The cyclic hexadepsipeptide enniatin is known as a phytopathogenic compound from Fusaria causing necrosis and wilt. The molecule consists of three alternating residues each of a branched chain amino acid and D-hydroxyisovaleric acid (D-Hiv). Enniatins are synthesized by a 347kDa multienzyme (enniatin synthetase) via a thiol template mechanism. The corresponding gene esyn1 has an open reading frame of 9393 nucleotides and harbours two modules, one responsible for D-hydroxy acid activation and one for L-amino acid activation with an integrated N-methyltransferase domain. Such methyltransferases build an homologous group among N-methyl peptide synthetases. Enniatins are synthesized by step-wise condensation of dipeptidol building blocks in an iterative manner resembling fatty acid synthesis. A key enzyme in enniatin biosynthesis is the NADPH-dependent D-2-hydroxyisovalerate dehydrogenase, that supplies enniatin synthetase with D-Hiv. Enniatins contribute to the wilt toxic character of Fusaria. Virulence was significantly reduced in F. avenaceum after disruption of the esyn1 gene. 相似文献

Buckwheat is a pseudocereal that has gained increasing interest of industry and consumers over the past decade. Little, however, is known about its dietary fiber composition and nonstarch polysaccharide structures. Analysis of the monosaccharide composition indicated large amounts of pectic polysaccharides in both insoluble and soluble fiber from buckwheat. Methylation analysis gave further insights into the structures of the polysaccharides. The corresponding partially methylated alditol acetates suggested only low amounts of galactans. Xyloglucans were the main hemicellulosic polysaccharides in the insoluble fiber fraction. Highly branched arabinans, exclusively substituted at position O3, were of higher abundance. These results were confirmed by screening endo‐arabinanase and endo‐galactanase liberated oligosaccharides with high‐performance anion‐exchange chromatography with pulsed amperometric detection and LC‐MS2. Application of this method also demonstrated highly branched arabinan areas within the pectic polysaccharides. Only low amounts of fiber associated hydroxycinnamic acids and diferulic acids were liberated by alkaline treatment, and no evidence was found for their attachment to polysaccharides, suggesting only a minor impact of these structural elements on fiber structures in dehulled buckwheat seeds. 相似文献

ABSTRACTInvasive alien plant species (IAPs) represent one of the main biological threats to biodiversity worldwide. Information about their phenotypic plasticity are needed to increase awareness about their future invasive potential. A study about phenotypic plasticity in response to contrasting light regimes and its quantification by a plasticity index (PI) of two IAPs (Ailanthus altissima and Robinia pseudoacacia) inside a Strict Nature Reserve was conducted. R. pseudoacacia showed a 70% higher PI, with a strongly greater value at morphological leaf level, associated with a greater ability to survive and grow in forest understory, explaining its greater widespread. Otherwise, A. altissima showed its highest PI at physiological level, which was associated with the ability to colonize and grow in environments with high-light regimes. Based on these results, the conservative management has limited the presence of A. altissima by its lower ability to grow in forest understory. In fact, the small-scale gaps in the forest infrastructure, that could allow its recruitment, are originated only from the death of a single tree or small group of trees. Regarding R. pseudoacacia, it is critical to maintain this type of management because any disturbances resulting in large openings could further promote its presence inside the Reserve. 相似文献

Interleukin-8 plays a critical role in inflammatory processes. Hence generation of molecules with anti-IL-8 activity is likely to be important for successful feeding and for survival of the ticks. Anti-IL-8 activity was studied in saliva of three ixodid tick species--Dermacentor reticulatus (Fabricius, 1794), Rhipicephalus appendiculatus Neumann, 1901, and Amblyomma variegatum (Fabricius, 1794). The greatest activity was shown in saliva prepared from D. reticulatus. The activity was attributed to tick salivary gland molecules that bind to IL-8, preventing binding of the chemokine to its specific receptor, rather than to occupation of the IL-8 cell receptor by the tick molecules. The distribution of anti-IL-8 activity in fast protein liquid chromatography (FPLC) fractions of salivary gland extracts (SGE) derived from adult female D. reticulatus, R. appendiculatus and A. variegatum was compared directly by both ELISA and receptor-binding inhibition assays. The correspondence in results with fractions of SGE from ELISA is consistent with detection of tick molecules that inhibit IL-8 binding to its receptor. As IL-8 is an important chemoattractant and activator of neutrophils, the presence of an anti-IL-8 activity in tick saliva indicates that neutrophils play an important role in the host response to parasitism by ticks. 相似文献

Little is known about the potentially disrupting effects of wind farms on the habitat connectivity of flying vertebrates at the landscape scale. We developed a regional-scale model to assess the wind farm impact on bat migration and commuting routes. The model was implemented for the bat Nyctalus leisleri in a region of central Italy currently undergoing considerable wind farm development. A Species Distribution Model (SDM) for N. leisleri was generated using the MaxEnt algorithm based on 47 presence records (reduced to 19 after the autocorrelation procedure) and 10 environmental variables derived from topographic and land cover maps. We used the SDM to create a map of connectivity using the software UNICOR to identify potential commuting corridors (PCCs). The incidence of each wind farm on bat flight corridors was assessed by overlaying the existing (380) and planned (195) turbine locations onto the PCCs. The SDM was statistically robust (AUC > 0.8). Most of the corridors were concentrated in the western part of the region, which hosts the largest suitable areas for the species; most of the existing (54 %) and planned (72 %) wind farms interfered with important corridors connecting the western and the eastern parts of the region. Our results provide key information on the impact of the wind farm industry on biodiversity on a regional scale. The novel approach adopted, based on SDM and connectivity analysis, could be easily extended to other flying vertebrates and landscapes and constitutes a promising planning tool necessary for harmonizing the development of renewable energy infrastructures with issues of biodiversity conservation. 相似文献

The aim of this article is the determination of uranium accumulation in plants tissue in shoots and roots of corn—maize (Zea mays), grown on two types of soils, pseudogley and chernozem, together with its phytotoxic effect on the plant growth and development. The soils was contaminated with different rates (10 to 1,000 mg U(VI) kg?1) of uranyl nitrate (UO2(NO3)2·6H2O). Vegetative tests performed with maize indicated uranium phytotoxic effect on plant height, yield, and germination of seeds. This effect was stronger on the plants grown on pseudogley in comparison with those grown on chernozem. Soil properties determined the tolerance and accumulation of U in plants. A linear dependence between the content of uranium in soil and in plants tissue, including maximal content of 1,000 mg U?kg?1, indicates that maize could be used for phytoremediation of uranium-contaminated soils. 相似文献

Both capacitive indicators derived from the water retention curve and dynamic measurements of the flow‐weighted mean pore radius, R0, were used to assess the soil physical quality of two agricultural areas (cropland and olive orchard) and two natural areas (grassland and managed woodlot plantation) potentially subject to soil degradation. The overall idea of the study was to investigate whether a dynamic indicator quantitatively derived from hydraulic conductivity measurements could be used to supplement the traditionally applied capacitive indicators retrieved from water retention measurements. According to the available criteria, only the surface layer of the cropland site showed optimal soil physical quality. In the grassland and woodlot sites, the physical quality was deteriorated also as a consequence of compaction because of grazing. Overall, the physical quality was better in tilled than nontilled soils. The optimal soil in terms of capacitive indicators had hydraulic conductivity close to saturation that was intermediate among the different land uses, and it remained 1·3–1·9 times higher than that observed in the natural sites even when the largest pores emptied. A depth effect on R0 was observed only when larger macropores were activated. It was suggested that water transmission parameters are more affected by changes in large pore domain. The plant available water content and Dexter's S‐index showed inverse statistically significant regressions with R0. The empirical relationships were physically convincing given that, at increasing R0, the contribution of macropores increases, water is transmitted faster below the root zone and the soil's ability to store water is reduced. Diapause is a protective response to unfavorable environments that results in a suspension of insect development and is most often associated with the onset of winter. The ls-tim mutation in the Drosophila melanogaster clock gene timeless has spread in Europe over the past 10,000 years, possibly because it enhances diapause. We show that the mutant allele attenuates the photosensitivity of the circadian clock and causes decreased dimerization of the mutant TIMELESS protein isoform to CRYPTOCHROME, the circadian photoreceptor. This interaction results in a more stable TIMELESS product. These findings reveal a molecular link between diapause and circadian photoreception. 相似文献

To analyze the accuracy of the Klason lignin method as applied for the determination of lignin contents in plant based-food products, Klason lignin preparations from curly kale, pears, whole wheat grains, and corn bran were chemically characterized. Characterization included routine ash and protein determinations and the extraction of fat/waxes as well as cutin/suberin depolymerization and extraction of the liberated monomers. Fat/wax and cutin/suberin amounts in the Klason lignin preparations were determined gravimetrically, and their compositions were analyzed by using GC-MS. Typical fat, wax, and cutin (and suberin) constituents such as saturated and unsaturated fatty acids, hydroxy and/or epoxy fatty acids, and phenolic acids were identified in all samples, whereas the detection of long-chain hydrocarbons, alcohols, and ketones, sterols, stanols, and dioic acids was dependent on the sample analyzed. Estimation of the contribution of non-lignin compounds to the Klason lignin contents reduced the noncorrected Klason lignin contents of the insoluble fibers from 28.7% (kale), 22.8% (pear), 14.8% (wheat), and 9.9% (corn) to maximum lignin contents of 6.5% (kale), 16.4% (pear), 4.9% (wheat), and 2.3% (corn). These data demonstrate that certain commonly used statements such as "cereal brans are highly lignified" need to be revised. 相似文献

Formation of multinucleated giant cells (MGCs) by macrophage fusion is a typical cytopathic effect of lentiviral replication in caprine monocytes and MGC formation from cultured caprine peripheral blood mononuclear cells (PBMCs) has been considered to be diagnostic for small ruminant lentivirus (SRLV) infection. In this study, formation of MGCs was observed after 7–14 days when PBMCs were cultured from healthy goats free from SRLV infection. These MGCs expressed tartrate-resistant acid phosphatase, calcitonin receptor, integrin αVβ3, cathepsin K and matrix metalloproteinase 9 and were able to resorb bone in vitro in the absence of RANKL and macrophage colony stimulating factor, consistent with an osteoclast phenotype. 相似文献
