Increasing number of Salmonella paratyphi B isolates from slaughtered poultry sent in to the national Salmonella reference laboratory.]

In the last years the number of isolations of Salmonella enterica subspecies enterica serovar paratyphi B (S. paratyphi B) sent to the national salmonella reference laboratory of Germany has increased steadily. Most of the isolates originated from fowl or poultry products. The bacteriological, serological and biochemical properties of the isolates were investigated. Special emphasis was given to the utilization of d-tartrate which subgroups the serovar. All of them belonged to the d-tartrate positive variant, which is generally considered less virulent for humans and was formerly called S. java. The performance of various tests is compared and in addition the possibility of the spread within the production line is discussed.     

Estimation of the costs of mastitis, using National Animal Health Monitoring System and milk somatic cell count data

Data collected by the National Animal Health Monitoring System in Ohio for a 12-month period during 1986 and 1987 were used to determine the relative magnitude of costs associated with mastitis in the following categories: milk production loss, veterinary services, drugs, producer labor, and "other" factors. The cost of milk loss associated with mastitis that was reported by producers cooperating in the National Animal Health Monitoring System program was compared with estimates based on bulk tank somatic cell counts and individual cow milk somatic cell counts. Using producer-reported estimates, milk loss accounted for about one third of the total cost associated with mastitis. When estimates of milk loss were replaced by estimates based on bulk tank somatic cell counts, milk loss accounted for over 80% of the total cost of mastitis. Estimates of the cost of milk loss based on studies relating milk yield to somatic cell counts differed considerably. Consequently, it was unclear how to best estimate the relative magnitude of the milk loss component of mastitis costs.     

Stable carbon isotope ratios of rock varnish organic matter: a new paleoenvironmental indicator

Stable carbon isotope ratios of organic matter in rock varnishes of Holocene age from western North America and the Middle East show a strong association with the environment. This isotopic variability reflects the abundance of plants with different photosynthetic pathways in adjacent vegetation. Analyses of different layers of varnish on late Pleistocene desert landforms indicate that the carbon isotopic composition of varnish organic matter is a paleoenvironmental indicator.     

Binding Sites for Ca2+-Channel Effectors and Ryanodine in Periplaneta americana—Possible Targets for New Insecticides

The calcium channel and the ‘calcium release channel’ of muscle membrane of the cockroach Periplaneta americana have been characterized. Biological assays with calcium channel blockers and ryanodine on different insects and acari revealed pronounced insecticidal effects with ryanodine, but not with calcium channel blockers, at concentrations between 0·1 and 300 μg ml⁻¹. Skeletal muscle membranes derived either from the tubular network or from the sarcoplasmatic reticulum of P. americana were characterized with respect to the binding of the dihydropyridine (DHP) [³H]isradipine (PN 200-110), the phenyl-alkylamine [³H]verapamil and the alkaloid [³H]ryanodine. Preliminary binding studies with the benzothiazepine [³H]diltiazem suggest a low-affinity binding site with a IC₅₀ value of 3·3 μM . All binding sites tested were sensitive to treatment with proteinase K. Optimal conditions for binding of the radioligand ryanodine revealed the highest specific binding at pH 8 and at calcium chloride concentrations between 100 and 500 μM . EGTA at 10 μM abolished 95% of the ryanodine binding. Binding studies with calcium channel binding sites revealed a pronounced effect of low Ca²⁺ concentrations on specific isradipine binding, whereas verapamil and diltiazem binding were only reduced by the presence of 200 μM EGTA. With respect to high Ca²⁺ concentrations, specific binding of diltiazem, isradipine and verapamil was reduced by 73, 40 and 20%, respectively, at 5 mM Ca²⁺. Radioligand binding experiments showed high-affinity binding sites for ryanodine and isradipine. K_D values of 0·95 nM (B_max=550 fmol mg⁻¹ protein) and 0·75 nM (B_max=213 fmol mg⁻¹ protein) were determined respectively. A lower-affinity binding site was identified in binding studies with verapamil (K_D=7·4 nM and B_max=27 fmol mg⁻¹ protein). [³H]isradipine displacement studies with several dihydropyridines revealed the following ranking of affinity: nitrendipine>isradipine>Bay K8664≪nicardipine. Displacement of [³H]verapamil binding by effectors of the phenylalkylamine binding site showed that bepridil and S(-)verapamil had the highest affinities of the compounds tested followed by (±)verapamil, nor-methylverapamil and R(+)verapamil.     

Report on Salmonella isolates submitted to the German National Veterinary Salmonella Reference Laboratory in the year 1999

The National Salmonella Reference Laboratory (NRL-Salm) of the Federal Institute for Health Protection of Consumers and Veterinary Medicine receives putative Salmonella isolates originating from animals, food, feed and the environment for typing. This report summarises the results of the sero- and phagetyping studies. In livestock S. Typhimurium DT 104 with 24% and S. Enteritidis PT 4 with 9% of all isolations predominate like in the human isolates. These sero- and phagetypes are frequently isolated from food as well and consequently can reach the consumer via the food chain.
