Wheat yellow rust (WYR), caused by Puccinia striiformis f. sp. tritici (PST), is a major disease of wheat, and deployment of a single cultivar often leads to disease epidemics. Effect of inoculum level, foliar fungicide spray, and wheat cultivar mixtures were evaluated on disease development in the field and greenhouse in Nepal. Treatments were arranged in a split–split plot design with three replications in both experiments. Two inoculum levels of PST (low and high) were main plot factors; nontreated control and foliar spray of fungicides (Mancozeb and Bayleton) were subplot factors; and two-component cultivar mixtures, composed of different ratios of a susceptible (S) and a resistant (R) cultivars (90:10, 80:20, and 50:50, 100:0, and 0:100) were sub–subplot factors. WYR severity was assessed at different time intervals, and disease development was calculated as area under the disease progress curve (AUDPC). Inoculum level did not cause significant differences in AUDPC in the field but did in the greenhouse. Foliar spray of fungicides reduced the AUDPC in the greenhouse and field. In both experiments, AUDPC values were low in cultivar mixtures compared with a pure stand of a susceptible cultivar. As the proportion of resistant cultivar increased compared with the susceptible cultivar in the S:R mixture component, disease severity decreased with a consequent increase in grain yield. The greater yield obtained with cultivar mixtures compared with only the susceptible cultivar, independent of inoculum level and fungicide spray in the field, revealed a promising strategy to manage WYR in Nepal.  相似文献

Commercial turkey eggs, free of antibodies to avian metapneumovirus subtype C (aMPV/C), were inoculated with aMPV/C at embryonation day (ED) 24. There was no detectable effect of virus inoculation on the hatchability of eggs. At 4 days post inoculation (DPI) (the day of hatch (ED 28)) and 9 DPI (5 days after hatch), virus replication was detected by quantitative RT-PCR in the turbinate, trachea and lung but not in the thymus or spleen. Mild histological lesions characterized by lymphoid cell infiltration were evident in the turbinate mucosa. Virus exposure inhibited the mitogenic response of splenocytes and thymocytes and upregulated gene expression of IFN-γ and IL-10 in the turbinate tissue. Turkeys hatching from virus-exposed eggs had aMPV/C-specific IgG in the serum and the lachrymal fluid. At 3 week of age, in ovo immunized turkeys were protected against a challenge with pathogenic aMPV/C.  相似文献

The H3N2 triple reassortant (TR) influenza viruses emerged in swine in 1998 and then in turkeys in 2003. It was then hypothesized that these viruses crossed the species barrier and transmitted from pigs to turkeys. In previous work we identified viruses with different transmission behavior between the two species, of which A/turkey/Ohio/313053/04 (TK04) transmitted both ways between swine and turkeys, and A/swine/North Carolina/03 (SW03) did not transmit either way between the two species. Utilizing the 12-plasmid reverse genetics (RG) system, we rescued two viruses (TK04 and SW03) with potentially different transmission behavior between pigs and turkeys. Single gene reassortants (SGR) were generated by switching the hemagglutinin (HA) or the neuraminidase (NA) genes between both viruses, and were evaluated for replication in vitro (pig and turkey tracheal/bronchial epithelial cells) and in vivo (pigs and turkeys). RG-created TK04 replicated more efficiently than SW03 in vitro and in vivo. Additionally, TK04 exhibited better binding affinity to plasma membrane preparations (PMP) from pig and turkey tracheal/bronchial epithelial cells compared to SW03. In study with SGR viruses, the HA protein was found to be essential for TK04 virus transmission amongst turkeys, but not sole factor contributing to the efficient replication of virus in turkeys and pigs. Such findings further highlight the polygenic nature of influenza virus pathogenesis.  相似文献

There is an increasing recognition of the contribution of forests to food security of poor and marginalized people. However, empirical findings remain limited on how forests contribute to food security. Drawing on four case studies of community forestry in Nepal, this paper discusses pathways through which forests are contributing to food security needs of local communities. The evidence presented here was gathered through 4 years of action research and draws insights from the past 40 years of Nepal’s community forestry practice, which is often regarded as a successful case of conservation and development. It is shown that there are four distinct pathways through which community forests contribute to food security as a source of: (1) income and employment; (2) inputs to increase food production; (3) directly for food; and (4) renewable energy for cooking. Despite emerging pathways linking forest management to food systems at the local level, forestry policies and institutions have neither explicitly recognized nor strengthened the linkage between forest and food security. The paper highlights that there is a need for a fundamental shift in thinking from the conventional notion of ‘forests for soil conservation’ to ‘sustainable forest management for food security’.  相似文献

Tropical theileriosis, caused by Theileria annulata, is an important tick-borne disease of cattle. A cell culture attenuated vaccine has been developed in our laboratory by long-term in vitro propagation of the schizont stage of the parasite. A longitudinal study was conducted at selected farms housing indigenous, cross-bred and exotic animals to investigate the effect of vaccination on the epidemiology of the disease. A total of 120 animals in 4 age groups were vaccinated with the vaccine before the onset of disease season. An equal number of age-matched animals were kept as controls at the same sites. Animals were monitored for 14 months at monthly intervals. The 97.5% vaccinated animals showed a rise in antibody titres 1 month post-vaccination, as determined by single dilution ELISA. The 78.3% of non-vaccinated animals became sero-positive over the period of observation. Mean antibody titres were significantly higher in vaccinated than non-vaccinated animals. Cross-bred animals showed higher antibody titres followed by exotic and indigenous animals in both the vaccinated and non-vaccinated groups. However, the antibody titres in animals of different ages were similar. The 36.7% vaccinated and 64.2% non-vaccinated animals became carriers (<0.5% piroplasms in erythrocytes) during the observation period. Clinical cases of theileriosis were recorded only in the non-vaccinated group suggesting that vaccinated animals were sufficiently immune to withstand field tick challenge for at least 14 months.  相似文献

The spatial and temporal variations commonly found in the infiltration characteristic for surface-irrigated fields are a major physical constraint to achieve higher irrigation application efficiencies. Substantial work has been directed towards developing methods to estimate the infiltration characteristics of soil from irrigation advance data. However, none of the existing methods are entirely suitable for use in real-time control. The greatest limitation is that they are data intensive. A new method that uses a model infiltration curve (MIC) is proposed. In this method a scaling process is used to reduce the amount of data required to predict the infiltration characteristics for each furrow and each irrigation event for a whole field. Data from 44 furrow irrigation events from two different fields were used to evaluate the proposed method. Infiltration characteristics calculated using the proposed method were compared to values calculated from the full advance data using the INFILT computer model. The infiltration curves calculated by the proposed method were of similar shape to the INFILT curves and gave similar values for cumulative infiltration up to the irrigation advance time for each furrow. More importantly the statistical properties of the two sets of infiltration characteristics were similar. This suggests that they would return equivalent estimates of irrigation performance for the two fields and that the proposed method could be suitable for use in real-time control.  相似文献

ABSTRACT: Infectious bursal disease (IBD) is an important immunosuppressive disease of chickens. The causative agent, infectious bursal disease virus (IBDV), consists of two serotypes, 1 and 2. Serotype 1 consists of classic IBDV (cIBDV) and variant IBDV (vIBDV). Both of these strains vary in antigenicity and pathogenesis. The goal of this study was to compare the immunopathogenesis of cIBDV and vIBDV. Three-week-old specific pathogen free chickens were inoculated intraocularly with standard challenge strain (STC) (cIBDV) and a variant strain Indiana (IN) (vIBDV). The cIBDV produced more pronounced bursal damage, inflammatory response and infiltration of T cells as compared to vIBDV. There were significant differences in the expression of innate (IFN-α and IFN-β), proinflammatory cytokine and mediator (IL-6 and iNOS) in cIBDV- and vIBDV-infected bursas. The expression of chemokines genes, IL-8 and MIP-α was also higher in cIBDV-infected chickens during the early phase of infection. The expression of Toll like receptor 3 (TLR3) was downregulated at post inoculation days (PIDs) 3, 5, and 7 in the bursas of vIBDV-infected chickens whereas TLR3 was upregulated at PIDs 3 and 5 in cIBDV-infected bursas. In vIBDV-infected bursa, TLR7 expression was downregulated at PIDs 3 and 5 and upregulated at PID 7. However, TLR7 was upregulated at PIDs 3 and 7 in cIBDV-infected bursas. The expression of MyD88 was downregulated whereas TRIF gene expression was upregulated in cIBDV- and vIBDV-infected bursa. These findings demonstrate the critical differences in bursal lesions, infiltration of T cells, expression of cytokines, chemokines and TLRs in the bursa of cIBDV-and vIBDV-infected chickens.  相似文献

The responses were monitored of young crossbred calves vaccinated against tropical theileriosis during the winter against a field tick challenge in the disease season. Thirty-eight calves below 2 months of age, born after the end of the disease season, were selected at an organized farm. Twenty-five animals were vaccinated with Theileria annulata (Hisar) cell culture vaccine (developed at CCS HAU Hisar laboratory) after the end of the disease season and 13 calves were kept as non-vaccinated controls. These calves were observed for their susceptibility to theileriosis in the new disease season. There was an increase in antibody titre in 18 of the 25 vaccinated animals one month after vaccination. The antibody titre then declined gradually, but remained higher than those of the non-vaccinated animals at month 0. No fever or other clinical signs of tropical theileriosis were observed in any of the vaccinated animals. Nine out of 25 (36%) vaccinated calves showed occasional piroplasms (<;0.5%) in blood smears. All the vaccinated animals withstood the field tick challenge. On the other hand, 9 of the 13 (69%) unvaccinated calves exhibited occasional piroplasms, and included three clinical cases of tropical theileriosis. These observations suggest that young crossbred calves vaccinated with the T. annulata (Hisar) cell culture vaccine at the end of the disease season were relatively resistant during the next disease season.  相似文献
