The body complex of the soybean seed (BCSS) was isolated from the single cells (27.2%) by a sequential procedure of autoclaving with water, cellulase digestion for the primary cell wall, pectinase digestion for the secondary cell wall, and defatting with hexane washing. Its characteristics were then investigated. The defatted BCSS (DBCSS) consisted of protein (76.5%) and mannose-rich carbohydrates (3.2%). Screening of the food-processing protease for the digestion of DBCSS was carried out, and a kind of alkaline protease was selected. The inner protein of DBCSS was easily extracted with 0.1 M sodium carbonate buffer, pH 10, and the insoluble shell of the body complex (SDBCSS) was left. SDBCSS consisted of hydrophobic amino acid-rich protein. SDBCSS was easily digested by the selected alkaline protease. SDBCSS was dissolved by boiling with sodium dodecyl sulfate-mercaptoethanol, and it was found to consist of a protein of approximately 3 kDa. The high enzymatic digestion including the selected protease for soybean seed and defatted soybean meal was carried out; both were extracted and digested with a yield of >99.5%. The final indigestible residue was found as paired hexagonal and filamentous organs of the soybean cells.  相似文献

The effects of inoculating arbuscular mycorrhizal (AM) fungi on the growth, phosphorus (P) uptake, and yield of Welsh onion (Allium fistulosum L.) were examined under the non-sterile field condition. Welsh onion was inoculated with the AM fungus, Glomus R-10, and grown in a glasshouse for 58?days. Non-inoculated plants were grown as control. Inoculated and non-inoculated seedlings were transplanted to a field with four available soil P levels (300, 600, 1,000, and 1,500?mg P2O5?kg?1 soil) and grown for 109?days. AM fungus colonization, shoot P concentration, shoot dry weight, shoot length, and leaf sheath diameter were measured. Percentage AM fungus colonization of inoculated plants was 94% at transplant and ranged from 60% to 77% at harvest. Meanwhile, non-inoculated plants were colonized by indigenous AM fungi. Shoot length and leaf sheath diameter of inoculated plants were larger than those of non-inoculated plants grown in soil containing 300 and 600?mg P2O5?kg?1 soil. Shoot P content of inoculated plants was higher than that of non-inoculated plants grown in soil containing 300 and 600?mg P2O5?kg?1 soil. Yield (shoot dry weight) was higher for non-inoculated plants grown in soil containing 1,000 and 1,500?mg P2O5?kg?1 soil than for those grown in soil containing 300 and 600?mg?P2O5 kg?1 soil. Meanwhile, the yields of inoculated plants (200?g plant?1) grown in soils containing the four P levels were not significantly different. Yield of inoculated plants grown in soil containing 300?mg P2O5 kg?1 soil was similar to that of non-inoculated plants grown in soil containing 1,000?mg P2O5?kg?1 soil. The cost of AM fungal inoculum for inoculated plants was US$ 2,285?ha?1 and lower than the cost of superphosphate (US$ 5,659?ha?1) added to soil containing 1,000?mg P2O5 kg?1 soil for non-inoculated plants. These results indicate that the inoculation of AM fungi can achieve marketable yield of A. fistulosum under the field condition with reduced application of P fertilizer.  相似文献

Expression of calbindin-D28K in sporadic nephroblastomas of the chicken   总被引:2,自引:0,他引:2  
Calbindin-D28K (Ca-D28K) is a calcium-binding protein. In the kidney, Ca-D28K is present in the distal nephron, but not in the proximal nephron. This site-specific distribution in the kidney indicates that Ca-D28K is a potential marker for the differentiation of the distal nephron. In this study, we have examined the expression of Ca-D28K in 25 sporadic cases of chicken nephroblastomas. All cases of nephroblastomas were composed of atypical tubular structures, blastemal cells, and fibrous stroma in varying degrees of differentiation. Immunohistochemically in all nephroblastoma specimens, Ca-D28K was expressed in the epithelial cells of the subsets of tubular structures, but not in the blastema or the stroma. These results suggested that the tubuli in the nephroblastomas are able to differentiate into the phenotype of distal nephrons. Furthermore, Ca-D28K might develop as a novel diagnostic marker for nephroblastomas because this molecule is reported to be completely negative in other renal tumors, including renal cell carcinoma, chromophobe carcinomas, and oncocytoma.  相似文献

We used ten discs from two Zelkova serrata trees - five discs from each tree at sampling heights of 1, 4, 7, 10, and 13 m above the ground - and investigated the radial variation in ray characteristics, i.e., ray area (cross-sectional area of rays on a tangential section), ray density (number of rays/mm2 on a tangential section), and ray proportion (the percentage of the area occupied by rays on a tangential section) and analyzed the pattern of variation with respect to the three stages (early, middle, and late) of radial stem increment as estimated using the Gompertz growth function. A juvenile-mature pattern of variation was observed in ray area and ray density. Ray area increased in the inner part of stem and fluctuated around a certain value in the outer part of the stem, and ray density decreased in the inner part of stem and tended to be constant in the outer part of the stem. The maturation age of ray density was similar to the age at the boundary between the early and the middle stages of radial stem increment, but ray area and ray proportion did not relate to the stages of radial stem increment.  相似文献
