Clinical, parasitological and biochemical parameters were evaluated in Corriedale and Crioula Lanada sheep after a single experimental infection with Haemonchus contortus. Ten 4-month-old worm-free lambs, of each breed, were infected with 200 L3 H. contortus per kg live weight and four uninfected animals of each breed were used as controls. Every week, the animals were weighed and blood and faecal samples were collected for measurement of packed cell volume (PCV), total serum protein (TSP) and albumin (ALB), and the number of eggs per gram of faeces (EPG), respectively. Twelve weeks after infection, the animals were slaughtered. The worm burden was determined and samples of the abomasal mucosa were processed for determination of the number of eosinophils, mast cells and globule leukocytes. No significant differences in PCV, TSP, ALB, parasite burden or the cell populations of the abomasal mucosa were observed between breeds, but Crioula lambs had a lower EPG count. The comparison of the infected groups with their respective controls revealed significant alterations in PCV, TSP and ALB in the Corriedale lambs and in PCV, TSP, ALB and the density of eosinophils and mast cells in the Crioula lambs.  相似文献

Purpose

Soils provide a variety of ecosystem services (ESs) that are crucial to food security, water security, energy security, climate change abatement, and biodiversity, especially in densely populated countries such as China. At present, China is facing great challenges from serious soil heavy metal (HM) contamination which has damaged soil ESs and soil security. In this paper, we evaluate the ESs that contaminated soils can potentially provide before and after remediation, and we explore the connections between these ESs and the achievement of soil security in China.

Materials and methods

After an introduction to the concepts of ESs and soil security and a review of the current status of soil HM contamination in China, the ESs that can potentially be provided by HM-contaminated soils are discussed. Finally, we discuss the current remediation status of HM-contaminated soils from the standpoint of optimizing the ability of these soils to provide ESs.

Results and discussion

The status of the provision of ESs by HM-contaminated soils of croplands, brownfields, and mining wastelands is described in detail. Contaminated cropland soils fail to provide provisioning (e.g., food production), cultural, and regulating services, while the regulating and supporting services of brownfield soils are greatly reduced. The ESs of mining wasteland soils have been severely damaged, resulting in a high potential for contamination of surrounding ecosystems. Recent soil remediation projects have demonstrated that the damaged ESs of HM-contaminated soils can be restored, which would enhance Chinese soil security. However, it has often been the case that only visible ESs (e.g., food production and vegetation cover) are addressed, while other less noticeable but important services (e.g., water quality and biodiversity) are neglected. Therefore, we propose a framework for the evaluation of ESs provided by HM-contaminated soils.

Conclusions

The ESs that could potentially be provided by HM-contaminated soils would help to achieve soil security in China, not only by improving food security, water security, and energy security but also by helping to protect soil biodiversity and abate global climate change. The ESs provided by HM-contaminated soils should be taken into account in soil policy and management systems as well as by the remediation industry.

Following evidence of reduced efficacy of ivermectin in a field population of Haemonchus contortus in Brazil, this strain of the parasite was submitted to a controlled anthelmintic test. Eighty worm-free lambs were randomly split into two groups of 40. Each lamb in the first group was infected with 5000 third stage larvae (L3) of the ivermectin-resistant strain; the remaining 40 lambs were each infected with 5000 L3 of a H. contortus strain of known susceptibility to the major groups of anthelmintic compounds used in sheep. On Day 28 post-infection, each group was subdivided according to egg counts and at random into four sub-groups of ten lambs, each of which was treated with albendazole (ABZ) at 3.8 mg kg-1, levamisole (LEV) at 7.5 mg kg-1 or ivermectin (IVM) at 0.2 mg kg-1, or was left as untreated control. At slaughter, 7 days later, all the anthelmintics reduced worm burdens in animals infected with the susceptible strain (ABZ 98.9%, LEV and IVM 100%). By contrast, in the lambs infected with the ivermectin-resistant strain, LEV was 99.8% effective, but ABZ reduced worm counts by only 14.7% and IVM by only 10.4%. Interestingly at necropsy on Day 7 post-treatment, the egg counts in the resistant strain treated with ABZ had been reduced by 92.5%, although worm counts were reduced by only 14.7%.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

An experiment was conducted to estimate the relative bioavailability of inorganic Se sources based on tissue Se deposition following supplementation at high dietary levels. Twenty-eight crossbred wethers averaging 50 kg initial weight were assigned randomly to seven treatments that were fed for 10 d. The basal diet contained .18 mg/kg Se (DM basis). Dietary Se was added at 0, 3, 6 or 9 mg/kg as reagent grade sodium selenite (Na2SeO3) and 6 mg/kg from either calcium selenite (CaSeO3), Na2SeO3 + fumed amorphous carrier or sodium selenate (Na2SeO4). There were four sheep per treatment group, housed in individual, raised pens with slatted floors. Daily feed intake was restricted to 1,200 g and tap water was available ad libitum. The basal diet was fed for a 10-d adjustment period, then sheep were fed experimental diets for 10 d. At the termination of the experiment, blood samples were taken; sheep were stunned and killed, and livers and kidneys were removed and frozen for Se analysis. There was a linear (P less than .001) uptake of Se in liver, kidney and serum. The CaSeO3 and Na2SeO4 sources resulted in greater (P less than .05) Se concentrations in liver and kidney than did Na2SeO3, but these differences were not significant when the analyzed dietary Se concentrations were used as a covariate in the statistical model. Based on linear and multiple linear regression slopes and average increases in serum, liver and kidney Se concentrations, estimated relative bioavailability values corrected for analyzed dietary concentration, were 100, 101, 90 and 133 for Na2SeO3, CaSeO3, Na2SeO3 + carrier and Na2SeO4, respectively.  相似文献

Two separate field trials involving naturally infected cattle were carried out on two farms known to have a history of Fasciola hepatica infection. On the first farm, 15 animals per group were allocated as follows: G1, triclabendazole (TCBZ) four times a year; G2, TCBZ twice a year (May and September); G3, untreated control. All groups grazed together and after 3.5 years the animals were slaughtered and their livers examined by federal meat inspectors who condemned 100% of livers in G3 and 8.3% in G2 owing to the presence of lesions of fasciolosis. In G1 no livers were condemned. Significant differences in weight gains were not detected and fluke counts remained at low levels in the treated groups. Also, in the control group, egg counts started to decrease when animals were 2 years old. On the second farm, groups of 20 animals were treated as follows: G1, TCBZ three times a year (May, September and December); G2, TCBZ twice a year (May and September); G3, nitroxynil twice a year (May and September); G4, rafoxanide twice a year (May and September); G5, untreated controls. All animals were weighed and faecal samples examined at approximately 28-day intervals. During the period of the study, larger weight gains were detected in the TCBZ treated groups than in the others. TCBZ treatment kept F. hepatica egg counts at a lower level for longer periods than the other drugs and significant differences in weight gains were only obtained between the group receiving TCBZ three times a year and the control group.  相似文献

Three different strains of Haemonchus contortus (susceptible to ivermectin, S-IVM; selected for resistance to ivermectin, R-IVM; a multiple resistant strain, i.e. resistant to benzimidazole and ivermectin, R-IVM/SA) were examined for isoenzyme variation by starch gel electrophoresis. Using stains for seven enzymes separated in five different buffer systems, no differences in the electrophoretic mobility could be detected between any of the strains. Results demonstrate a low level of enzyme variation in H. contortus and no differences in enzyme electrophoretic profile between IVM-sensitive and IVM-resistant parasites. Differences between the ivermectin-sensitive and both ivermectin-resistant strains were observed with the propionyl esterases and although some of the differences are probably associated with benzimidazole resistance, others are associated with resistance to ivermectin. The three strains of H. contortus are generally identical; however, differences between all strains of H. contortus and a strain of Dictyocaulus viviparus were detected.  相似文献

Thirty crossbred wethers (60 kg avg initial wt) were used to study the time-dose response to dietary Se as sodium selenite (Na2SeO3). Sheep were fed a basal diet (.20 mg/kg Se, M basis) for 10 d; three wethers were killed and tissues were collected for controls. The remaining 27 sheep were assigned randomly to diets supplemented with either 3, 6 or 9 mg/kg Se (as-fed basis) from reagent grade Na2SeO3 and fed for 10, 20 or 30 d. Feed offered was restricted to 1,200 g daily and tap water was available ad libitum. Sheep were stunned and killed by exsanguination and liver, kidney, muscle, heart and spleen were removed and frozen for Se analysis. No toxic effects were noted as expressed by feed intake or hemoglobin concentration. Added dietary Se increased Se linearly (P less than .01) in liver, kidney, and serum. Selenium in liver, kidney and serum also increased (P less than .01) as time advanced. Serum, liver and kidney were more sensitive to dietary Se than were muscle, heart and spleen. Ten days appeared to be an adequate length of time for further Se bioassay studies of this nature. Reagent grade Na2SeO3 was nontoxic when fed to sheep for 30 d at levels up to 90 times the Se requirement.  相似文献

Twenty-four yearling 11-month old Nellore steers initially weighing 180 kg were assigned to three treatments in a randomised complete block experiment lasting 392 days. Animals grazed continuously either Yaragua grass (Hyparrhenia rufa) or Yaragua/Stylo legume (Stylosanthes guianensis) and received the following treatments: A) Control, Yaragua plus common salt, B) Yaragua/Stylo plus common salt, and C) Yaragua/Stylo plus dicalcium phosphate/common salt (1:1). Mineral consumption in the three groups averaged 52 g per animal day-1. At 13 months daily liveweight gains/steer were 0.182, 0.409 and 0.483 kg for treatments of Yaragua, Yaragua/Stylo, and Yaragua/Stylo plus dicalcium phosphate/common salt mixture. Daily liveweight gains per ha were 0.488, 1.289, and 1.522 kg steer-1 for Yaragua, Yaragua/Stylo plus common salt and Yaragua/Stylo plus dicalcium phosphate/common salt. Steers receiving a legume (stylo) in the diet more than doubled weight gains (P less than 0.05) compared to controls with dicalcium phosphate further (P less than 0.05) increasing these gains.  相似文献

Agromining is an emerging technology that utilizes selected ‘metal crops' (= hyperaccumulator plants) to extract valuable target metals from unconventional resources for profit from mineralised soils. Growth characteristics, shoot metal concentrations, and agrominable locations are important considerations in economic agromining. Globally, the greatest potential for nickel (Ni) agromining exists in the tropics. However, the agronomic systems of tropical ‘metal crops' have not been previously tested. Furthermore, it is currently unknown whether nutrient dosing of prospective tropical agromining Ni ‘metal crops’ could possibly cause a shoot Ni‐dilution effect which may ultimately limit economically viable Ni yields. We undertook a pioneering study on Ni uptake and growth responses to nutrient dosing in two promising tropical ‘metal crops' (Phyllanthus rufuschaneyi and Rinorea cf. bengalensis). The experiment consisted of a large randomised block growth trial in large pots over 12 months in Sabah (Malaysia). At 3‐month intervals, the plants were exposed to soluble treatments that altered available concentrations of nitrogen (N), phosphorus (P), and potassium (K). We found strong positive growth responses to N and P additions in P. rufuschaneyi, whereas K additions had negative growth effects. In R. cf. bengalensis, all treatments had positive growth effects. The increases in biomass in response to nutrient dosing did not significantly reduce shoot Ni concentrations in both species, with the exception of N addition in P. rufuschaneyi. This study reveals that Ni uptake and growth responses to nutrient dosing are species‐dependent, primarily influenced by the ecophysiology of the species. Inorganic fertilization could possibly be an important component of the management of local ‘metal crops' to be used in viable commercial agromining in the tropics, but this needs to be tested in the field with different formulations of N, P, and K.  相似文献
