Innovative approaches to genome editing in avian species   总被引:1,自引:0,他引:1  
The tools available for genome engineering have significantly improved over the last 5 years, allowing scientist to make precise edits to the genome. Along with the development of these new genome editing tools has come advancements in technologies used to deliver them. In mammals genome engineering tools are typically delivered into in vitro fertilized single cell embryos which are subsequently cultured and then implanted into a recipient animal.In avian species this is not possible, so other methods have been developed for genome engineering in birds. The most common involves in vitro culturing of primordial germ cells(PGCs), which are cells that migrate through the embryonic circulatory system to the developing gonad and colonize the gonad, eventually differentiating into the gonadocytes which produce either sperm or ova. While in culture the PGCs can be modified to carry novel transgenes or gene edits, the population can be screened and enriched, and then transferred into a recipient embryo. The largest drawback of PGC culture is that culture methods do not transfer well across avian species, thus there are reliable culture methods for only a few species including the chicken. Two newer technologies that appear to be more easily adapted in a wider range of avian species are direct injection and sperm transfection assisted gene editing(STAGE).The direct injection method involves injecting genome engineering tools into the circulatory system of the developing embryo just prior to the developmental time point when the PGCs are migrating to the gonads. The genome engineering tools are complexed with transfection reagents, allowing for in vivo transfection of the PGCs. STAGE utilizes sperm transfection to deliver genome engineering tools directly to the newly fertilized embryo. Preliminary evidence indicates that both methodologies have the potential to be adapted for use in birds species other than the chicken, however further work is needed in this area.  相似文献

The aim of the present study was to define whether elevated rumen-undegradable protein (RUP) and tannin-rich peanut skin (PS) supplementation would affect animal growth performance, average daily gain (ADG), blood metabolites, carcass traits associated with lipogenic and immune-related gene expressions in meat goats grazing winter wheat (WW). Thirty-six Kiko-crossbreed male goats at approximately 6 months of age were blocked by body weight (BW; 25.6 ± 1.1 kg) and randomly assigned to one of the four treatments with two replicates based on a 2 × 2 factorial design. Diets contained PS replacing alfalfa meal (ALM), without or with RUP supplementation. Both PS and ALM were incorporated into grain mix portion of the diet and pelletized, with remaining diets fed ad libitum of WW forage for a period of 51 days. Lipogenic genes examined included SCD, ACLY, YWHAZ, PPIA and FABP4, while immune-related genes examined included ACTB (as a control gene), H3F3A, PPIA, IRF3, STAT2, HERC3 and IFIT3 antibody genes. The meat goats on PS-pellet-supplemented group with or without RUP supplementation grew 38.5% faster ADG (p < .001) when compared to control-supplemented group. When goats received PS diet, empty body weight, hot carcass, cold carcass, shoulder, hind shank, rack, loin and fat thickness were greater (p < .05) than control diet. Animals on PS-pellet had higher ACLY, YWHAZ, PPIA and FABP4 gene expression (p < .05) when compared to ALM-pellet control, with RUP by PS-pellet interactions (p < .01). Goats receiving additional RUP supplementation had increased (p < .05) STAT2 gene expression, whereas goats receiving PS-pellet supplementation showed increased STAT2 (p < .05) and a tendency to increase IRF3 (p = .07) gene expressions. In conclusion, the addition of PS-pellet or RUP supplementation has the potential to improve ADG and altered selected lipogenic and immune-related gene expressions.  相似文献

Research on the nutrition transition often treats dietary changes as an outcome of increased trade and urban living. The Northern Food Crisis presents a puzzle since it involves hunger and changing diets, but coincides with a European ban on trade in seal products. I look to insights from economic sociology and decolonizing scholarship to make sense of the ban on seal products and its impacts. I examine how trade arrangements enact power imbalances in ways that are not always obvious. I explain how the ban’s exemption for Inuit-produced seal goods explicitly aims to protect Inuit from the harshness of capitalism and preserve their traditions. In this respect, the Northern Food Crisis is an embodiment of European visions of who Inuit are expected to be and how they are supposed to act in the global economy.  相似文献

One challenge for statisticians and epidemiologists in projecting the future health risks of climate change is how to estimate exposure-response relationships when temperatures are higher than at present. Low dose extrapolation has been an area of rich study, resulting in well-defined methods and best practices. A primary difference between high dose and low dose extrapolation of exposure-response relationships is that low dose extrapolation is bounded at no exposure and no (or a baseline) response. With climate change altering weather variables and their variability beyond historical values, the highest future exposures in a region are projected to be higher than current experience. Modelers of the health risks of high temperatures are making assumptions about human responses associated with exposures outside the range of their data; these assumptions significantly affect the magnitude of projected future risks. Further, projections are affected by adaptation assumptions; we explore no adaptation (extrapolated response); individual (physiological) adaptation; and community adaptation. We present an example suggesting that linear models can make poor predictions of observations when no adaptation is assumed. Assumptions of the effects of weather above what has been observed needs to be more transparent in future studies. Statistical simulation studies could guide public health researchers in identifying best practices and reducing bias in projecting risks associated with extreme temperatures. Epidemiological studies should evaluate the extent and time required for adaptation, as well as the benefits of public health interventions.  相似文献

More diverse sources of energy are needed for countries to progress toward energy independence and to meet future food production needs. The US Task Force on Strategic Unconventional Fuels concluded that to achieve this objective it is essential to develop a domestic unconventional fuels industry. Rangelands, which cover 50% to 70% of the earth's terrestrial surface and dominate much of the western half of the United States, represent a major source of alternative energy resources. A framework to systematically identify biophysical-socioeconomic links that influence the delivery of ecosystem services affected by alternative uses of rangelands has been lacking. The Integrated Social, Economic, and Ecological Conceptual framework was developed by the Sustainable Rangeland Roundtable to address this deficiency. We apply this framework to demonstrate how the effect on ecosystem services of exploiting rangeland-based biofuel, natural gas, and wind energy resources can be systematically compared. We also demonstrate the use of this framework for selecting suitable indicators to monitor changes in the biophysical-socioeconomic links affected by the development of these unconventional energy sources. This type of approach can potentially enhance coordination between federal, state, and local agencies that are attempting to set polices and regulations for the sustainable development of unconventional energy resources on rangelands.  相似文献

Blood glucose was found to be below the reference range in most farmed elk calves examined at the Western College of Veterinary Medicine teaching hospital. Hypoglycemia was associated with diarrhea, sepsis, and malnutrition. Blood glucose analysis and, when indicated, intravenous dextrose administration are recommended when treating moribund farmed elk calves.  相似文献
