Semi-hardwood cuttings of Cotoneaster divaricata grown in 926 cm3-containers were top-dressed with controlled-release Osmocote (18.0 N-2.6 P-10.0 K) at levels of 0, 2, 3 or 6 g/l of container. Optimum shoot and root growth was obtained at 2 g/l. Applications of controlled-release fertilizer significantly increased foliar N and P concentration, but not foliar K. The most desirable shoot-to-root ratio occurred at 2 g/l.  相似文献

Aspiration of lytic bone lesions is an excellent diagnostic test in the initial evaluation of primary bone neoplasia. However, cytologically, it can be difficult to differentiate osteosarcoma (OSA) from other bone neoplasms, including fibrosarcoma, chondrosarcoma, synovial cell sarcoma, and plasma cell myeloma. The purpose of this study is to determine the sensitivity and specificity of alkaline phosphatase (ALP) staining to differentiate OSA from other tumors that express vimentin by immunocytochemistry or immunohistochemistry. ALP is a hydrolytic enzyme present in multiple tissues including liver, kidney, intestine, placenta, and bone. Hypothetically, neoplasms actively producing bone should be specifically positive for ALP staining. Unstained, cytologic specimens were incubated for 8-10 minutes with nitroblue tetrazolium chloride/5-bromo-4-chloro-3-indolyl phosphate toluidine salt-phosphatase substrate. A positive reaction stains the membrane of the cells gray to black. Samples were counterstained with a Romanowsky's stain to determine whether the sample was of representative cellularity. A total of 61 vimentin-positive neoplasms have been evaluated and confirmed histopathologically. Tumors that expressed vimentin and were positive for ALP included 33 OSAs, one multi-lobular tumor of bone, one amelanotic melanoma, and one chondrosarcoma. Tumors that expressed vimentin and were negative for ALP included chondrosarcomas (three of four), multiple fibrosarcomas, and multiple synovial cell sarcomas. The sensitivity is 100%, and the specificity is 89%. In conclusion, ALP appears to be a highly sensitive and fairly specific marker in the diagnosis of OSA.  相似文献

BACKGROUND: Increased serum lipase activity has been used historically to support the diagnosis of acute pancreatitis, a common disease in dogs. Most of the lipase assays that are currently in use lack optimum sensitivity and specificity. OBJECTIVE: The objectives of this study were to 1) validate the 1,2-o-dilauryl-rac-glycero-3-glutaric acid-(6'-methylresorufin) ester (DGGR) assay for determination of lipase activity in canine serum and 2) compare results, reference intervals, sensitivity, and specificity of the DGGR assay with a standard 1,2-diglyceride (1,2 DiG) assay for diagnosing acute pancreatitis in dogs. METHODS: Precision, linearity, and interference studies were performed for method validation on a Hitachi 911 analyzer. Lipase results from the DGGR and 1,2 DiG assays were compared by linear regression analysis. Sensitivity, specificity, and diagnostic efficacy were determined for both assays on a population of 30 dogs, 15 of which had acute pancreatitis based on history, clinical signs, and ultrasound findings. RESULTS: Within-run and within-day coefficients of variation (CVs) were low (<3%), with higher day-to-day CVs (< or =14 %). The assay was linear between 8 and 2792 U/L. No significant interference by hemolysis and lipemia was found. Poor correlation was found between the assays (r(s)=0.84). The lipase reference interval was 8-120 U/L for the DGGR assay and 30-699 U/L for the 1,2 DiG assay. Sensitivity and specificity for the diagnosis of pancreatitis were 93% and 53%, respectively, for the DGGR assay and 60% and 73% for the 1,2 DiG assay. Receiver operating characteristic curve analysis showed similar areas under the curve. CONCLUSIONS: On the basis of this study, the DGGR method is considered adequate for assaying serum lipase activity in dogs. The high sensitivity of the DGGR assay suggests it may be a useful screening test for canine pancreatitis.  相似文献

In this study, the results of investigations of organic matter decomposition in natural and dredged areas of the inner Puck Bay (Baltic Sea) are presented. The dredging of relatively deep pits causes environmental problems. Researched post-dredging pits are sediment traps in which 3 times more organic carbon (Corg), 3.5 times more total nitrogen (Ntot), about 1.5 times more organic phosphorus (Porg) and 1.7 times more total phosphorus (Ptot) accumulate as compared to the non-dredged regions; they are also characterized by very intensive decomposition of organic matter. About 42, 44, 95 and 50% of the annual load of, respectively, Corg, Ntot, Porg and Ptot undergo decomposition in the dredged area, whereas the respective values for natural seabed are ca. 11, 44, 41 and 21%. Reduction of nitrogen in the pit occurs mainly through ammonification, while in the natural areas of seabed denitrification prevails. In non-dredged sediments, 84% of the released nitrogen comes from denitrification whereas in the pit, it is only 18%. Organic matter degradation in the pit sediments manifests itself by a 7-fold increase in the phosphate flux into near-bottom water as compared to natural seabed. The observed phosphate flux originated from the organic matter as well as from the decomposition of inorganic phosphorus compounds. Periodically, sulphate reduction in the pit sediments resulted in hydrogen sulfide occurrence.  相似文献

Root and shoot growth rates of Cotoneaster divaricata were determined using a growing system of polyvinyl chloride pipe cylinders in which longitudinal sections could be removed for periodic root observations. Plants were fertilized with nitrogen (N), phosphorus (P), and potassium (K). N, as NH4NO3, was applied weekly at levels of 0, 250 or 500 mg/l and P, as H3PO4, was incorporated in the medium at levels of 0, 5 or 50 mg/l. K, as KCl, was maintained in the medium at a 150 mg/l level by soil tests conducted weekly. Shoot growth was increased after N application. However, no difference was observed between N levels. P increased shoot growth only at the highest N level applied. Although root growth was not increased by either N or P, high N levels inhibited root growth, whereas P stimulated root growth. No correlation (r = 0.19) was observed between shoot and root growth.  相似文献

Rice (Oryza sativa L.) plants were grown over a 30‐day‐period in nutrient solutions containing increasing copper (Cu) concentrations (0.002, 0.01, 0.05, 0.25, 1.25, and 6.25 mg/L). It was observed that in both root and leaf tissues the total activity of l‐aminocyclopropane‐l‐carboxylate synthase decreased at concentrations above 0.05 mg/L Cu treatment, whereas the total activity of the ethylene forming enzyme slightly increased until the 1.25 mg/L Cu treatment. In the root and leaf tissues, the 1‐aminocyclopropane‐l‐carboxylic acid concentrations decreased after the 0.05 mg/L and 0.01 mg/L Cu treatments, respectively, whereas the ethylene production decreased in both tissues after the 0.05 mg/L Cu treatment. It is proposed that excess Cu in both root and leaf tissues decrease the conversion of S‐adenosylmethionine to 1‐aminocyclopropane‐l‐carboxylic acid through the inhibition of the total l‐aminocyclopropane‐l‐carboxylate synthase activity. The concomitant effect of this inhibition on adventitious root formation and leaf senescence is evaluated.  相似文献

Seventy-eight (33 Bulgarian and 45 foreign) common bean (Phaseolus vulgaris L.) genotypes included in Bulgarian collection were screened for ISSR (Inter Simple Sequence Repeats) and AFLP™ (Amplified Fragment Length Polymorphism) markers. ISSR analysis was performed with 13 primers, and 55 (36.7 %) out of the 150 bands observed were polymorphic. One hundred and sixty-four AFLP™ fragments were obtained with three primer combinations, of which 54 (32.9%) were polymorphic. UPGMA (Unweighted Pair Group Method Arithmetic Averages) analysis was performed using DICE coefficient and dendrograms were constructed using either the data from each method (ISSR and AFLP™) separately or combined in a single joint matrix. Despite some genetic heterogeneity observed in both pools (Middle American origin: M and Andean origin: A) the genotypes were separated in to main groups: one gathering genotypes mainly from pool M and the other more genotypes from pool A. Moreover, Bulgarian genotypes were spread over the two groups suggesting that they are not subject to genetic erosion and that the Phaseolus genetic diversity is conserved.  相似文献

Purpose

Mercury speciation in sediments is linked to environmental conditions and processes. Domination of particular mercury species depends on its source, displays considerable seasonal behavior, and may be further modified due to oxygen levels, icing conditions, or the input of fresh organic matter. The purpose of this study was to examine the coastal area of the Gulf of Gdańsk in terms of mercury contamination and the influence of seasonal environmental changes on its speciation.

Materials and methods

In three highly dynamic coastal sites, mercury speciation in sediments was studied in relation to other environmental parameters (redox conditions, organic matter concentration, bacteria abundance, etc.). Sediment and water samples were collected monthly during a 3-year study. Sequential extraction of sediments was used for identification of four inorganic mercury species: dissolved, bound with fulvic and humic acids, mercury sulfide, and residual fraction. Cold vapor atomic fluorescence spectrometry (CV-AFS) was used for extracts and liquid sample analysis. Total mercury in sediments was measured with atomic absorption spectrometry (AAS). Changes in salinity were analyzed by measurements of chloride and sulfate ion concentrations using ion chromatography. Bacteria number and biomass were measured by direct counting using epifluorescence microscopy.

Results and discussion

Seasonal changes in mercury speciation were observed at all sites and attributed to different processes. Labile mercury fraction contribution varied from 0 to 80%. The speciation patterns varied locally as the stations selected for the study are diversified in terms of anthropogenic impact, water dynamics, and output from land. Mercury concentrations at all stations fluctuated during phytoplankton blooms, icing of marine waters, precipitation, or increased surface runoff from the land. In this local scale, the global climate changes are visible already as environmental conditions in studied area changed in comparison to elder research.

Conclusions

Obtained results suggest that although mercury emissions to the environment have decreased in recent years, local weather conditions, which may be intensified by climate change, seriously affect the bioavailability of past mercury deposits in coastal sediments.
