analysis and restriction enzyme digestion confirmed the presence of conserved SalI site at 2380 bp position in both Indian and Taiwan isolates. Phylogenetic analysis showed that all BTV-2 isolates formed one distinct group in which BTV-2 Indian and Taiwan isolate is more closely related and further demonstrated that BTV’s of the same serotype from different geographical regions were closely related at nucleotide and amino acid level, respectively.  相似文献

  1. To our knowledge, this is the first inclusive assessment of the status of Sparidae fisheries in the territorial waters of Kuwait, integrating information on catch trends, consumer preferences, recreational fishing behaviour, and molecular based identification of fish species by DNA barcoding
  2. Fisheries landing data were obtained from the official fisheries bulletin released by the Kuwait Central Statistical Bureau. Surveys were conducted to assess consumer preferences on seabream species, landed seabream species, and the behaviour of recreational anglers towards seabream species. DNA barcodes were generated to authenticate commercial seabream using the mitochondrial cytochrome c oxidase subunit I (COXI) gene and then compared with GenBank sequence entries; these sequences were then used to construct a neighbour-joining (NJ) phylogenetic tree.
  3. Yellowfin seabream Acanthopagrus latus was the prime and most favoured commercial seabream species (45%), followed by Sparidentex hasta, which was the top-rated secondary, unassessed seabream species. Approximately 54% of households conduct recreational fishing more than twice a month, and most (77%) primarily target S. hasta and Ac. latus. Consumer demand for secondary seabreams, which were not assessed and are listed in the bulletin under the category ‘others’, was evident.
  4. The NJ phylogenetic tree revealed that Arabian Gulf seabreams, including those of Kuwait, are genetically different from their counterparts inhabiting neighbouring waters.
  5. The data presented here highlight the urgency to modify the existing fisheries list using accurate identification tools, such as DNA barcoding, for the sustainable and conservation-oriented management of local fisheries, which are in decline.

Porcine reproductive and respiratory syndrome (PRRS) is a chronic viral disease of pigs caused by PRRS virus (PRRSV). The PRRSV VR2332 is the prototype North American parental strain commonly used in the preparation of vaccines. Goal of this study was to understand missing information on VR2332 induced immune modulation at the lungs and lymphoid tissues, the sites of PRRSV replication. Pigs were infected intranasally and samples collected at post-infection day (PID) 15, 30, and 60. Microscopically, lungs had moderate interstitial pneumonia, and the virus was detected in all the tested tissues. Peak antibody response and the cytokine IFN-γ secretion were detected at PID 30, with increased TGF-β until PID 60. Population of CD8+, CD4+, and CD4+CD8+T cells, Natural killer (NK) cells, and γδ T cells in the lungs and lymphoid tissues were significantly modulated favoring PRRSV persistence. The NK cell-mediated cytotoxicity was significantly reduced in infected pigs. In addition, increased population of immunosuppressive T-regulatory cells (Tregs) and associated cytokines were also observed in VR2332 strain infected pigs.  相似文献

Cytoplasmic male sterile system in ridge gourd has been converted to cytoplasmic genetic male sterile (CGMS) system through the development of analogues of male sterile (MS) line, maintainer line and fertility restorer line. These lines were developed by crossing the MS mutant, regenerated through in vitro culture, with monoecious pollen parents Deepthi, Haritham, LA 101, CO 2, IC 92761 and IC 92685. All hybrids and the BC1 generation developed by crossing with the recurring pollen parents Deepthi, Haritham and LA 101 were male sterile. Male sterile BC1 plants have been advanced to BC6 generation and the parental line LA 101 was proved to be a successful maintainer line, producing male sterile progeny in successive back cross generations. Analogue of cytoplasmic male sterile line, MS LA 101, was developed through back crossing and on crossing with fertility restorer lines Arka Sumeet and LA 102, this line excelled as female parent, resulting heterotic combinations. Mitochondrial marker rpS14 and SCAR Tm-53 were identified to yield male sterility specific markers whereas SSR marker 18956 has generated the male fertility specific marker. These primers are recommended for marker assisted selection of ridge gourd, for utilizing male sterility for hybrid seed production and for developing A, B and C lines in CGMS system.  相似文献

A 7-month-old, 180-kg, female foal was presented with open diaphyseal fracture of the left third metacarpal bone. The fractured limb was stabilized preoperatively with external coaptation. Open reduction and internal fixation were done using 4.5-mm broad dynamic compression plate, which was applied in a neutralization manner. Postoperatively, external support with padded bandage reinforced with gutter splint made of fiber glass was provided. The incisional infection and the contaminated wound on medial aspect of the metacarpal were managed with regular dressing. Complete radiographic union and functional recovery were noticed by 4 months postoperatively. Open diaphyseal fractures can be managed by proper preoperative fracture stabilization, wound management, and fixation methods using bone plate and external coaptation.  相似文献

Live attenuated homologous vaccine against peste des petits ruminants of sheep and goats was produced on a large scale basis in roller culture bottles using seed virus developed at the Indian Veterinary Research Institute, Muktheswar, India. Vero cells between 130–150 passages with six percent foetal calf serum were used for the production of vaccine. The cells were infected with 0.01 multiplicity of infection and harvested when the cytopathic effect was 80%. The vaccine was freezedried in order to maintain the stability of the vaccine. Identity test and titration was performed and the vaccine titre was monitored to be minimum of 105/100 doses. In-house sterility tests and quality control tests using experimental animals and small ruminants were performed. The vacuum and moisture content of the vaccine were also regulated to be within the normal limits.  相似文献

Sterility mosaic disease (SMD), considered as the “green plague of pigeonpea” and caused by pigeonpea sterility mosaic virus (PPSMV) is one of the major biotic factors, which leads to heavy yield losses and hence poses a big challenge for pigeonpea production in the Indian subcontinent. Variability in the sterility mosaic pathogen revealed the occurrence of five different isolates in India. Among them, three distinct SMD isolates have been characterized, viz., Patancheru, Bangalore and Coimbatore. Molecular tools offer a viable option to tackle these biotic stresses via identification of the genomic regions associated with the trait such as SMD resistance. With an aim of identifying the gene(s)/QTLs linked with SMD resistance, two F2 populations, i.e. ICP 8863 × ICPL 20097 (segregating for Patancheru SMD isolate) and TTB 7 × ICP 7035 (segregating for both Patancheru and Bangalore SMD isolates) were developed and F2:3 families were phenotyped for resistance to respective isolate(s) of SMD. After screening over 3000 SSR markers on parental genotypes of each mapping population, intra-specific genetic maps comprising of 11 linkage groups and 120 and 78 SSR loci were developed for ICP 8863 × ICPL 20097 and TTB 7 × ICP 7035 populations, respectively. Composite interval mapping (CIM) based QTL analysis by using genetic mapping and phenotyping data provided four QTLs for Patancheru SMD isolate and two QTLs for Bangalore SMD isolate. Identification of different QTLs for resistance to Patancheru and Bangalore SMD isolates is an indication of involvement of different genes conferring the resistance to these two SMD isolates. One QTL namely qSMD4 identified within an interval of 2.8 cM on LG 7 explaining 24.72% of phenotypic variance, once it is validated in other genetic background, seems to be a promising QTL for use in marker assisted selection. In summary, this is the first study on development of intra-specific genetic maps and identification of QTLs for SMD resistance in pigeonpea.  相似文献

  1. The Eurasian otter (Lutra lutra), smooth‐coated otter (Lutrogale perspicillata), and Asian small‐clawed otter (Aonyx cinereus) have all been reported previously from the Indian state of Uttarakhand. However, little information is available about their current distribution in a mountainous region that is subject to increasing human‐induced stressors (such as hydropower plants, pollution, sand and boulder mining, destructive fishing techniques, poaching).
  2. Owing to important roles played by these otters in structuring riverine food webs (particularly taking account of their roles as top carnivores), it is critical that they receive suitable protection in the face of projected temperature rise, change in precipitation patterns, and associated river flows in this Himalayan biodiversity hotspot. This study assesses otter distribution in four rivers of Uttarakhand as a basis for informing future conservation actions.
  3. Field surveys were conducted (October 2018–January 2019) in reaches of the Kosi, Ramganga, Khoh, and Song rivers, supported by semi‐structured interviews (N = 379) conducted with members of local communities to collect qualitative data on views and perceptions of otter species. In addition, community‐based otter awareness camps were organized for local youths (N = 105), adults (N = 115), and schoolchildren (N = 256 covering 10 schools).
  4. Habitat suitability maps were created using remote‐sensing data, survey findings, and a geographic information system to provide information about priority reaches of river to be targeted for future conservation efforts.
  5. This study provides critical interdisciplinary baseline information to guide decision‐makers towards developing a targeted, otter‐specific conservation programme for this important Himalayan biodiversity hotspot.
  6. The otter conservation education programmes conducted during this study resulted in a proposal to set up a community‐based conservation initiative to monitor and report otter sightings from the area, potentially representing a way forward for achieving simultaneous otter conservation and associated ecosystem benefits for local communities.
