Technetium-99m hexamethylpropyleneamine oxine (99mTc-HMPAO) and Indium-111 oxine (111In-oxine) labeled canine gramulocytes were evaluated in vitro over a six hour period. Labeling efficiency for 99mTC-HMPAO and 111In-oxine labeled granulocytes was 39.6%± 8.0% and 60.6%± 17.6% (mean ± SD) respectively. The mean in vitro elution of the radiolabel ranged from 8.7-14.0% for the 99mTc-HMPAO grannulocytes and from 6.1-9.0% for the 111In-oxine granulocytes. Mean cell viability, for the 99mTc-HMPAO, 111In-oxine and non-radiolabeled control granulocytes ranged from 97.8-99.4%, 96.4-98.5% and 98.2-99.0%, respectively. The phagocytic ability of the 99mTc-HMPAO, 111In-oxine and control granulocytes ranged from 47.5-54.1%, 38.9-56.2% and 46.6-57.8% respectively over the six hour study period. Although labeling efficiency using 111In-oxine was significantly (P=0.05) better than 99mTc-HMPAO, there was no significant difference in label retention of the two radiolabels. There was no significant difference in viability or phagocytic function during the six hour study period. Considering the potential cost advantage and the superior imaging qualities of Technetium-99m relative to Indium-111, 99mTc-HMPAO appears to be a good alternative to 111In-oxine as a granulocyte label.  相似文献

Equilibrium radionuclide ventriculograms were performed in 6 normal dogs during resting and isoproterenol challenged states. A new method of analysis of the equilibrium radionuclide ventriculograms using a simple digital subtraction technique was compared to traditional methods of analysis and echocardiography. There was no significant difference between ejection fraction values from the three methods tested. The equilibrium radionuclide ventriculograms ejection fraction was significantly increased following isoproterenol administration using both the new and traditional methods of analysis but the pre versus post-isoproterenol increase ejection fraction derived from echocardiographic measurements of ejection fractions was not significantly different. The new digital subtraction method consistently produced reproducible, high quality ventricular volume curves for the left and right ventricle. Left and right ventricular maximum and average emptying and filing rates were determined from the slopes of the volume curve and the results were similar to those previously published for man and dog. The emptying and filling rates of both ventricles were significantly increased following isoproterenol administration. The time to peak emptying significantly decreased following isoproterenol. Time to peak filing decreased following isoproterenol but the difference was not significant.  相似文献

Isolated equine granulocytes (WBC), radiolabeled with 99mTc-hexamethylpropyleneamine oxime (99mTc-HMPAO) or 111In-oxine, were evaluated in vitro for their labeling characteristics, viability, and phagocytic function over a 6-hour postlabeling period. Mean +/- SD labeling efficiency for 111In-oxine-WBC was 62.2 +/- 15.3%, which was significantly (P less than 0.001) higher than that for 99mTc-HMPAO-WBC (32.0 +/- 17.0%). In vitro elution of radiolabel from cells was significantly (P less than 0.02) greater for 99mTc-HMPAO-WBC at 0.5, 2, and 4 hours, but was not significantly different from elution of radiolabel for 111In-oxine-WBC at 6 hours. Viability, assessed by trypan blue dye exclusion, for 99mTc-HMPAO-WBC, 111In-oxine-WBC, and nonlabeled control WBC ranged from 97 to 100%, and was not significantly different among groups. Cell function was assessed by use of a phagocytosis assay and was reported as phagocytic index. The phagocytic index ranged from 0.86 to 0.96 for 99mTc-HMPAO-WBC, and from 0.76 to 0.97 for 111In-oxine-WBC. The phagocytic index was not significantly different at 0.5, 2, or 4 hours, but was significantly (P = 0.038) greater at 6 hours for 99mTc-HMPAO-WBC. Because of the superior imaging characteristics of 99mTc-HMPAO-WBC and equal or better labeling characteristics than those for 111In-oxine at 6 hours, 99mTc-HMPAO-WBC appear to be a good alternative to 111In-oxine-WBC.  相似文献

A serpentinite-hosted ecosystem: the Lost City hydrothermal field   总被引:1,自引:0,他引:1  
The serpentinite-hosted Lost City hydrothermal field is a remarkable submarine ecosystem in which geological, chemical, and biological processes are intimately interlinked. Reactions between seawater and upper mantle peridotite produce methane- and hydrogen-rich fluids, with temperatures ranging from <40 degrees to 90 degrees C at pH 9 to 11, and carbonate chimneys 30 to 60 meters tall. A low diversity of microorganisms related to methane-cycling Archaea thrive in the warm porous interiors of the edifices. Macrofaunal communities show a degree of species diversity at least as high as that of black smoker vent sites along the Mid-Atlantic Ridge, but they lack the high biomasses of chemosynthetic organisms that are typical of volcanically driven systems.  相似文献

The marine bivalved Brachiopoda are abundant throughout the geological record and have apatitic (CaPO4-rich) or calcitic (CaCO3-rich) shells. Vesicles covering the larval valves of living apatitic-shelled discinids contain tablets of silica. The tablets are cemented into close-packed mosaics by spherular apatite in glycosaminoglycans. They are usually lost as vesicles degrade but leave imprints on the underlying apatitic shell. Similar imprints ornament larval surfaces of some of the earliest Paleozoic apatitic-shelled brachiopods and may also be indicators of siliceous biomineralization.  相似文献

Osmolar control of prolactin secretion in man   总被引:1,自引:0,他引:1  
To study the effect of changing serum osmolality on serum prolactin concentration 11 volunteers were given oral and intravenous hypotonic and hypertonic fluids. Mean serum prolactin fell to 10.5 percent of baseline after oral water loading and to 15 percent of baseline after intravenous hypotonic saline infusion. Conversely, mean prolactin rose to 417 percent of baseline after intravenous hypertonic saline administration. The correlation coefficient of simultaneously determined serum prolactin and osmolality was highly significant (P .001). Isoosmolar changes in extracellular fluid volume did not consistently affect the concentration of prolactin in the serum. Thus, prolactin may be involved in the physiologic regulation of osmolar balance and the kidney may be an important farget organ for prolaction.  相似文献
