A three-phase laboratory procedure suitable for predicting protein degradability in the rumen and digestibility of undegraded protein is reported. In the first phase the feed was incubated with starch and buffered rumen fluid. In the incubation mixture the viability of protease-active bacteria was checked by anaerobic culturing, whereas changes in protease activity were monitored by azocasein degradation. In the second and third phase rumen undegradable protein (UDP) was digested with pepsin and pancreatin, respectively. The measurements showed that 63.2, 5.2 and 4.7% of the crude protein of green lucerne was decomposed by rumen fluid, pepsin and pancreatin, respectively. Degradability of the crude protein of extracted sunflower meal was 68.3, 17.7 and 5.5% in the three phases, respectively. Repeated determination yielded crude protein degradabilities of 66.7, 27.1 and 5.1% for the three phases, respectively. 相似文献

The concept of a spatial-velocity hodograph is introduced to describe quantitatively the extrusion of a carbon tubule from a catalytic particle. The conditions under which a continuous tubular surface can be generated are discussed in terms of this hodograph, the shape of which determines the geometry of the initial nanotube. The model is consistent with all observed tubular shapes and explains why the formation process induces stresses that may lead to "spontaneous" plastic deformation of the tubule. This result is due to the violation of the continuity condition, that is, to the mismatch between the extrusion velocity by the catalytic particle, required to generate a continuous tubular surface, and the rate of carbon deposition. 相似文献

There is a growing interest in highly fermentable dietary fibers having the potential to reduce risks of disease through the production of short-chain fatty acids (SCFA). Recently a digestion-resistant retrograded maltodextrin (RRM), classified as type 3 resistant starch was developed. Systematic work to determine its molecular and physiological properties was carried out to determine (1) the fraction resistant to digestion in vitro and in vivo, (2) its postconsumption effect on blood glucose in healthy volunteers, and (3) its in vitro fermentation pattern, at different ages, by use of pooled fresh human fecal inoculum. RESULTS: The digestion resistant fraction obtained in vivo from ileostomy patients (59.4%) is similar to that obtained by the AOAC method for measuring retrograded resistant starch (59.7%). The relative glycemic response after consumption of 50 g of RRM was 58.5% compared to glucose set as 100%. When exposed to colonic microbiota, in vitro obtained indigestible fractions behave similarly to those obtained in vivo in ileostomy patients. Fermentation of RRM and production of butyric acid is negligible during the first months of life but develops subsequently during weaning. In adults, RRM fermentation results in a high yield of SCFA, with butyrate representing 21-31 mol % of total SCFA. The high yield of SCFA during colonic fermentation, observed from weaning age on, as well as the potential to help reduce glycemic load may be of benefit to a number of health-related functions in the host. Application of the aroma extract dilution analysis on a flavor distillate prepared from freshly ground rye flour (type 1150) revealed 1-octen-3-one (mushroom-like), methional (cooked potato), and (E)-2-nonenal (fatty, green) with the highest flavor dilution (FD) factors among the 26 odor-active volatiles identified. Quantitative measurements performed by stable isotope dilution assays and a comparison to the odor thresholds of selected odorants in starch suggested methional, (E)-2-nonenal, and hexanal as contributors to the flour aroma, because their concentrations exceeded their odor thresholds by factors >100. Application of the same approach on a rye sourdough prepared from the same batch of flour revealed 3-methylbutanal, vanillin, 3-methylbutanoic acid, methional, (E,E)-2,4-decadienal, 2,3-butanedione, and acetic acid as important odorants; their concentrations exceeded their odor thresholds in water and starch by factors >100. A comparison of the concentrations of 20 odorants in rye flour and the sourdough made therefrom indicated that flour, besides the fermentation process, is an important source of aroma compounds in dough. However, 3-methylbutanol, acetic acid, and 2,3-butanedione were much increased during fermentation, whereas (E,E)-2,4-decadienal and 2-methylbutanal were decreased. Similar results were obtained for five different flours and sourdoughs, respectively, although the amounts of some odorants in the flour and the sourdough differed significantly within batches. 相似文献

The aims of the present study were to determine whether salsolinol (SAL), a dopamine-related compound, is present in the bovine posterior pituitary (PP) gland, and to clarify the effect of SAL on the secretion of prolactin (PRL) in ruminants. SAL was detected in extract of bovine PP gland using high-pressure liquid chromatography with electrochemical detection (HPLC-EC). A single intravenous (i.v.) injection of SAL (5 and 10mg/kg body weight) significantly and dose-dependently stimulated the release of PRL in goats (P<0.05). Plasma PRL levels reached a peak 10min after the injection, then gradually returned to basal values in 60-80min. The PRL-releasing pattern was similar to that in response to sulpiride (a dopamine receptor antagonist). The intracerebroventricular (i.c.v.) injection of 1mg of SAL had no significant effect on the release of PRL in calves, however, 5mg significantly stimulated the release (P<0.05) with peak values reached 30-40min after the injection. Moreover, SAL significantly stimulated the release of PRL from cultured bovine anterior pituitary cells at doses of 10(-6) and 10(-5)M, compared to control cells (P<0.05). Taken together, our data clearly show that SAL is present in extract of the PP gland of ruminants, and has PRL-releasing activity both in vivo and in vitro. Therefore, this endogenous compound is a strong candidate for the factor having PRL-releasing activity that has been previously detected in extract of the bovine PP gland. 相似文献

Chocolate is often labeled with percent cocoa solids content. It is assumed that higher cocoa solids contents are indicative of higher polyphenol concentrations, which have potential health benefits. However, cocoa solids include polyphenol-free cocoa butter and polyphenol-rich nonfat cocoa solids (NFCS). In this study the strength of the relationship between NFCS content (estimated by theobromine as a proxy) and polyphenol content was tested in chocolate samples with labeled cocoa solids contents in the range of 20-100%, grouped as dark (n = 46), milk (n = 8), and those chocolates containing inclusions such as wafers or nuts (n = 15). The relationship was calculated with regard to both total polyphenol content and individual polyphenols. In dark chocolates, NFCS is linearly related to total polyphenols (r2 = 0.73). Total polyphenol content appears to be systematically slightly higher for milk chocolates than estimated by the dark chocolate model, whereas for chocolates containing other ingredients, the estimates fall close to or slightly below the model results. This shows that extra components such as milk, wafers, or nuts might influence the measurements of both theobromine and polyphenol contents. For each of the six main polyphenols (as well as their sum), the relationship with the estimated NFCS was much lower than for total polyphenols (r2 < 0.40), but these relationships were independent of the nature of the chocolate type, indicating that they might still have some predictive capabilities. 相似文献

This study was aimed at construction of reference (physiological) value intervals determined by lower 2.5% and upper 97.5% quantiles for total plasma protein (TPP) in farmed rainbow trout, Oncorhychus mykiss (Kamloops strain), aged 10–12 months (n=755). Total plasma protein in male trout from raceway culture was found to be significantly greater (38–58.3 g L?1) relative to females (31–51.5 g L?1). Total plasma protein in trout from cage culture was significantly greater (32–54 g L?1) compared with the raceway culture (31–49 g L?1). Moderate product–moment correlations between TPP and cholesterol (r=0.8730), TPP and total calcium (r=0.8188), TPP and alanine aminotransferase (r=0.6200) and between TPP and aspartate aminotransferase (r=0.6430) were estimated. In order to describe the effect of complementary artificial light supply on the nutrient status of tomato seedlings, fluorescent lamps were compared to each other. Nitrogen (N), phosphorus (P), and potassium (K) concentrations were measured (mg g?1 dry matter). Blue, red and far-red relationships were established. The N, P and K higher concentration and uptake have been found under high efficiency fluorescent lamps. Phosphorus uptake increasing was observed at high levels of irradiation. Potassium concentration is significantly higher under compact fluorescent lamps. The relationship between ratios and N, P and K extractions were used as indicators of limitation for plant. The ratio N/P and N/K present a low relation with N extraction. The ratio N/P presents a significant correlation with P extraction, also the ratios N/K and P/K with K extraction. Organic acids released into the rhizosphere may perform many beneficial functions to the plant including metal detoxification and enhancement of nutrient acquisition. Typically, these organic acids are studied in isolation; however, roots simultaneously exude a cocktail of organic acids and other substances, and their combined impact on rhizosphere processes may be quite different. It has been hypothesized that some exudates may play secondary roles (e.g. inhibitors of microbial activity, blockage of sorption sites), which might enhance the longevity and nutrient-mobilization capacity of others. Here we investigated how the decomposition, sorption and P-solubilizing effects of citrate, malate and oxalate are affected by the presence of malonate and shikimate. We found that in a range of agricultural soils the decomposition of citrate, malate and oxalate was rapid, but not influenced by the presence of large quantities of shikimate or malonate. This suggests that the individual organic acids are taken up by different transport mechanisms or components of the microbial community. At large concentrations, malonate decreased sorption of citrate, malate and oxalate on the soil, whilst shikimate had little effect. The capacity of citrate, malate and oxalate to desorb P was significantly greater in cocktails containing malonate compared with the single organic acid; no effect was seen with shikimate. We conclude that neither malonate nor shikimate at realistic concentrations will significantly affect the biodegradation of citrate, malate or oxalate in the rhizosphere, and while malonate did enhance P desorption, this effect is additive rather than synergistic. Overall, we found little evidence that malonate and shikimate act as secondary regulators of citrate, malate and oxalate behavior in soil. 相似文献

Interest in native species is growing across the tropics as reforestation of degraded lands becomes more widespread. In this study four tree species native to Panama - Cedrela odorata, Pachira quinata, Samanea saman, and Tabebuia rosea - were grown on rural farms at two dry tropical sites in Panama for up to five years. Survivorship and growth data at these “on-farm” trials are compared to data recorded from nearby experimental or “species selection” trial sites and evaluated in terms of soil fertility and management. Participant farmers were also asked about their interest in planting trees in general as well as their interest in 61 species grown in the species selection trial.Although, on-farm survivorship was variable and generally lower than that found on the species selection trial, one species (S. saman) experienced high and consistent survivorship. High survivorship combined with growth data from farms at both sites for this species suggests it would be a good candidate for extension projects working with rural farmers. Survivorship of other species appears more sensitive to farmer management and/or local site conditions. Generally lower growth on the Los Santos farms as compared to the species-selection trial is attributed to the lower soil fertility (plant available P) at the on-farm sites compared to the species selection trial. In contrast, only one species - P. quinata - had a growth variable found to be significantly lower between the on-farm and species selection trial sites in Rio Hato. C. odorata, P. quinata T. rosea can all be used in on-farm conditions with consideration to specific site and management conditions.By 2009, approximately 80% of the farmers planting trees still wished to participate in tree planting activities. All of the farmers no longer wishing to continue with the project expressed slow growth rates of trees as a principal reason. All but one of these farmers had growth rates for his/her trees markedly below those of the species selection trial nearby. Some farmers wishing to continue had very high mortality rates (>70% for all species), suggesting non-tangible benefits for participating in project activities. Other species that were not tested on-farm but grew well in the species-selection trials and were of interest to local farmers are discussed.As long as specific site and management conditions are carefully considered, data from species selection trials can be useful in informing tree planting projects with rural farmers; however, care should be taken to manage expectations. 相似文献
