This study evaluated the effects of fat and sugar levels on the surface properties of Lactobacillus rhamnosus GG during storage in food model systems, simulating yogurt and ice cream, and related them with the ability of the bacterial cells to adhere to Caco-2 cells. Freeze-dried L. rhamnosus GG cells were added to the model food systems and stored for 7 days. The bacterial cells were analyzed for cell viability, hydrophobicity, ζ potential, and their ability to adhere to Caco-2 cells. The results indicated that the food type and its composition affected the surface and adhesion properties of the bacterial cells during storage, with yogurt being a better delivery vehicle than ice cream in terms of bacterial adhesion to Caco-2 cells. The most important factor influencing bacterial adhesion was the storage time rather than the levels of fats and sugars, indicating that conformational changes were taking place on the surface of the bacterial cells during storage.  相似文献

Interferon gamma (IFN-γ) or type II interferon is a cytokine that is critical for innate and adaptive immunity against viral and some bacterial and protozoal infections. The importance of IFN-γ in the immune system lies in its ability to inhibit viral replication directly and most importantly from its immunomodulatory effects. Previously, we successfully co-administered IFN-γ along with GAPDH gene of Edwardsiella tarda as bicistronic DNA vaccine in Labeo rohita. In order to ascertain the individual role of IFN-γ, the present study involves cloning and expression of 552-bp IFN-γ open-reading frame (ORF) of L. rohita in striped snakehead (SSN-1) cell line using eukaryotic expression vector system (pQE-TriSystem) followed by transfection in peripheral blood lymphocytes (PBMCs) to evaluate its immunomodulatory ability in comparison to polyinosinic-polycytidylic acid (Poly I:C)-treated PBMCs. The 18.7-kDa protein, expressed in the pQE-IFNγ-transfected SSN-1 cells, reacted with anti-His antibody in Western blot confirming it to be recombinant IFN-γ, whereas the relative expression of IFN-γ, iNOS, Mx, and IL-1β genes in PBMCs was quantified at 24 h and 48 h post treatment by qPCR. The comparative kinetics of all four genes showed significantly (p?<?0.05) high upregulation pattern in both pQE-IFNγ-transfected cell group and Poly I:C-treated cell group demonstrating recombinant IFN-γ as an equally efficient inducer like Poly I:C. Thus, our in vitro experiment results highlight the immunomodulatory potential of recombinant IFN-γ as an analogue to synthetic Poly I:C which warranted future studies to further explore the potential of recombinant IFN-γ as an effective vaccine adjuvant against different microbial invasion.  相似文献

The study reports diversity of arbuscular mycorrhizal fungal (AMF) species in the rhizosphere of an endangered anticancerous herb – Curculigo orchioides Gaertn. in its natural habitat. A total of 18 species of AMF, belonging to three genera (Acaulospora, Glomus and Gigaspora) were recorded, with Glomus microcarpum being the most abundant species type. The AMF species composition across the study sites appeared to be influenced by soil pH rather than soil P and vegetation. Acaulospora laevis spores were restricted to sites where the soil pH was acidic. The effectiveness of these native AMF species on growth performance of C. orchioides plants was compared under experimental conditions. In general, the mycorrhizal plants were superior in most of the evaluated parameters, but the extent to which the growth of mycorrhizal plants was influenced varied with the inocula used. The plants inoculated with mixed consortia containing maximum AMF species richness exhibited improved growth in comparison to consortia containing lower AMF diversity and monospecies cultures. The variable plant responses observed with any two consortia having same species richness in the present study could be due to variable component AMF species and their relative abundance. These results emphasize the need to protect the below-ground diversity of AMF and recommend their usage for restoration practices.  相似文献

A wild non-progenitor species from wheat (Triticum aestivum L.) tertiary gene pool, Aegilops peregrina (Hack.) Maire & Weiller accession pau3519 (UUSS), was used for introgression of leaf rust (caused by Puccinia triticina) and stripe rust (caused by Puccinia striiformis f. sp. tritici) resistance in bread wheat. The accession was crossed and backcrossed with hexaploid wheat line Chinese Spring PhI to develop two homozygous BC2F6 wheat-Ae. peregrina introgression lines (ILs), viz., IL pau16058 and IL pau16061, through induced homoeologous recombination. Homozygous lines were screened against six Puccinia triticina and two Puccinia striiformis f. sp. tritici pathotypes at the seedling stage and a mixture of prevalent pathotypes of both species at the adult plant stage. IL pau16061 showed resistance to leaf rust only, whereas IL pau16058 was resistant to both leaf and stripe rust pathotypes throughout plant life. Molecular profiling of these ILs with simple sequence repeat (SSR) markers indicated that alien introgressions were mainly terminal and very few were interstitial. Identification of linked markers with advanced genomic technologies will aid in marker-assisted pyramiding of alien genes in cultivated wheat background.  相似文献

Over the past few years, micropropagation techniques are being widely used as an important and advantageous tool for rapid propagation of several commercially important varieties of horticultural and forest tree species. Although, this technology has got several successful applications, low survival rates and poor growth while shifting these plantlets to field conditions are the most common problems which hinders its usage for commercial plant production practices. Inoculation of arbuscular mycorrhizal fungi (AMF) to the roots of micropropagated plantlets plays a beneficial role on their post-transplanting performance. This review discusses the multiple roles played by AMF in the establishment of micropropagated plantlets. Previous studies have shown that carbon (C) mineralization in saline or sodic soils is affected by various factors including organic C content, salt concentration and water content in saline soils and soil structure in sodic soils, but there is little information about which soil properties control carbon dioxide (CO2) emission from saline-sodic soils. In this study, eight field-collected saline–sodic soils, varying in electrical conductivity (ECe, a measure of salinity, ranging from 3 to 262 dS m−1) and sodium adsorption ratio (SARe, a measure of sodicity, ranging from 11 to 62), were left unamended or amended with mature wheat or vetch residues (2% w/w). Carbon dioxide release was measured over 42 days at constant temperature and soil water content. Cumulative respiration expressed per gram SOC increased in the following order: unamended soil<soil amended with wheat residues (C/N ratio 122)<soil with vetch residue (C/N ratio 18). Cumulative respiration was significantly (p < 0.05) negatively correlated with ECe but not with SARe. Our results show that the response to ECe and SARe of the microbial community activated by addition of organic C does not differ from that of the less active microbial community in unamended soils and that salinity is the main influential factor for C mineralization in saline–sodic soils.  相似文献

Stripe rust is considered as the current major rust disease affecting winter cereal production across the world. A quick, reliable PCR-based marker was developed here to detect, identify and rapidly monitor Puccinia striiformis f. sp. tritici (Pst) in wheat-growing areas. Three respective sets of primers, designed from β-tubulin, squalene monooxygenase and ketopantoate reductase genes selected from the full genome of Puccinia striiformis f. sp. tritici, amplified sequences of 239, 358 and 1518 bp, respectively, in Pst pathotypes. A fragment of 1518 bp unique to Pst pathotypes was amplified using primer set PstKeto F1_30/Pst KetoR1_1547 and distinguished the pathogen clearly from different Puccinia spp. and other fungal pathogens. The detection limit of the marker (KetoPstRA1500, accession no. KU240073) by conventional PCR assay was 10 pg. This marker could detect the pathogen in the host before symptom expression. The sensitivity and utility of the marker were further enhanced in a qPCR-based assay that was developed with a newly designed primer set PstKeto F1_1246/Pst KetoR1_1547, which amplified a product of 302 bp and detected as little as 10 fg of DNA. This PCR/qPCR based marker is suitable for studying cultivar resistance, which requires accurate quantification of the pathogen in diseased host tissue.  相似文献

Cowpea mild mottle virus (CPMMV) is an emerging severe disease of soybean. The resistant genotypes, DS 12‐5 and SL958, were crossed with susceptible genotypes F4C7‐32 and JS335, respectively. Resistance reactions of sap‐inoculated F2 plants and individual F2 plant‐derived F3 families indicated that resistance was controlled by a single dominant gene. Molecular mapping with bulked segregant analysis showed that Satt635 and UO8405 are linked to resistance gene which is located on linkage group H.  相似文献
