Four components of partial resistance toPhytophthora infestans were measured after inoculation in the greenhouse and in the field ofSolanum arnezii x hondelmannii, S. berthaultii, S. circaeifolium, S. leptophyes, S. microdontum, S. sparsipilum, S. sucrense andS. vernei, and four hybrid progenies ofS. microdontum withS. tuberosum. The four components were infection efficiency, lesion growth rate, generation time and sporulation capacity. The results were compared with resistance ratings derived from field experiments, and with observations made on the potato cultivars Bintje, Bildstar, Libertas and Pimpernel. Genetic variation for all components was found, while the relative importance of the components of partial resistance appeared to vary between the species. InS. microdontum, generation time, infection efficiency and lesion growth rate, and inS. tuberosum infection efficiency, lesion growth rate and sporulation capacity appeared positively associated, but in other species no such association was found. A strong hypersensitive reaction, the expression of which appeared to depend on environmental conditions, was found inS. microdontum. ForS. berthaultii, infection efficiency appeared to be the main resistance component.Abbreviations ADPC

We have amplified, cloned and sequenced (part of) the open reading frame of the S1 segment encoding the sigma C protein of avian reoviruses isolated from chickens with different disease conditions in Germany and The Netherlands during 1980 up to 2000. These avian reoviruses were analysed phylogenetically and compared with sequences of avian reoviruses in the Genbank database. The avian reoviruses could be grouped in 5 different genotyping clusters and this classification was identical when the sequences were compared of the 5' end, the 3' end or the whole open reading frame of the sigma C protein. Therefore sequencing of either part of the gene encoding the sigma C protein seems to be reliable for classification. We were unable to identify a correlation between sigma C sequences of the avian reoviruses and the disease condition they were isolated from. The sequences found in The Netherlands and in Germany are, like those in Taiwan, more dispersed than the known avian reovirus sigma C sequences in the USA and Australia. We did not establish temporal or geographic differences in the avian reoviruses studied. 相似文献

- Sawfish (Pristidae) are considered to be among the most threatened families of elasmobranch (sharks and rays). There is a need to gather information on the status of poorly known sawfish populations to assist in global recovery initiatives.
- This study used interviews with local fishers to investigate the presence of sawfish in southern Papua New Guinea (PNG) and their interactions with and uses and values for small-scale fishers.
- A range of sawfish size classes are still encountered throughout southern PNG, while juvenile largetooth sawfish Pristis pristis were additionally reported in the freshwater reaches of all rivers surveyed. Reports of large size classes in estuarine and marine environments provide an optimistic outlook that sawfish populations persist throughout southern PNG.
- Most fishers that catch sawfish retain them for various uses including consumption and for the sale of meat, fins and occasionally rostra. Negative population trends including decreases in catch frequency and/or size classes were reported by 66% of interviewees, with the largest declines being reported in the Kikori River. The increasing technical capacity of small-scale fishers, their preference for gillnetting and the emerging market for teleost swim bladder (a high-value fishery product) present a major ongoing threat to sawfish in southern PNG. Furthermore, the tendency of fishers to kill or remove rostra from entangled sawfish results in high fishing mortality regardless of any use by the fisher.
- This study indicates that considerable community engagement will be necessary to manifest any legislative actions or increased enforcement on international trade regulations for sawfish in PNG. This is due to traditional land and waterway ownership values throughout PNG and the local perception of sawfish as a traditional food resource rather than an animal of intrinsic biodiversity value as perceived by global conservationists. Future research should consider exploring culturally appropriate conservation initiatives that are likely to achieve engagement and participation from local fishers.

A Laboratory Assay for Phytophthora infestans Resistance in Various Solanum Species Reflects the Field Situation 总被引:4,自引:0,他引:4

Physiological and molecular research on resistance responses of Solanum tuberosum cultivars and partially resistant Solanum species to Phytophthora infestans requires a reliable resistance test that can be used in the laboratory. Laboratory tests performed on detached leaves and intact plants were compared with field tests for similarity of late blight reactions. Detached leaves from field-grown plants were as resistant as detached leaves from climate chamber-grown plants when challenged with P. infestans. However, detached leaves incubated in covered trays at high relative humidity were more susceptible than detached leaves kept in open trays or leaves on intact plants. The incubation conditions of detached leaves in covered trays rather than detachment itself appeared to affect the resistance expression. Detached leaves of some wild Solanum genotypes became partially infected, whereas intact plants were completely resistant when inoculated. Inoculation of leaves on intact plants, however, resulted in lower infection efficiencies. These limitations should be taken into account when choosing the appropriate inoculation method for specific purposes. For resistance screening, laboratory tests proved to be a good alternative for field tests. The ranking of resistance levels for twenty plant genotypes was similar under laboratory and field conditions. 相似文献

Feline coronaviruses (FCoV) comprise two biotypes: feline enteric coronaviruses (FECV) and feline infectious peritonitis viruses (FIPV). FECV is associated with asymptomatic persistent enteric infections, while FIPV causes feline infectious peritonitis (FIP), a usually fatal systemic disease in domestic cats and some wild Felidae. FIPV arises from FECV by mutation. FCoV also occur in two serotypes, I and II, of which the serotype I viruses are by far the most prevalent in the field. Yet, most of our knowledge about FCoV infections relates to serotype II viruses, particularly about the FIPV, mainly because type I viruses grow poorly in cell culture. Hence, the aim of the present work was the detailed study of the epidemiologically most relevant viruses, the avirulent serotype I viruses. Kittens were inoculated oronasally with different doses of two independent FECV field strains, UCD and RM. Persistent infection could be reproducibly established. The patterns of clinical symptoms, faecal virus shedding and seroconversion were monitored for up to 10 weeks revealing subtle but reproducible differences between the two viruses. Faecal virus, i.e. genomic RNA, was detected during persistent FECV infection only in the large intestine, downstream of the appendix, and could occasionally be observed also in the blood. The implications of our results, particularly our insights into the persistently infected state, are discussed. 相似文献

- One-third of all elasmobranch species currently known to occur in Papua New Guinea are taken as bycatch in the Gulf of Papua trawl fishery. An ecological risk assessment was conducted on the 16 species of sharks and 23 species of rays caught by the fishery.
- Eight species were classified to be at low risk, 28 species were at medium risk while three species – Rhynchobatus palpebratus (eyebrow wedgefish), Carcharhinus coatesi (Australian blackspot shark) and Maculabatis astra (blackspotted whipray), all endemic to New Guinea and northern Australia – faced the highest level of risk from the fishery at current fishing levels.
- There is potential for more vessels to enter the fishery in future, which is likely to elevate the risk for a greater proportion of elasmobranch species. Ongoing monitoring of bycatch levels, supported by biological and ecological studies on frequently caught species, is required to assess and manage risk in the future.
- The development and use of trawl excluder devices or bycatch reduction devices in this fishery have the potential to reduce bycatch levels and reduce risk levels.
