Papillomaviruses (PVs) infect a wide range of animal species and show great genetic diversity. To date, excluding equine sarcoids, only three species of PVs were identified associated with lesions in horses: Equus caballus papillomavirus 1 (EcPV1-cutaneous), EcPV2 (genital) and EcPV3 (aural plaques). In this study, we identified a novel equine PV from aural plaques, which we designated EcPV4. Cutaneous samples from horses with lesions that were microscopically diagnosed as aural plaques were subjected to DNA extraction, amplification and sequencing. Rolling circle amplification and inverse PCR with specific primers confirmed the presence of an approximately 8 kb circular genome. The full-length EcPV4 L1 major capsid protein sequence has 1488 nucleotides (495 amino acids). EcPV4 had a sequence identity of only 53.3%, 60.2% and 51.7% when compared with the published sequences for EcPV1, EcPV2 and EcPV3, respectively. A Bayesian phylogenetic analysis indicated that EcPV4 clusters with EcPV2, but not with EcPV1 and EcPV3. Using the current PV classification system that is based on the nucleotide sequence of L1, we could not define the genus of the newly identified virus. Therefore, a structural analysis of the L1 protein was carried out to aid in this classification because EcPV4 cause lesion similar to the lesion caused by EcPV3. A comparison of the superficial loops demonstrated a distinct amino acid conservation pattern between EcPV4/EcPV2 and EcPV4/EcPV3. These results demonstrate the presence of a new equine PV species and that structural studies could be useful in the classification of PVs.  相似文献

Essential fatty acids should be included in the diet to ensure adequate fish growth. Despite the great number of studies on fatty acid nutrition of fish, there are still several unknowns. The aim of the present study was to investigate fatty acid nutrition of jundiá, a Latin American freshwater catfish. Four diets were formulated containing (i) coconut oil (?C, negative control), (ii) coconut oil + high‐docosahexaenoic‐acid‐fish oil (+C, positive control) and coconut + sunflower + linseed oils at different ratios, producing either (iii) a diet rich in linoleic acid (LA) (HighLA) or 4) a diet low in LA (LowLA). All diets contained significant amounts of saturated fatty acids (at least 57.5% total fatty acids in HighLA) and monounsaturated fatty acids (at least 19.1% total fatty acids in ?C). Diets were fed to jundiá fingerlings (1.5 g) for 70 days; growth, body composition and liver histology were evaluated. The ?C diet, without essential fatty acids, promoted significantly lower fish growth, body fat accumulation and hepatic lipidosis. Fish fed HighLA and LowLA diets presented similar growth as fish fed +C diet. These findings suggest that diet formulations for jundiá catfish fingerlings can include only plant oils without negative effect on growth, survival, body composition, fish health or parameters of feed utilization (ingestion rate and protein utilization).  相似文献

Natural compounds have shown good potential for the discovery of new chemotherapeutics for the treatment of Chagas’ disease. Recently, our group reported the effective trypanocidal activity of (−)-elatol, extracted from the red macroalgae Laurencia dendroidea present in the Brazilian coast against Trypanosoma cruzi. However, the mechanism of action of this compound has remained unclear. There are only hypotheses concerning its action on mitochondrial function. Here, we further investigated the mechanisms of action of (−)-elatol on trypomastigotes of T. cruzi. For this, we evaluated some biochemical alterations in trypomastigotes treated with (−)-elatol. Our results show that (−)-elatol induced depolarization of the mitochondrial membrane, an increase in the formation of mitochondrial superoxide anion and loss of cell membrane and DNA integrity. Additionally, (−)-elatol induced formation of autophagic vacuoles and a decrease in cell volume. All together, these results suggest that the trypanocidal action of (−)-elatol involves multiple events and mitochondria might be the initial target organelle. Our hypothesis is that the mitochondrial dysfunction leads to an increase of ROS production through the electron transport chain, which affects cell membrane and DNA integrity leading to different types of parasite death.  相似文献

Globally, the area of land cultivated with genetically modified (GM) crops has increased a thousand-fold over the last two decades. Although this technology has become important for food production, the regulatory frameworks that underpin these outcomes are based on a list of requirements for a risk assessment that differ from country to country. In recent years, policymakers have had the opportunity to learn from the controversies over transgenics to create effective regulatory milestones for emerging technologies, allowing them to reach their potential for a more sustainable agriculture, ensuring food security. In Brazil, Law No. 11.105 of 24 March 2005 established a framework with four main organizations responsible for risk assessment and management. However, most of new breeding technologies did not exist at that time and were not considered in this law. In 2016, Normative Resolution No. 16 of the National Biosafety Technical Commission (CTNBio) was established to address this gap based on the evaluation of the products obtained through these techniques (termed Innovative Precision Improvement Techniques in the resolution), in a case-by-case consultation system. Briefly, if the product is designated to be a GM, the developer will have to go through the biosafety requirements and will be approved only after CTNBio risk assessment. If the product is designated not to be GM (for the purposes of the legislation), then it can be registered using the existing procedures. Currently, 152 GM products are commercially approved in Brazil. In 2018, CTNBio assessed the first consultation on commercial release of plants generated using the new breeding technologies and has subsequently approved six products. It is expected that many institutions would be able to participate in Brazilian and world markets, developing and introducing new biotechnological solutions and products through a more sustainable approach and without facing public disapproval, a common issue for GM crops.  相似文献

In agricultural fields soil compaction is a major cause of physical degradation. Degree of compactness (DC) is a useful parameter for characterizing compaction and the response of crops for different soils. The objectives of this study were: (1) to identify the critical DC and PR values for soybean [Glycine max (L.) Merrill] using plant growth variables and (2) to verify the relationship between DC and PR, and assess which parameter is recommended for the evaluation of soil compaction. The study was conducted in a greenhouse in a completely randomized factorial design of 4 textures × 5 compaction levels for sandy loam and sandy clay loam soils, and 3 compaction levels for the clayey and very clayey soils. Soil samples were collected from the surface of a Xantic Kandiudox from the NE region of the State of Pará, Brazil. The DC was calculated from the maximum bulk density obtained by the Proctor test, and the PR curve was determined in undisturbed samples equilibrated in different matric potentials. The growth and development of the soybean was favored in the DC range of 80 to 85%, regardless of soil texture. The critical degree of compactness for the growth of soybean was around 98% regardless of soil texture, while the critical values for penetration resistance at field capacity varied according to soil texture and bulk density and were 28.2, 5.6, 3.5, and 5.2 MPa for the sandy loam, sand clay loam, clayey and very clayey soils, respectively. The root length was the plant growth variable most susceptible to soil compaction. Change in soil penetration resistance was poorly related with change in degree of compactness showing that one parameter cannot be replaced by the other. Because PR is quickly determined in field and have a direct relationship with plant growth, for the soils evaluated in this study we recommend the use of soil PR to assess the state of soil compaction.  相似文献

The proteins encoded by ATRX and DAXX participate in chromatin remodeling at telomeres and other genomic sites. Because inactivating mutations of these genes are common in human pancreatic neuroendocrine tumors (PanNETs), we examined the telomere status of these tumors. We found that 61% of PanNETs displayed abnormal telomeres that are characteristic of a telomerase-independent telomere maintenance mechanism termed ALT (alternative lengthening of telomeres). All of the PanNETs exhibiting these abnormal telomeres had ATRX or DAXX mutations or loss of nuclear ATRX or DAXX protein. ATRX mutations also correlate with abnormal telomeres in tumors of the central nervous system. These data suggest that an alternative telomere maintenance function may operate in human tumors with alterations in the ATRX or DAXX genes.  相似文献

Oligodendrogliomas are the second most common malignant brain tumor in adults and exhibit characteristic losses of chromosomes 1p and 19q. To identify the molecular genetic basis for this alteration, we performed exomic sequencing of seven tumors. Among other changes, we found that the CIC gene (homolog of the Drosophila gene capicua) on chromosome 19q was somatically mutated in six cases and that the FUBP1 gene [encoding far-upstream element (FUSE) binding protein] on chromosome 1p was somatically mutated in two tumors. Examination of 27 additional oligodendrogliomas revealed 12 and 3 more tumors with mutations of CIC and FUBP1, respectively, 58% of which were predicted to result in truncations of the encoded proteins. These results suggest a critical role for these genes in the biology and pathology of oligodendrocytes.  相似文献

Coffee is a much enjoyed beverage because of its unique flavor and taste characteristics. The lipid fraction of coffee beans, composed of wax, oil and unsaponifiable matter, prevents volatilization and loss of flavor during the roasting process. The lipid fraction extracted from green beans is high in linoleic acid and has an ultraviolet absorption. These properties that can be very useful and suitable for cosmetic products such as skin moisturizers and sunscreens. The aim of this study was to characterize the lipid fraction and to determine the sun protection factor of 10 species of Coffea. Significant variability was found for all the parameters investigated: the wax content varying between 0.0 and 2.8%, the oil content ranging from 6.9% to 32.4%, unsaponifiable matter from 0.3% to 13.5% and the sun protection factor from 0.0 to 4.1. Fatty acids widely used in the cosmetics industry, such as linoleic and oleic acid were found to be present in excellent proportion.  相似文献

Increasing plasma estrogen (E) levels during the follicular phase of the estrous cycle trigger the pre-ovulatory surge of gonadotropin-releasing hormone (GnRH)/LH. Noradrenaline (NA)-producing cells of the brain stem are involved in regulating GnRH cells and project to the preoptic area (POA) and bed nucleus of stria terminalis (BnST). Input to GnRH cells may be direct or indirect, via relay neurons in the POA/BnST. To investigate this, we ascertained whether an 1-adrenergic antagonist would block/delay the LH surge in ovariectomised (OVX), E-treated ewes. E benzoate (EB) (50 μg) was injected (i.m.) and Doxazosin (100 nmol/h) or vehicle was infused into the third ventricle 2–26 h after EB injection. Doxazosin reduced the magnitude of the LH surge, but did not affect timing. To determine if NA is released in the POA/BnST of cyclic ewes, we immunostained dopamine-β-hydroxylase (DBH) in terminal fields. Reduced numbers of varicosities staining for DBH indicates release of NA. The number of varicosities immunostained for DBH was reduced in the dorsal and lateral BnST during the follicular phase and during the preovulatory LH surge compared to the luteal phase. These data suggest that noradrenergic mechanisms are involved in generation of the GnRH/LH surge via projections to the BnST and relay to GnRH cells. Since Doxasozin reduced the magnitude of the LH surge in the E-treated OVX ewe, and release of NA in cyclic ewes occurred during the follicular phase of the estrous cycle, we speculate that NA is a permissive factor in surge generation. Thus, increased noradrenergic activity is not a trigger mechanism for initiation of the surge.  相似文献

The contents of cholesterol oxides, cholesterol, and total lipid, and the fatty acid composition were determined in frozen turkey meat. The 7-ketocholesterol content varied from 33 microg/100 g in the breast to 765 microg/100 g in the skin, and the levels of 7 beta-hydroxycholesterol varied from not detected in the leg, breast, and skin to 370 microg/100 g in the skin. The values for total lipid (g/100 g) in the wings, legs, breast, and skin were 0.9 +/- 0.4, 1.1 +/- 0.2, 0.5 +/- 0.1, and 12 +/- 3, respectively. The contents for cholesterol (mg/100 g) were 46 +/- 5, 35 +/- 2, 27 +/- 3, and 81 +/- 6 in the wing, legs, breast, and skin, respectively. The main fatty acids identified in all cuts were C18:2n6, C18:1n9, C16:0, C18:0, and C20:4n6.  相似文献
