The results of vaccination trials carried out on pups with maternally derived antibodies (MDA) to canine parvovirus (CPV), using a modified‐live CPV‐2b variant vaccine (29‐97/40 strain), are reported. The vaccine was able to overcome the obstacle of MDA, and to elicit protective immunity in 100% of the pups whose antibody titres were 1:10–1:40, 83% of the pups with titres of 1:80, 57% of the pups with titres of 1:160, and even in 60% of the pups with antibody titres of 1:320. 相似文献

The application of artificial insemination techniques as a way to overcome the lack of mating of the closed thelycum Farfantepenaeus paulensis under large scale conditions is reported. The performance of artificially inseminated females (49.4±11.6 g) maintained in conventional versus unisex maturation systems was compared. Groups of unilaterally eyestalk-ablated females were artificially inseminated with one compound spermatophore soon after molting. The percentage of fertilized spawns increased from 26% before the use of artificial insemination to 57% afterwards. The reproductive performance of inseminated females held in conventional or unisex maturation systems showed no significant differences. Artificial insemination was considered a practical approach to overcome the lack of mating of F. paulensis under large-scale conditions with acceptable reproductive performance. The results also indicate the possibility of holding females separately from males, which could mean significant improvements in maturation systems through the increase in the stocking density of spawning females. 相似文献

The reproduction of wild-caught Farfantepenaeus paulensis in captivity has been successfully achieved since 1983. However, year-round supply of wild broodstock is costly and unpredictable. This study compares the reproductive performance of animals of similar size (females from 32–33 g and males from 19–21 g) but from different sources: estuary-caught juveniles reared indoors for 9 mo and wild-caught adults from shallow coastal waters. Groups of 20 males and 30 unilaterally eyestalk-ablated females were maintained for 60 d in four maturation tanks under controlled environmental conditions. A higher number of eggs per spawning event (78,414 ± 34,491) and total egg production (4,469,600) were observed for the wild females. Nevertheless, captive females produced a higher number of nauplii (1,788,259) due to their higher percentage of fertilized spawns. Therefore, within the size range of this study, the reproductive performance of estuary-caught F. paulensis juveniles over-wintered indoors was found to be equivalent to that of wild-caught females from shallow coastal waters. 相似文献

The relationship between maternally derived antibody (MDA) levels and protection to canine parvovirus (CPV) infection in pups is reported. Twelve pups with a wide range of haemagglutination inhibiting (HI) titres of MDA to CPV were divided into four groups, with each group balanced for antibody titres. The dogs were inoculated with a field CPV-2b strain and clinical signs, virus shedding and antibody response were assessed. The CPV was not detected in the faeces of dogs with HI titres of 320 at any time. In dogs with HI titres up to 160, active CPV replication after challenge was demonstrated by real-time polymerase chain reaction. The successful infection of dogs with HI titres of 80 and 160 was confirmed by seroconversion, evaluated at day 14 post-infection. These findings demonstrated that CPV infection could also occur in the presence of MDA HI titres (> or =80) usually considered fully protective. 相似文献

Protein-folding intermediates have been implicated in amyloid fibril formation involved in neurodegenerative disorders. However, the structural mechanisms by which intermediates initiate fibrillar aggregation have remained largely elusive. To gain insight, we used relaxation dispersion nuclear magnetic resonance spectroscopy to determine the structure of a low-populated, on-pathway folding intermediate of the A39V/N53P/V55L (A, Ala; V, Val; N, Asn; P, Pro; L, Leu) Fyn SH3 domain. The carboxyl terminus remains disordered in this intermediate, thereby exposing the aggregation-prone amino-terminal β strand. Accordingly, mutants lacking the carboxyl terminus and thus mimicking the intermediate fail to safeguard the folding route and spontaneously form fibrillar aggregates. The structure provides a detailed characterization of the non-native interactions stabilizing an aggregation-prone intermediate under native conditions and insight into how such an intermediate can derail folding and initiate fibrillation. 相似文献

The effects of total ammonia (TAN; NH4++ NH3) on the reproductive performance, survival, growth and moulting of wild Penaeus paulensis (Pérez-Farfante) broodstock were studied to determine optimal rearing conditions. Based on previously established ‘safe levels’ for P. paulensis adults (3.4 and 4.2 mg L?1 TAN), two 46-day trials were performed. In the first trial, six females and four males were stocked in 700-L tanks under three treatments (0.37, 2.53 and 6.86 mg L?1 TAN) with at least two replicates per treatment. In trial 2, ammonia levels of 0.68, 1.55 and 2.62 mg L?1 TAN were assigned to three 6000-L tanks, each stocked with 36 females and 24 males. Ammonia only influenced the survival of females and the growth of males exposed to 6.86 mg L?1 TAN (0.21 mg L?1 NH3). No further effects of ammonia on moulting and reproductive performance were detected. The present results demonstrate that up to 2.62 mg L?1 TAN, coupled with 0.07 mg L?1 NH3 and 1.50 mg L?1 NO2, will not impair reproductive performance of P. paulensis. It is suggested that water quality for the maturation of P. paulensis may be maintained through lower daily water exchange rates instead of the usual high levels (150-300%) employed on penaeid shrimp maturation systems. 相似文献

Biofloc rearing media provides a potential food source for shrimp reared in limited or zero water exchange systems. This culture
system is environmentally friendly as it is based on limited water use and minimal effluent is released into the surrounding
environment. In this study, we evaluated the survival, growth performance and salinity stress tolerance of pink shrimp Farfantepenaeus paulensis postlarvae reared from PL10 to PL25 in a biofloc technology limited water exchange system. PL (mean ± SD weight and length of 14 ± 10 mg and 8.10 ± 0.7 mm, respectively)
were reared in nine 40-L plastic tanks with a stocking density of 10PL/L. Three culture treatments were applied (1) culture
in the presence of bioflocs and commercial feed supply (FLOC + CF); (2) culture in the presence of biofloc without feed supply
(FLOC) and (3) culture in clear water with feed supply (control). Final biomass and survival were significantly higher in
FLOC + CF treatment than the control (P < 0.05), but did not differ from FLOC. PL reared in the FLOC + CF treatment achieved a significantly higher final weight,
weight gain and length in comparison with the other two treatments (P < 0.05). No significant difference (P > 0.05) between treatments was found for salinity tolerance over 24 and 48 h durations. The proximate analysis of floc shown
high levels of crude protein (30.4%), but low levels of crude lipids (0.5%). The continuous availability of bioflocs had a
significant effect on growth and survival of F. paulensis postlarvae cultured in BFT nursery systems. 相似文献
