We studied the effects of the root endoparasitic nematode Heterodera trifolii on rhizodeposition and the root architecture of white clover (Trifolium repens). Rhizosphere solutions were collected from the root systems of plants growing with and without H. trifolii (200 juveniles per inoculated plant) in sand-based microlysimeters. The organic carbon (C) content of these solutions was analyzed, and they were applied to plant-free soils to investigate microbial responses. Although plant biomass was unaffected by nematodes, the architecture of the root systems was significantly altered, with a decrease in overall root length and an increase in the density of lateral branches from the primary root. The presence of nematodes reduced the concentration of organic compounds in the rhizosphere solutions but only on the final sampling date (75 days). Analysis of microbial signature phospholipid fatty acids revealed no change in the structure of the microbial communities in soils to which rhizosphere solutions were applied. However, these microorganisms did respond with changes in substrate utilization patterns (community-level physiological profiles). Microbes in soils that received rhizosphere solutions from the nematode-infected clover showed lower utilization of most substrates but higher utilization of oligosugars. These responses appear to be related to changes in roots and rhizodeposition associated with nematode infection of clover roots. The results of this study suggest that root herbivory can negatively impact carbon-limited soil microbial communities via changes in root architecture that moderate rhizodeposition.  相似文献

The brain keenly depends on glucose for energy, and mammalians have redundant systems to control glucose production. An increase in circulating glucose inhibits glucose production in the liver, but this negative feedback is impaired in type 2 diabetes. Here we report that a primary increase in hypothalamic glucose levels lowers blood glucose through inhibition of glucose production in rats. The effect of glucose requires its conversion to lactate followed by stimulation of pyruvate metabolism, which leads to activation of adenosine triphosphate (ATP)-sensitive potassium channels. Thus, interventions designed to enhance the hypothalamic sensing of glucose may improve glucose homeostasis in diabetes.  相似文献

The surface potential of biological membranes varies according to their lipid composition. We devised genetically encoded probes to assess surface potential in intact cells. These probes revealed marked, localized alterations in the charge of the inner surface of the plasma membrane of macrophages during the course of phagocytosis. Hydrolysis of phosphoinositides and displacement of phosphatidylserine accounted for the change in surface potential at the phagosomal cup. Signaling molecules such as K-Ras, Rac1, and c-Src that are targeted to the membrane by electrostatic interactions were rapidly released from membrane subdomains where the surface charge was altered by lipid remodeling during phagocytosis.  相似文献

Variable Rate Technology (VRT) has the potential to increase crop yields and improve water quality relative to Uniform Rate Technology (URT). The effects on profitability and water quality of adopting VRT for nitrogen (N) and lime were evaluated for corn production on four claypan soil fields in north central Missouri under average to better than average weather conditions. Variable N and lime rates were based on measured topsoil depth and soil pH, respectively. VRT rates were compared to two different uniform N applications (URT-Nl based on the topsoil depth within these claypan soil fields, and URT-N2 based on a typical N rate for corn production in this area). Expected corn yield was predicted based on topsoil depth, soil pH, N rate, and lime rate. Water quality benefits of VRT relative to URT were evaluated based on potential leachable N. Sensitivity analyses were performed using simulated topsoil data for topsoil depth and soil pH. Results showed that VRT was more profitable than URT in the four sample fields under URT-N1, and in two of the four fields under URT-N2. Greater variation in topsoil depth and soil pH resulted in higher profitability and greater water quality benefits with VRT. Results support adoption of VRT for N and lime application for other claypan soil fields with characteristics similar to those in the fields used in this study.  相似文献

BACKGROUND: Measurement of high concentrations of urine catecholamines and metanephrines is useful in diagnosing pheochromocytoma in humans. Stress increases catecholamine excretion in urine. HYPOTHESIS: Stress of a hospital visit increases urinary catecholamine and metanephrine excretion in dogs. ANIMALS: Fourteen clinically normal dogs, 2 dogs with pheochromocytoma. METHODS: Voided urine samples were collected by the owners 7 days before (t-7), during the hospital visit immediately after diagnostic procedures (t0), as well as 1 (t1) and 7 days (t7) after the hospital visit. Urine catecholamine and metanephrine concentrations were measured using high-pressure liquid chromatography and expressed as ratios to urine creatinine concentration. RESULTS: In client-owned dogs epinephrine and norepinephrine ratios at t0 were significantly higher compared with ratios at t7. Metanephrine and normetanephrine ratios at t-7, t0, and t1 did not differ significantly from each other; however, at t7 they were significantly lower compared to values at t-7. In staff-owned dogs no significant differences were detected among the different collecting time points for any variable. Metanephrine and normetanephrine ratios were significantly higher in client-owned dogs compared to staff-owned dogs at t-7, t0, and t1 but not at t7. CONCLUSIONS AND CLINICAL IMPORTANCE: Stress associated with a hospital visit and with the sampling procedure causes increases in urine catecholamine and metanephrine excretion. Urine collection for the diagnosis of pheochromocytoma probably should take place at home after adaptation to the sampling procedure.  相似文献

Since 2007, the Open Access journal PLOS ONE has published 194,622 expert-reviewed scientific papers, including 85 related to the human food sciences such as food safety, general food science, and nutrition. Seventy-five of their corresponding authors had previously published. Their most cited articles in other journals were identified and citation counts compared against those earned by their PLOS ONE papers. No PLOS ONE food sciences paper has yet been cited more than an author's best effort in the field in other journals. Nitrous oxide (N2O) emissions make up a significant part of agricultural greenhouse gas emissions. There is an urgent need to identify new approaches to the mitigation of these emissions with emerging technology. In this short review four approaches to precision managements of agricultural systems are described based on examples of work being undertaken in the UK and New Zealand. They offer the opportunity for N2O mitigation without any reduction in productivity. These approaches depend upon new sensor technology, modeling and spatial information with which to make management decisions and interventions that can both improve agricultural productivity and environmental protection.  相似文献

This article considers the experiences of a Samoan female geographer undertaking community‐based field research in her home country, and identifies some lessons for researchers working in similar circumstances. The paper suggests that ethical behaviour in research is ‘place‐specific’, and there is therefore a need to employ culturally sensitive and ‘location‐specific’ methodologies when undertaking community‐based research fieldwork. For the lead researcher, growing up in Samoa played a significant role in gaining an understanding of a set of robust and effective procedures to evaluate citizens' resilience to climatic and other environmental challenges. Fluency in everyday and more formal local languages, and awareness of local differences, each with multi‐layered connections to local communities, understanding cultural nuances and practising respect for va tapuia [sacred space or relationships] protocols, together facilitated the process of knowledge acquisition and helped to confirm the validity of the conclusions derived from the study.  相似文献

Palmar foot pain is a common cause of lameness. Magnetic resonance imaging (MRI) has the potential to detect damage in all tissues of the equine foot, but an understanding of the differences in magnetic resonance (MR) images between feet from horses with and without palmar foot pain is required. This study aimed to describe MR findings in feet from horses with no history of foot-related lameness, and to compare these with MR findings in horses with lameness improved by palmar digital local analgesia. Thirty-four limbs from horses euthanized with a clinical diagnosis of navicular syndrome (ameness >2 months duration, positive response to palmar digital nerve blocks and absence of other forelimb problems) (Group L), and 25 feet from age-matched horses with no history of foot pain (Group N) were examined. For each anatomic structure, MR signal intensity and homogeneity, size, definition of margins, and relationships with other structures were described. Alterations in MR signal intensity and homogeneity were graded as mild, moderate, or severe and compared between Groups L and N. Results revealed that there were significant differences in MR images between Groups N and L. Multiple moderate-severe MR signal changes were present in 91% of limbs from Group L and moderate (none were graded severe) in 27% of limbs from Group N. In most Group L limbs, more than three structures and frequently six to eight structures were abnormal. Concomitant abnormalities involved most frequently the deep digital flexor tendon, distal sesamoidean impar ligament, navicular bone, collateral sesamoidean ligament, and navicular bursa (with significant associations in severity grade between these structures), sometimes with involvement of the distal interphalangeal joint and/or its collateral ligaments. It was concluded that findings on MR images were different between horses with and without foot pain, and that pain localized to the foot was associated with MR changes in a variety of structures, indicating that damage to several structures may occur concurrently and that MR imaging was useful for evaluation of foot pain.  相似文献
