The influence of genotypic differences in resistance to infection from Lymphoid Leukosis (LL) on mortality and egg production was studied in three pairs of sublines. One subline of each pair had been selected for resistance and the other for susceptibility to infection. At 10 months of age, almost no hens with neutralising antibodies against LL were found in the resistant sublines, while in each susceptible subline more than 40% of the hens had LL antibodies. At 21 months of age, very few hens of the resistant sublines had group-specific (gs) antigen in their eggs. Susceptible sublines differed considerably in this respect, from 81% gs antigen-positive hens in one subline to 9% or no positive hens in the others. On the whole, the susceptible sublines had 6% higher mortality during the laying period (8 months) but the size of the difference was influenced by vaccination against Marek's disease (MD) and varied considerably between pairs of sublines. Significant interactions between strain and genotype of resistance to LL infection were observed for mortality from "other causes" in hens vaccinated against MD, as well as for mortality from "Leukosis" and from "MD" in non-vaccinated hens. The average rate of lay from 1 to 8 months was 8% lower in susceptible hens. In the subline with the highest frequency of eggs containing LL group-specific antigen in the albumen, the reduction of laying rate was twice as large as in the other susceptible sublines. 相似文献
Cell cultures infected with BHV-1/F(syn), a recombinant bovine herpesvirus 1 (BHV-1) which expresses a synthetic open reading frame encoding the fusion (F) protein of the bovine respiratory syncytial virus (BRSV), showed a cytopathic effect (CPE) indistinguishable from that induced by wildtype BHV-1 although transient transfection experiments demonstrated that expression of the F protein leads to formation of large syncytia. Since it has been shown that glycoprotein M (gM) of pseudorabies virus inhibits BRSV F-induced syncytium formation in transient plasmid transfection experiments [Pseudorbies virus glycoprotein M inhibits membrane fusion. J. Virol. 74 (2000) 6760], the gM ORF of wtBHV-1 and BHV-1/F(syn) was interrupted. Infection of cell cultures with the resulting gM(-) mutant of BHV-1/F(syn) led to formation of syncytia, whereas the CPE in gM(-)BHV-1 infected cells was comparable to the CPE in wtBHV-1 infected cultures. Our results demonstrate that gM is not essential for BHV-1 replication in cell culture and that gM is involved in inhibition of the cell fusion activity of the BHV-1 expressed BRSV F protein. 相似文献
OBJECTIVE: To determine the diagnostic value of aerobic microbial culture and cytologic evaluation of corneal specimens in the diagnosis of infectious ulcerative keratitis (IUK). DESIGN: Prospective study. ANIMALS: 48 animals (26 dogs, 13 horses, 7 cats, 1 bird, and 1 llama) with corneal ulcers. PROCEDURE: Scrapings from corneal ulcers were examined cytologically. Corneal swab specimens were submitted for microbial culture. Animals were grouped according to whether they had been receiving antimicrobials at the time of admission. RESULTS: Of the 38 animals receiving antimicrobials, 19 had positive results for IUK on cytologic evaluation, 20 on microbial culture, and 26 on cytologic evaluation, microbial culture, or both. Of the 10 animals not receiving antimicrobials at the time of admission, 7 had positive results for IUK on cytologic evaluation, and 9 had positive results on microbial culture. In this group of 10 animals, additional animals with IUK were not identified on the basis of cytologic evaluation alone. When all 48 animals were considered irrespective of antimicrobial treatment, 26 and 29 had positive results for IUK on cytologic evaluation and microbial culture, respectively, whereas IUK was confirmed in 35 animals on the basis of cytologic evaluation, microbial culture results, or both. CONCLUSIONS AND CLINICAL RELEVANCE: Microbial culture and cytologic evaluation of corneal specimens maximizes identification of IUK, especially in animals receiving antimicrobial treatment. Because of serious consequences of untreated IUK, we recommend that both diagnostic tests be used to tailor treatment and reduce risk of vision impairment in animals. 相似文献
BACKGROUND: Sepsis is still a major cause of death in both human and veterinary medicine. Early diagnosis is essential for appropriate treatment. Identification of patients at risk for developing sepsis is already possible in human medicine through the measurement of plasma interleukin-6 (IL-6) levels. In veterinary medicine, however, this has been investigated only in canine experimental models. OBJECTIVES: The purpose of this study was to measure IL-6 plasma levels in dogs with naturally occurring systemic inflammatory response syndrome (SIRS) and sepsis and to analyze the value of IL-6 as a predictive parameter for severity and mortality. METHODS: Included in the study were 79 dogs that had been admitted to the small animal clinics of Munich and Berlin from July 2004 to July 2005 and that satisfied the diagnostic criteria for SIRS and sepsis as defined using established parameters. Measurement of plasma IL-6 levels on days 0, 1, and 2 was performed by the use of a colorimetric bioassay based on IL-6-dependent cell growth. RESULTS: Septic foci were identified in 43 patients (septic group), and 36 patients were enrolled in the SIRS group. The frequency of positive blood cultures was 11%. The overall mortality rate was 48%. Higher plasma IL-6 levels on the day of admission were significantly correlated with a more severe degree of disease, increased mortality rate, and earlier fatality. CONCLUSIONS: Plasma IL-6 concentration is predictive of outcome in canine SIRS and sepsis and may be a valuable laboratory parameter for assessing critically ill dogs. 相似文献
1. Sudden Death Syndrome (SDS) is a disease of well‐developed, predominandy male broiler chickens where death appears to occur because of cardiovascular failure. The role of skeletal muscle sarcoplasmic calcium regulation as a potential cause of SDS has been investigated.
2. Calcium regulation of skeletal muscle sarcoplasmic reticulum was compared between broiler and Leghorn chickens. Calcium regulation matured from the 2nd to the 11 th week and, at any age, broiler chickens showed significandy lower calcium transport rates and transport efficiencies. The mechanism of calcium transport in broiler chickens was more energy‐consuming than that of the Leghorn chickens.
3. Sarcoplasmic calcium regulation is pivotal for muscle metabolism. As in porcine malignant hyperthermia, weaker calcium regulation might lead to hyperactivation of skeletal muscle, followed by elevated lactic acid concentrations and cardiovascular failure. 相似文献
In the sandy soils of northeast Thailand, root development is generally limited to the topsoil (0–20 cm depth) but a simple slotting intervention (20–40 cm) significantly increased the root frequency in the slotted material (Eslot) compared with the undisturbed subsoil (E horizon). The aim of this study was to investigate the consequences of slotting on the soil structure by analysing at different scales the pore characteristics of the original soil profile and of the soil material inside the slot. These characteristics were studied using bulk density measurements, image analysis of thin sections and mercury porosimetry. Our results showed that the total porosity of the E horizon and Eslot material was similar when measured in 100 cm3 cylinders, but that the pore size distribution had been changed by slotting. The unaltered E horizon contained mainly small pores characterized by a narrow distribution related to close packing of the sand grains, associated with some biological macropores probably with poor continuity as they did not contain roots despite their size. On average, pores were larger in the Eslot material, with a broader distribution resulting from looser packing of the sand grains but with fewer biological macropores. Although slotting reduced the number of biological pores, the looser packing appeared to be more favourable to root development than the presence of macropores in the E horizon. Finally, the comparison of the porosity in the different horizons with the porosity of the Eslot material, indicated the significance of the closeness of the sand packing on root development. 相似文献
The genetic structure of Azospirillum amazonense populations isolated from the rhizosphere soil and washed and surface-sterilised roots of rice, maize and sorghum plants, cropped simultaneously in two different soils (clay loam and sandy loam) was characterised. Genetic diversity was measured by restriction fragment length polymorphism of the amplified 16S–23S rDNA intergenic spacer region (RISA-RFLP) and cluster analysis. Four genetically distinct clusters of isolates were observed with 78% similarity, suggesting that the A. amazonense population was heterogeneous at the strain level regardless of the soil type or host plant. Analysis of molecular variance (AMOVA) demonstrated that the host plant had a highly significant selective effect on the genetic structure of this species, especially on those isolates intimately associated with them, but also to a lesser extent on isolates from the rhizosphere and washed roots. The soil type also had a highly significant selective effect on A. amazonense genetic diversity, especially for those isolates from the rhizosphere soil. The selective effect of the soil type combined with that of the host plant suggests that environmental factors, such as soil texture and composition of exudates provided by C3 or C4 plants, play major roles in the overall genetic structure of A. amazonense populations associated with these cereals. 相似文献
Different barley varieties, consisting of hulled and hull-less types, of normal, waxy, and high amylose starch, as well as two-rowed and six-rowed types, were analyzed for their main proanthocyanidins and bound phenolic acids. Variations in proanthocyanidin and phenolic acid contents were studied in different barley types as well as inter-relationships between the phytochemicals and polysaccharides. The main flavanols found in the analyzed barley varieties were two dimeric as well as four trimeric forms in addition to catechin. The total amount of flavanols ranged from 325 to 527 microg/g of fresh weight of barley flour. No evident associations were found between variations in proanthocyanidin levels and different barley types. The total amount of phenolic acids ranged from 604 to 1346 microg/g of fresh weight of barley flour, with ferulic acid as the dominating acid. The amount of phenolic acids varied according to occurrence or lack of hull, with significantly higher levels in the hulled varieties. 相似文献
