Pharmacokinetic and clinical effectiveness of liposome-encapsulated N-methylglucamine antimoniate (LMA) was performed in dogs suffering from experimental leishmaniosis. LMA was compared with N-methylglucamine antimoniate (MGA), the same drug in its free form. Sb plasma concentrations for LMA were always higher than those for MGA. Mean residence time (MRT), half-life time (t(1/2)) and clearance (Cl) showed that Sb was eliminated slower after liposome administration. The high volume of distribution (Vd) obtained with LMA suggests that Sb could achieve therapeutic concentrations in parasite-infected tissues. Average plasma concentration at steady state (Css(ave)) shows that Sb body concentrations after LMA treatment (9.8 mg/kg Sb, each 24h) would be effective in Leishmania infantum canine infection. Comparing LMA with MGA in a 1-year follow-up we observed no relapses for LMA and total protein and gammaglobulin concentrations were within normal range, while for MGA both began to rise 3 months after treatment. Use of antimonial liposomal formulations may restore effectiveness to an existing drug and reduce toxicity.  相似文献

Photoinhibition was examined in four co-occurring Mediterranean evergreen tree species during two consecutive winters. In response to low temperatures and saturating light, Juniperus phoenicea L., Pinus halepensis Mill., Quercus coccifera L. and Q. ilex ssp. ballota (Desf.) Samp. exhibited marked chronic photoinhibition, indicated by low predawn maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm). Low Fv/Fm values were correlated with high concentrations of xanthophyll cycle components (VAZ) and with the maintenance of high concentrations of zeaxanthin overnight (DPSpd). In all species, however, chronic photoinhibition was enhanced as the winter progressed in the absence of changes in DPSpd, suggesting cumulative damage toward the end of winter. Photoinhibition differed among species: P. halepensis always displayed significantly higher Fv/Fm values; and Q. coccifera had the lowest Fv/Fm values, showing a high sensitivity to the combination of high light and low temperatures. Differences among species were not fully explained by differences in the xanthophyll pool or its de-epoxidation state. Chronic photoinhibition overlapped with a dynamic photoinhibition as shown by the low values of photochemical efficiency of the open reaction centers of PSII at midday. Winter photoprotective strategies differed among species and may involve photoprotective mechanisms in addition to those associated with xanthophylls. The observed species-specific differences matched results obtained for the same species in summer; however, comparison of the two seasons suggests that the higher VAZ concentration observed in winter has an additional structural photoprotective role.  相似文献

This paper discusses the results of a lichenometrical and geomorphological study of one of the few remaining active snow hollows in the central region of the Iberian Peninsula. The study area, located on a glacial shoulder, is called Hoya del Cuchillar de las Navajas. A protalus rampart occurs at the base of the hollow. Our studies, conducted between 1992 and 1998, were designed to determine the geomorphological characteristics of Hoya, the mobility of the deposits, and the characteristics of the snow cover. These data formed the basis for a study of the lichen colonization on the blocks and on the wall surrounding the snow hollow. All of the lichen species found were analyzed according to their abundance, distribution and the extent of their surface cover. Measurements of the diameter of the thalli of the species Rhizocarpon geographicum were also obtained. Thalli of this species were found to require a mean snow-free growing season of at least 95 days (13.5 weeks) per year. Maximum mean thallus diameters indicate that the protalus rampart was formed during the Little Ice Age and became inactive 130 years ago.  相似文献

OBJECTIVE: To determine pharmacokinetic variables and pharmacologic effects of the S(-) isomer of bupivacaine (S[-]-BPV) in dogs. ANIMALS: 6 adult male Beagles. PROCEDURE: Dogs received S(-)-BPV (1 mg/kg of body weight) i.v., and 15 days later, the same dogs received 1.8 mg/kg epidurally. Pharmacokinetic variables and pharmacologic effects were determined for each route of administration. RESULTS: After i.v. administration, plasma concentration versus time curves were adjusted, using biexponential equations that indicated a rapid distribution phase followed by a slower elimination phase, with a mean +/- SD half-life of 33.5 +/- 17.0 minutes. Mean plasma clearance was 21.0 +/- 10.7 ml/min/kg, and mean volume of distribution at steady state was 0.8 +/- 0.2 L/kg. After i.v. administration, mean peak plasma concentration was 2.6 +/- 0.7 micrograms/ml; after epidural administration, it was 0.9 +/- 0.5 microgram/ml at approximately 3 minutes. Half-life after epidural administration was 5 times longer than that observed after i.v. administration. Motor block began immediately after the end of epidural administration and lasted for 3 to 4 hours. Changes in systolic blood pressure and heart rate after epidural administration were slight but occurred at the same time that plasma concentration peaked. After i.v. administration, motor block or variations in physiologic variables studied were not observed. CONCLUSIONS AND CLINICAL RELEVANCE: In dogs, the pharmacologic behavior of S(-)-BPV was similar to that of the bupivacaine racemate, but motor block attributable to S(-)-BPV lasted longer than that attributable to the racemate, with lower plasma concentrations observed at equivalent sample collection times.  相似文献

Although decline of Aleppo pine was observed long ago and several climatic and biotic factors have been previously associated with this complex process, site factors involved in this decline remain poorly understood. The objective of the work described here was to identify site factors associated with canopy condition. Canopy condition was estimated both by a visual estimation of defoliation, and by an indirect estimation of leaf area index (LAI) and other stand‐ and light‐related parameters through the analysis of hemispherical photographs. A high percentage of damaged trees (81%) along with high levels of defoliation in plots (up to 53%) and trees (up to 85%) were recorded. Regression models showed that the site factors associated with defoliation were basal area, age, crown depth and elevation, while those associated with LAI were diameter at breast height, tree density and canopy openness. Analysis of hemispherical photographs proved to be a useful method for LAI estimation, but not for estimation of defoliation due to heterogeneous defoliation patterns caused by fungal pathogens detected in the study area. Soils and climatic conditions were common to all plots, so their influence could not be tested, but poor soil conditions and climatic restraints are known in this area, including low soil productivity, frequent summer droughts and high numbers of frost days. The results obtained suggest that several factors were associated with the decline of Pinus halepensis, including age, basal area, canopy openness, diameter, height and tree density. These factors can influence canopy condition, and thus, they might be acting as predisposing factors for the decline. The modulation of these factors is possible if suitable forest management strategies are applied, which could lead to a decrease of the decline incidence.  相似文献

Abstract

Egg extrusion patterns were investigated as an inducible defensive mechanism in potato plants against the leafminer Liriomyza huidobrensis (Blanchard) (Diptera: Agromyzidae). Increased multiplication rates in leaf cells surrounding an egg of L. huidobrensis leads to its exposure through the leaf cuticle, which might increase the risk of mortality. Extrusion rates were evaluated in laboratory tests, in relation to plant and leaf age in Solanum tuberosum L. var. Spunta. Stratified field sampling was also carried out in order to assess leafminer distribution on new and old leaves. Extrusion was higher in young plants, being restricted in older ones to new, still expanding leaves. Plant reaction was independent of egg density. In the field, damage was concentrated in older foliage, in line with the extrusion results. The relevance of egg extrusion for pest management is discussed.  相似文献

We compare dry mass (DM) and storage of starch (St) and nitrogen (N) in seedlings of three Mediterranean oaks, two evergreens (Quercus coccifera L. and Q. ilex L. subsp. ballota (Desf.) Samp) and one deciduous (Q. faginea Lam.), across different scenarios of nutrient and water availabilities. Three fertilization (5, 50 and 200 mg of N per plant and growing period) and watering (28–39, 55–71 and 70–85 g H2O 100 g soil ?1 gravimetric soil water) treatments were applied to current-year seedlings between May and October 2002 in two independent experiments. The three species showed a similar response to fertilization, storing nitrogen instead of increasing biomass, in agreement with adaptations to nutrient-poor habitats. However, they differed in their responses to water, reflecting the different water requirements in the field: Q. coccifera, from arid zones, showed no response to water regarding DM and St; Q. faginea, from humid zones, required higher water availability to simultaneously increase growth and storage; while Q. ilex, spanning over most of the water availability range, exhibited a balanced increase of both functions when water increased moderately. In the two evergreen species, N concentration increased with water supply, whereas the reverse occurred in Q. faginea. The latter species favoured growth over storage at moderate water supply (according to its more competitive strategy), although it was the species which accumulated more St and N at the end of the experiments (autumn).  相似文献

Leptin is an anorectic hormone secreted mainly by peripheral adipocytes but also by other central and peripheral tissues. It acts by means of a receptor called OB‐R, influencing not only appetite and body mass but being also involved in many fields like endocrinology, metabolism and reproduction. Immunohistochemistry and qRT‐PCR techniques were, respectively, used to demonstrate the presence of leptin and its receptor in the gut of adult zebrafish and to evaluate the leptin gene expression response to feeding and fasting. Immunoreactivity for the antibodies utilized was demonstrated in feeding but not in fasting fish, and the gene expression analysis corroborates the data obtained by immunohistochemistry. Therefore, all the obtained results support the hypothesis of the role of this hormone in food regulation in zebrafish.  相似文献

For Holm oak (Quercus ilex L.), a flush growing species, nutrient loading during the growing season is difficult and can lead to a low nutrient status of the seedlings. To provide insights about Holm oak nutrient dynamics during fall in the nursery and subsequent planting performance, a factorial nursery experiment was conducted in a mild fall–winter area testing the effects of timing of fertilization (early and late fall) and rate (two doses of a NPK fertilizer that applied 28 and 56 mg N per seedling), followed by an experimental plantation.  相似文献
