Calcium uptake by bell pepper (Capsicum annuum L. cv. ‘California Wonder') varied by stage of plant development and N form supplied (NO3 NH4 + ratios: 1:0, 3:1, 1:1, 1:3, and 0:1) in a hydroponic study. Uptake of Ca++ was highest at bloom and during fruit expansion, making the fruit development stage the highest demand period. Calcium uptake declined with each increasing increment of NH4 + relative to NO3 supplied, although fruit yield was not significantly reduced until the ratio of NH4 + to NO3 exceeded 50%. Tissue Ca++ levels in the blossom‐end of the fruit were reduced whenever NH4 + was included with N supplied. Vegetative yield of plants followed the same trend as that observed for total fruit dry weights. Our results indicate that pepper yields are higher when NO3 is the predominant form of N. Also, these results strongly suggest that Ca++ fertilizer applications should precede the bloom period and continue during fruit development to ensure adequate Ca++ availability for fruit development.  相似文献

A methodology for ranking and classifying performance of irrigation systems through multidimensional performance indicators is developed using fuzzy set theory. The procedure uses the concepts of fuzzy resemblance and fuzzy dominance. Preference levels reflecting management priorities are incorporated into the analysis using appropriate weighting factors. The application of the procedure is demonstrated through two case studies: the Shi-Jin irrigation district in Hebei Province, China and the Goulburn irrigation region in Victoria, Australia. The classification of performance periods for the Australian system shows three clusters indicating the predominant effect of the increase in waterlogged area when higher priority is given to this indicator. No clear trend appeared when equal weight was assigned to all indicators. The effect of economic reforms implemented in 1977–78 in China is clearly reflected in the resulting ranking and clustering of the performance periods in the Shi-Jin irrigation district. Transport of post‐larvae shrimp used in aquaculture is an important element of successful cultivation because of the potential for stress during stocking procedures. To find optimum transport conditions, several bioassays were performed in the laboratory to evaluate survival of whiteleg shrimp Litopenaeus vannamei 5–30‐day‐old postlarvae under conditions similar to those encountered during transport from the hatchery to nursery and shrimp ponds. Postlarvae were exposed for 4 h to different temperatures and pH levels ammonia concentrations. Survival was significantly reduced after a 4 h exposure to pH 9 and was inversely related to temperature with or without 7 mg L?1 of ammonia. The 15‐ and 20‐day‐old postlarvae had higher survival rates than other ages. The lowest survival occurred in alkali conditions (pH 9), with 7 mg L?1ammonia at 30 and 32°C. To assure optimal survival of postlarvae during transfer from the hatchery to the nursery and shrimp ponds, we recommend temperatures below 28°C, pH no higher than 8, no ammonia and post‐larval age at least 15 days.  相似文献

The confirmatory diagnosis of Mycobacterium bovis (M. bovis) in animal samples is carried out by culture in Stonebrink media. However, culture is very slow because of the extremely long duplication time of the bacillus and difficult because of the scarcity of bacilli in diagnostic samples. This study describes the development of a single-tube touch-down polymerase chain reaction (PCR) protocol for the detection of M. bovis using primers that target the IS6110 element. Spiked water and milk as well as routine diagnostic samples (milk and nasal swabs) from M. bovis-positive cattle were tested. This protocol allows the rapid and sensitive detection of M. bovis in bovine samples by enhancing the sensitivity of standard PCR amplification.  相似文献

OBJECTIVE: To evaluate chemotactic, phagocytic, and bactericidal activities of bovine and porcine alveolar macrophages (AM) exposed to tilmicosin. ANIMALS: 12 healthy calves and 12 healthy pigs. PROCEDURES: Lungs were obtained immediately after euthanasia; AM were collected by means of bronchoalveolar lavage and density gradient centrifugation. Chemotactic activity was evaluated by exposing AM to lipopolysaccharide or macrophage inhibitory peptide during incubation with tilmicosin. Phagocytic activity was evaluated by incubating AM with tilmicosin for 24 hours and then with tilmicosin-resistant Salmonella serotype Typhimurium. Bactericidal activity was evaluated by incubating AM with tilmicosin (0, 10, or 20 microg/ml for bovine AM; 0 or 10 microg/ml or 10 microg/ml but washed free of tilmicosin for porcine AM) and then with Mannheimia haemolytica (bovine AM) or with Actinobacillus pleuropneumoniae or Pasteurella multocida (porcine AM). RESULTS: Tilmicosin had no significant effects on chemotactic or phagocytic activities of bovine or porcine AM. The time-course of bactericidal activity was best described by polynomial equations. Time to cessation of bacterial growth and area under the time versus bacterial number curve were significantly affected by incubation of AM with tilmicosin. CONCLUSIONS AND CLINICAL RELEVANCE: Results show that bactericidal activity of bovine and porcine AM was enhanced by tilmicosin, but not in proportion to the reported ability of AM to concentrate tilmicosin intracellularly. With or without exposure to tilmicosin, the time-course of bactericidal activity of bovine AM against M haemolytica and of porcine AM against A pleuropneumoniae or P multocida was too complex to be reduced to a simple linear equation.  相似文献

Immunity to Taenia solium infection was investigated using an experimental intramuscular oncosphere infection assay (IMOA) model in pigs. Three naturally infected pigs with cysticercosis were treated with oxfendazole (OFZ), a drug demonstrated to kill cysts in porcine muscle. These animals were then challenged with oncospheres but did not develop any cysts while three uninfected pigs that were similarly challenged, did develop intramuscular cysts. In another study, two groups of three pigs each were immunized with crude T. solium oncosphere and metacestode antigens, respectively, and tested with the IMOA. Immunization with crude oncosphere antigens (OAs) induced 100% protection, while metacestode antigens provided only partial protection. Immunoblots showed that pigs with complete immune protection to oncosphere intramuscular challenge had antibodies to two OAs at 31.3 and 22.5 kDa, respectively. Antibody to these two antigens was absent in pigs immunized with metacestodes or in uninfected control pigs. This study demonstrated the presence of two antigens that are unique to the oncosphere. Although, antibody to these two antigens is consistently present in pigs that are protected from an oncosphere intramuscular challenge their role in preventing infection by T. solium larval cysts is still hypothetical.  相似文献

A model to predict the bending modulus of elasticity (MOE) of oriented strand board (OSB) panels produced by batch processing is presented. The approach developed herein is unique in its comprehensiveness since the MOE is determined from information on the panel structure, temperature and moisture profiles and vertical density profiles obtained from the mat formation and compression models presented in Part 1. Comparison of predicted MOE values with those measured from 24 commercially produced panels shows good agreement considering some of the uncertainties involved. Simulations show that the MOE can be increased by any of the following changes: reduced fines content, increased panel density, better flake alignment in each of the three layers within a panel, increased flake length and a larger difference between the density of the face and core layers. The model was also used in a genetic algorithm to carry out an optimization study of batch OSB manufacturing. This analysis showed that by combining the appropriate reduction in the amount of flakes used, increase in fines content, improvement in flake alignment within each of the face and core layers and shortening of the batch time, a significant theoretical profit increase from the base case scenario can be obtained.  相似文献

The dietary protein requirement of the freshwater prawn Macrobrachium rosenbergii to obtain the maximum body protein retention was assessed by the estimated protein needed daily for maintenance (M) and body protein increment (G). The body protein increment was estimated by quantifying body nitrogen increments when the prawn was fed high‐protein diets composed primarily of casein. The protein required for maintenance was estimated by regressing weight gains in feeding experiments using graded dietary levels of casein. The true daily retention (R) or increase in body protein of the prawn corresponds to the sum of M and G. The values of M, G, and R determined in the present study were 3.86, 0.24, and 4.10 g/kg body weight/d, respectively. On the basis of the daily body protein increment and the net protein utilization of casein (58%), dietary protein requirement of the juvenile M. rosenbergii for maximum body protein retention was suggested to be about 7.1 g/kg body weight/d.  相似文献

Nutrient content and pH of rain samples collected at nine sites during 1988-1996 were studied to determine the amounts of N, P and S added by precipitation over Maracaibo Lake basin. The basin is a wide depression occupied by Lake Maracaibo and extensive alluvial plains and delimited by a mountainous zone of variable physiolgraphic characteristics with different cattle ranching and industrial activities. Rain pHs averaged ≈ 4 to 6, individual samples occasionally reached as low as pH 3 in the south basin. NH4 + levels were significantly higher (up to 2.6 mg/l NH4-N) than NO3 ? plus NO2 ?. Phosphorus levels were low, ranging from undetectable to 1.5 mg/l. The amounts of inorganic N/ha/yr added ranged from ≈ 5.4 Kg in the north to ≈ 8 Kg in the south basin; around the lakeshore, it was estimated that ≈ 9.8–16.8 Kg TN/ha (≈ 55 % as organic-N) are annually deposited. The annual amounts of SO4-S/ha added range from ≈ 10 to 13 Kg. The contribution of precipitation to P in the ecosystem is very small; ≈ 0.02 – 0.8 Kg of PO4-P/ha/yr was added in basin. The results of this study show that precipitation represents a significant factor for the total nitrogen and sulfur loading to Maracaibo Lake Basin.  相似文献
