One hundred two fecal specimens from psittacine birds submitted to Veterinary Laboratory Services of the California Department of Food and Agriculture at Petaluma were screened for Chlamydia psittaci by a direct immunofluorescence assay using a fluorescein-labeled monoclonal antibody conjugate specific for Chlamydia sp. Results were compared with those obtained by isolation of chlamydia in cultures of McCoy mouse cells. The relative specificity of the direct fluorescent antibody test on fecal smears was 98.9% and the relative sensitivity was 62.5%. The results of this study suggested that the direct fluorescent antibody test was highly specific, and it proved to be a useful same-day antemortem diagnostic test for birds with symptomatic chlamydial infection. The use of centrifugation in the cell culture assay was found to significantly enhance the level of chlamydial infection in cell culture. 相似文献

Superintensive shrimp culture in zero‐exchange, biofloc‐dominated production systems is more biosecure and sustainable than traditional shrimp farming practices. However, successful application of this technology depends upon optimizing dietary formulations, controlling Vibrio outbreaks, and managing accumulative changes in water quality and composition. A 49‐d study investigated the effect of two commercial feeds of differing protein content and an indoor limited‐exchange, biofloc‐dominated culture environment on Litopenaeus vannamei performance and tissue composition, water quality and ionic composition, and Vibrio dynamics. Juveniles (5.3 g) were stocked at 457/m3 into four 40 m3 shallow raceways containing biofloc‐dominated water and fed one of two commercial feeds with differing protein content, 35 or 40%. Shrimp performance, Vibrio populations, and changes in shrimp and culture water composition were monitored. There were no significant differences (P > 0.05) in shrimp performance (survival, weight, growth, specific growth rate, total biomass, yield, feed conversion ratio, and protein efficiency ratio) or proximate composition between feed types. The 40% protein feed resulted in higher culture water nitrate and phosphate concentrations, alkalinity consumption and bicarbonate use, and higher phytoplankton density. The presence of Vibrio, specifically Vibrio parahaemolyticus, reduced shrimp survival. This survival decrease corresponded with increased culture water Vibrio concentrations. Culture water K+ and Mg2+ increased significantly (P < 0.05), and Sr2+, Br?, and Cl? decreased significantly (P < 0.05) over time. While Cu2+ and Zn2+ did increase in shrimp tissue, no heavy metals accumulated to problematic levels in culture water or shrimp tissue. These results demonstrate the importance of monitoring Vibrio populations and ionic composition in limited‐exchange shrimp culture systems. 相似文献

When the effect of calcium on the oxidation of linoleic acid by potato tuber 5-lipoxygenase (LOX) was investigated, it was seen to promote the enzyme's activity at pH values higher than the optimum pH of 6.3, resulting in an enzyme activation at alkaline pH. Kinetic analysis of calcium activation at different pH values revealed that the cation abolished the inhibition by high substrate concentration, which occurs in the absence of Ca(2+), thus leading to activation at high substrate concentration. Studies were conducted to investigate the influence of Ca(2+) on the physicochemical nature of the substrate and its effect on the LOX activity expression. Peanut and its derivatives, especially peanut butter, are extensively consumed in many countries, mainly in the United States, which is also the major exporter of these products. trans-Resveratrol is present in peanuts, and recently this compound has been quantified in peanut butter. It is well-known that there are beneficial effects of trans-resveratrol and its glucoside, the piceid, in health. The absorption of trans-resveratrol has been proven in animals, and certain studies show that the absorption of some phenols is enhanced by conjugation with glucose, so that it could be possible that trans-piceid would be more absorbed than its aglycon (trans-resveratrol). In our work, we have identified the presence of trans-piceid in peanut butter with a new method to quantify trans-resveratrol and trans-piceid (3-beta-glucose of trans-resveratrol). This fact is very interesting because the glucosilated form could be more easily absorbed by the intestinal gut; in this way trans-piceid would exercise its beneficial function more efficiently than trans-resveratrol. To our knowledge, this is the first time that trans-piceid has been quantified in peanut butter. Wildfires often modify soil properties, including the N status and net N mineralization rates, but their impacts on gross N fluxes have been scarcely evaluated. We aimed to ascertain the immediate effects of a medium–high severity wildfire on soil N transformations. Net and gross N rates were analytically and numerically (FLUAZ) quantified in burned (BS) and unburned (US) topsoils from the temperate–humid region (NW Spain). Analytical and numerical solutions were significantly correlated for both gross N mineralization (m) (r 2?=?0.815; p?<?0.001) and gross nitrification (n) (r 2?=?0.950; p?<?0.001). In BS, all NH4 +-N fluxes (net m, gross m and gross NH4 +-N immobilization, ‘ia’) increased, while those of NO3 ?-N decreased (gross n and gross NO3 ?-N immobilization, ‘in’) or did not vary (net n). In US and BS, gross m (0.26–3.60 and 4.70–15.42 mg N kg?1 day?1, respectively) predominated over gross n (0.026–2.45 and 0.001–0.002 mg N kg?1 day?1, respectively), and the same was true for the net fluxes. Compared with the few available data on recently burned soils (m?=?8–55 mg N kg?1 day?1; n?=?0.50–1.83 mg N kg?1 day?1), our gross m and n rates were similar and very low, respectively; gross n showed that nitrifiers were active in US and also in BS, despite the 98 % reduction observed immediately after the fire.
