Organic production of one of the most popular botanical supplements, Echinacea, continues to expand in the U.S. Echinacea seeds typically show a high degree of dormancy that can be broken by ethephon or gibberelic acid (GA), but these methods are currently disallowed in organic production. In order to determine the efficacy of non-chemical seed treatments, we evaluated the effect of varying seed source and supplying light, with and without cold-moist stratification, on seed germination of the three most important medicinal species of Echinacea, E. angustifolia DC, E. purpurea (L) Moench, and E. pallida (Nutt.) Nutt. Treatments included cold-moist stratification under 24 h light, 24 h dark, and 16/8 h light/dark to break seed dormancy. We found that germination was greater in the E. purpurea and E. pallida seeds from a commercial organic seed source compared to a public germplasm source. When seeds were not cold-moist stratified, 16-24 h light increased germination in E. angustifolia only. Echinacea angustifolia, E. purpurea, and E. pallida seeds that were cold-moist stratified under 16-24 h of light for 4 wk had a significantly greater percentage and rate of germination compared to seeds germinated in the dark. Therefore, cold-moist stratification under light conditions is recommended as a method to break seed dormancy and increase germination rates in organic production of Echinacea. 相似文献
The aim was to verify the effect of follicle‐stimulating hormone (FSH) supplementation to α‐MEM+ or TCM199+ media on the in vitro development of ovarian preantral follicles (PFs) derived from collared peccaries. Ovaries (n = 5 pairs) were collected and divided into fragments destined to control group (non‐cultured) or treatments that were cultured for 7 days. The PFs morphology, growth and activation were evaluated by classical histology. The immunohistochemistry markers Ag‐NOR and PCNA were used for nuclear proliferation analysis, and the picrosirius red labelling was used for ovarian extracellular matrix (ECM) evaluation. After 7‐day culture, only the TCM199+ treatment maintained the proportion of intact PFs similar to day 1 (63.2%), but no differences were found among treatments (p > .05). In addition, a significant increase in the growing follicles proportion was verified for all the treatments, indicating follicular activation (p > .05). By the Ag‐NOR analysis, only the TCM199+/FSH maintained the nuclear proliferation similar to the first day (p > .05). The picrosirius red staining revealed that the ECM remained intact in all the treatments (p > .05). We suggest the use of TCM199+ medium supplemented of FSH for the in vitro development of peccaries PFs under 7‐day culturing conditions. 相似文献
The comparative accuracy of 4 biochemical diagnostic kits (API, Enterotube, Minitek, and Pathotec) and the conventional (AOAC) tube system for identifying primarily Salmonella and other enteric isolates was collaboratively studied. Each of 11 participating analysts received 40 foodborne isolates (25 Salmonella and 15 non-Salmonella cultures), representing a total of 440 cultures examined by each identification system. In decreasing order of accuracy, the overall number of correctly identified cultures with each of the systems was as follows: AOAC, 423 (96.1%), Minitek, 403 (91.6%), Enterotube, 395 (89.8%), API, 394 (89.5%), and Pathotec, 373 (84.8%). A cost analysis showed that all 4 diagnostic kit systems were less expensive than the conventional AOAC tube system for a single culture identification. Three of the diagnostic kits have been adopted as official first action as alternatives to the AOAC biochemical tube system for presumptive generic identification of foodborne Salmonella and for screening and eliminating non-Salmonella isolates. Routine incorporation of any one of the 3 diagnostic kits, however, should be preceded by the demonstration in the analyst's own laboratory of adequate correlation between the kit and the AOAC system. 相似文献
The effects of high dose of microbial phytase and an emulsifier on the performance, apparent metabolisable energy (AME) and nitrogen (N) retention in broilers fed on diets containing different fat sources were examined in a 5-week trial. Two fat sources (soy oil and tallow), two inclusion levels of E. coli phytase (500 or 1000 phytase units (FTU)/kg diet) and two inclusion levels of lysolecithin emulsifier (0 or 3.5 g/kg of diet) were evaluated in a 2 × 2 × 2 factorial arrangement of treatments.
Throughout the 5-week trial, soy oil supplementation improved weight gain and feed per gain compared with tallow, but had no effect on feed intake.
The high dose of phytase increased the weight gain and feed intake and lowered the feed per gain during d 1–21, but had no effect on performance parameters over the whole trial period.
An effect of emulsifier was observed for feed intake during d 1–21 and over the whole trial period. Addition of emulsifier increased feed intake compared with diets without emulsifier.
During weeks 1, 2, 3 and 5, birds fed on soy oil–based diets had higher nitrogen-corrected AME (AMEN) compared with those fed on tallow-based diets. During weeks 2, 3 and 5, the effect of phytase was significant for AMEN, with the high dose increasing the AMEN. During week 2, AMEN was increased with emulsifier addition.
During weeks 1, 2, 3 and 5, birds fed on soy oil–based diets had higher fat retention compared with those fed on tallow-based diets. The high dose of phytase improved the retention of fat during week 5 and the addition of emulsifier resulted in higher fat retention during week 1.
During weeks 2, 3 and 5, an interaction between fat source × phytase × emulsifier was observed for N retention. In soy oil–based diets, emulsifier plus 1000 FTU/kg phytase increased N retention compared with other groups, while in tallow-based diets, emulsifier addition increased N retention in diets with 500 FTU/kg, but not in 1000 FTU/kg diet.
Overall, the present data suggest that the dietary fat source influenced performance, AMEN and fat retention in broiler chickens. There is opportunity to improve bird performance during d 1–21, AMEN and fat retention with higher doses of microbial phytase. Addition of the emulsifier increased the AMEN during week 2 and tract retention of fat during week 1, but this effect was not translated into improvements in performance.
In the Upper Zambezi and Okavango ecoregions, Brycinus lateralis, Hepsetus cuvieri, Schilbe intermedius and Serranochromis macrocephalus are important in subsistence fisheries, while S. intermedius and S. macrocephalus are often caught in commercial catches. Despite their importance, there is little information on their age and growth and age validation. Growth zone deposition rate in otoliths of B. lateralis, H. cuvieri, S. intermedius and S. macrocephalus was validated as annual using edge analysis in this study. Annual deposition of growth zones was corroborated by a fluorochrome marking experiment. Both methods demonstrated that one growth zone was deposited annually. Edge analysis demonstrated that the highest proportion of opaque zones was detected between August and December, most likely as a result of slow growth during lower temperature water during the preceding winter. With growth zone deposition validated as annual, future research should focus on determining the growth, maturity and mortality rates of these species, parameters which are important for their management. 相似文献
We investigated the effects of partial root-zone drying (PRD) applied at different periods on leaf water relations, vegetative development, fruit yield, must and wine quality in wine grapes (Vitis vinifera L. cv. Monastrell) during a 3-year field experiment in order to determine the importance of the timing of PRD application on physiological and agronomical vine response under semiarid conditions. Two irrigation treatments were applied: conventional drip irrigation (CI) and PRD. Both treatments received the same annual water quantity. Each year the PRD treatment was applied at different periods of the growth cycle. In 1999 PRD was applied from veraison to harvest (end July–early September); in 2000 from fruit set to harvest (mid June–early September); and in 2001 PRD from budburst to harvest (mid April–early September). Leaf water relations and gas exchange during the experimental period were not significantly affected by PRD treatment. In 1999 and 2000 there was no significant treatment effect on vegetative development, yield or fruit quality. However, in 2001 (when PRD was applied from budburst to harvest), reproductive and vegetative development was clearly altered in PRD vines. Fruit set percentage and vegetative development (shoot length, pruning weight and primary and lateral leaf area) were significantly increased in PRD vines compared to CI. This resulted in both higher yield (kg per vine) (43%) and water use efficiency (40%) compared to CI vines. Berry number per cluster and cluster weight were also significantly increased in PRD vines. Notwithstanding higher yield in PRD vines and a similar berry size, the must and wine quality was not significantly altered, indicating a higher synthesis and accumulation of photoassimilates and metabolites in the berries of PRD vines. We conclude that there was an positive effect on vegetative and reproductive growth when long-term PRD was applied from the beginning of growing season (budburst), suggesting that early onset of PRD is desirable to intensify PRD response under these semiarid conditions. Nevertheless from these results we need to further investigate the long- and short-term effects of PRD, with moderate water amounts, on vegetative and reproductive development such as flowering and fruit set processes in wine grapes. 相似文献
Adsorption, incubation and soil-column experiments with bentazone [3-isopropyl-1H-2,1,3-benzothiadiazin-4(3H)-one 2,2-dioxide] were carried out in ten different soils from the marches surrounding the Doñana National Park (Huelva, SW Spain). Adsorption isotherms for the different soils showed a good fit with the Freundlich equation. Bentazone was poorly adsorbed in all the soils studied, with no significant relationship between theKf values and soil characteristics. A significant correlation was obtained between the soil organic matter content and the distribution constant values (Kd) calculated at an equilibrium concentration of 200 μg cm−3. The low adsorption and non-degradation of bentazone on these soils suggest that the herbicide readily percolates through soils to reach the surface and ground waters. The mobility of bentazone through three soil columns was also studied. The mass balances carried out showed that bentazone was totally eluted from the soil columns. The theoretical model applied to explain bentazone leaching under our experimental conditions seems to be suitable for soil columns with a uniform water-flow rate. 相似文献
It is well known that healthy gut microbiota is essential to promote host health and well‐being. The intestinal microbiota of endothermic animals as well as fish are classified as autochthonous or indigenous, when they are able to colonize the host's epithelial surface or are associated with the microvilli, or as allochthonous or transient (associated with digesta or are present in the lumen). Furthermore, the gut microbiota of aquatic animals is more fluidic than that of terrestrial vertebrates and is highly sensitive to dietary changes. In fish, it is demonstrated that [a] dietary form (live feeds or pelleted diets), [b] dietary lipid (lipid levels, lipid sources and polyunsaturated fatty acids), [c] protein sources (soybean meal, krill meal and other meal products), [d] functional glycomic ingredients (chitin and cellulose), [e] nutraceuticals (probiotics, prebiotics, synbiotics and immunostimulants), [f] antibiotics, [g] dietary iron and [h] chromic oxide affect the gut microbiota. Furthermore, some information is available on bacterial colonization of the gut enterocyte surface as a result of dietary manipulation which indicates that changes in indigenous microbial populations may have repercussion on secondary host–microbe interactions. The effect of dietary components on the gut microbiota is important to investigate, as the gastrointestinal tract has been suggested as one of the major routes of infection in fish. Possible interactions between dietary components and the protective microbiota colonizing the digestive tract are discussed. 相似文献
