L-α-glycerophosphate oxidase (GlpO) plays a central role in virulence of Mycoplasma mycoides subsp. mycoides SC, a severe bacterial pathogen causing contagious bovine pleuropneumonia (CBPP). It is involved in production and translocation of toxic H2O2 into the host cell, causing inflammation and cell death. The binding site on GlpO for the cofactor flavin adenine dinucleotide (FAD) has been identified as Gly 12−Gly13Gly 14−Ile15−Ile16Gly 17. Recombinant GlpO lacking these six amino acids (GlpOΔFAD) was unable to bind FAD and was also devoid of glycerophosphate oxidase activity, in contrast to non-modified recombinant GlpO that binds FAD and is enzymatically active. Polyclonal monospecific antibodies directed against GlpOΔFAD, similarly to anti-GlpO antibodies, neutralised H2O2 production of M. mycoides subsp. mycoides SC grown in the presence of glycerol, as well as cytotoxicity towards embryonic calf nasal epithelial (ECaNEp) cells. The FAD-binding site of GlpO is therefore suggested as a valuable target site for the future construction of deletion mutants to yield attenuated live vaccines of M. mycoides subsp. mycoides SC necessary to efficiently combat CBPP.  相似文献

Wastewater treatment plants (WWTP) with insufficient technologies for wastewater purification often cause a distinct nutrient pollution in the receiving streams. The increased concentrations of dissolved nutrients can severely disturb the ecological integrity of streams, which has been recently shown for basic ecosystem processes like mineralization of coarse particulate organic matter (CPOM). The present study investigated the impact of a modern WWTP (Zentralkläranlage Jena) on breakdown rates of CPOM exposed in net bags (1 mm mesh size) to the effluent of a large municipal WWTP and an upstream control site in the Saale River (Thuringia, Germany) from April to October 2005. Control and effluent site differed significantly in water chemistry with increased concentrations of dissolved organic carbon (DOC), ammonium, sulfate, and chloride at the effluent site, while the control site displayed higher concentrations of nitrate. However, breakdown rates of toothpickers and small twigs were not significantly different between the sites, whereas breakdown rate of leaf litter was significantly higher at the effluent site (k?=?0.0124 day?1) than at the control site (k?=?0.0095 day?1). Benthic invertebrate assemblages inhabiting the sandy stream bed at both sites were dominated by Chironomidae and Oligochaeta, typical inhabitants of fine sediments. Although the Shannon diversity of the benthic invertebrates was slightly higher at the effluent site (0.85) than at the control site (0.63), no significant difference could be detected. Bacterial numbers in water samples and surface biofilms on glass slides also displayed no significant differences between the two sites. This study showed that the effluents of a WWTP with modern technologies for wastewater purification did not directly affect breakdown rates of CPOM, bacteria numbers in epibenthic biofilms and the water column, and the community composition of sediment inhabiting aquatic macroinvertebrates in an effluent-receiving river with already increased concentrations of dissolved nutrients.  相似文献

Ozone measured in cabin air on 14 flights over polar areas shows maximum concentrations during Spring. Ozone numbers close to atmospheric values were found in the air entering the cabin through the air conditioning system but a reduction to about half of this value takes place immediately within the cabin air. Aircraft with recirculating cabin air showed an additional reduction by about 25%. Maximum concentrations are considerably higher and the duration of such periods longer than those reported in FAA Technical Report ADS-5 in which, however, data from polar areas during O3 maximum are missing. Ozone destruction effects within the pressurization system as claimed in the FAA report are doubtful. Although O3 reduction occurs within the cabin air, the threshold value of 0.1 ppm. for breathing air was surpassed in about 75 % of the flight time (8 to 10 hr between Copenhagen and Seattle) and maximum concentrations (0.4 ppm during 4 hr and 0.6 ppm during 1 hr) were found to be close to critical values recommended by the Environmental Protection Agency. Assuming that no O3 destruction takes place in the pressurization system, O3 concentrations exceeding 1 ppm, can be expected to enter the cabin at flight levels for commercial flight on polar routes during Spring. Some experiences during periods of high O3 concentrations were increasing irritation in the eyes and the respiratory tract, effects different from those associated with low humidity in cabin air.  相似文献

A comparison was made between two soil climosequences on north- and south-facing slopes in northern Italy to determine the influence of slope aspect on soil processes. The climosequences span an elevational gradient ranging from moderate (1200 m a.s.l.) to high alpine (2420 m a.s.l.) climate zones on surfaces having an age of about 15 000 years. The soils were investigated with respect to organic C, oxalate and dithionite extractable Fe, Al and Si, elemental losses (Ca, Mg, K, Na, Fe, Al, Si, Mn) and clay minerals. The stocks of soil org. C as well as of oxalate-extractable Fe and Al was greatest in the subalpine zone near the timberline. There are no clear differences in organic C content between the soils on north- and south-facing sites. Fe-oxalate and to a lesser extent Alo-stocks were, however, greater on north-facing sites, indicating that weathering is greater there. Eluviation and illuviation of Al and Fe within the soil profile, typical for podzolisation, was more distinctly expressed on the N slopes. The probability of ITM (Imogolite-type-material) formation in the soil seemed to be greater on south-facing sites. On the north-facing sites, element leaching was most intense in the subalpine zone close to the timberline while on the south-facing sites this was only the case for the base cations. The N slopes exhibited higher leaching of elements which generally indicates a higher weathering intensity. On south-facing sites, typical podzolisation processes were measurable only above 2000 m a.s.l. The development of smectites is also a reflection of the weathering intensity; smectite was discernible in the surface horizon at all sites on N slopes but the highest amount was detected in the sub-alpine climate zone. For the south-facing sites only in the alpine climate zone could smectite be detected. Higher temperatures and an increased number of freeze-thaw cycles on south-facing slopes should theoretically enhance rates of chemical weathering. This could, however, not be confirmed with our measurements. The degree of chemical weathering increases from the south- to the north-facing sites that are characterised by lower temperatures, lower evapotranspiration and consequently by a higher humidity. Although precipitation in Alpine regions is abundant, the availability and flux of water through the soil is the prime factor in weathering intensity.  相似文献
