Biochars produced from different feedstocks (such as wood, pig manure) possess varying physical and chemical properties, which have influence on crack and evaporation rate of biochar-amended soil (BAS). Furthermore, influence of compaction state and drying-wetting cycles on evaporation rate and cracking of BAS has not been investigated comprehensively. The objective of this study was to investigate the effects of biochar types, compaction state of BAS, and drying-wetting cycles on crack propagation and retained water (or evaporation rate).
Material and methods
An animal and plant feedstock-based biochars were produced in-house from pig manure (PM) and wood (W), respectively. In addition, nano structured chalk and wheat biochar (CWB) were also produced. Soil amended with individual biochars was compacted in petri-glass discs at two densities. Disc specimens were subjected to multiple drying-wetting cycles, and evaporation rate of specimens and crack area were monitored throughout the experimental period (70 days). Images were captured after every 24 h and processed using image processing technique to obtain the crack intensity factor (CIF).
Results and discussion
The results show that plant-based W BAS showed the high water retention, i.e., low evaporation rate and low CIF. Furthermore, the crack potential of CW BAS was seen to be higher. In dense compacted soil, maximum CIF% can be reduced from 3.9 to 0.4% for W BAS, from 3.9 to 1.7% for PM BAS, and from 3.9 to 1.6% for CW BAS.
Conclusion
WB was able to resist cracking more efficiently than other types of biochar. Evaporation was found to be minimal for plant-based W BAS at 10% biochar percentage. Higher biochar content in soil was seen to increase the water retention of BAS significantly. Dense state of BAS at high biochar content (i.e., 10%) was effective in reducing evaporation rate and crack progression.
Cryopreservation of semen collected from dead fishes showed that it is possible to use it for fish production. We humanely killed individuals of the Indian major carp rohu, Labeo rohita and stored at different temperature regimes of 31 °C, 0 °C, − 10 °C and − 30 °C till 8 h. At every one hour interval semen from these fishes were collected by Pasteur pipette and evaluated for sperm yield/kg body weight, motility, pH, spermatocrit (%) and sperm count. The semen having suitable characteristics such as; 70% spermatocrit or above and motility index of 4 or above was cryopreserved by following a specific protocol. The cryopreserved semen of the dead fishes was stored for 7 days and then thawed in a water bath at 37 °C for 50 s. It was found that up to 8 h, spermatozoa of rohu were viable when stored at 0 °C or − 10 °C. Sperm collected after 8 h of fish death and maintained at 0 °C was the best stored condition that showed 30% larval survival. The spermatozoa collected 8 h after fish death was mostly normal as observed under scanning electron microscope and the total length of rohu spermatozoa was 25-30 μm. The hatchlings produced with this cryopreserved semen grew normally and juvenile fishes of rohu could be produced. This study suggests that germ cells such as spermatozoa of dead fishes can be cryopreserved and utilized for restoration of a species. The effects of including bovine lactoferrin (Lf) in the diet of the Asian catfish (Clarias batrachus) on specific and non-specific immunity as well as disease resistance were investigated. The catfish were fed four different diets for 2 weeks: a commercial diet as control and the same diet supplemented with 50, 100 and 200mg bovine Lf/kg feed. After 1 and 2 weeks, serum bacterial agglutination titre against Aeromonas hydrophila as a measure of specific immunity; natural serum haemolysin titre, lysozyme activity and oxidative radical production by neutrophils as a measure of non-specific immunity as well as disease resistance against A. hydrophila challenge to vaccinated and non-vaccinated animals were evaluated. The results showed that Lf supplements, particularly at 100mg level, significantly (P<0.05) enhanced serum lysozyme level, oxidative radical production and level of protection against A. hydrophila challenge in non-vaccinated animals irrespective of length of exposure. The specific immunity was not influenced by Lf feeding as evidenced from the bacterial agglutination titre and level of protection in vaccinated animals. As Lf feeding at 100mg/kg for 1 week is able to enhance the non-specific immunity and disease resistance of catfish efficiently, these results support the possible use of Lf as an immunostimulant for farmed catfish. 相似文献
Cassava is an important subsidiary food and industrial raw material in the tropics. Root rot disease, caused by Phytophthora palmivora, poses a serious threat to cassava cultivation in Tamil Nadu, India. Field experiments (2008–09) were conducted to study the effect of biocontrol agents (Trichoderma spp. and Pseudomonas fluorescens) and biofertilizers (Azospirillum, vesicular–arbuscular mycorrhizal fungi and phosphorus-solubilizing bacteria) on root rot, yield, harvest index and nutrient uptake of cassava at two NPK rates. The design of the experiment was a split plot with two NPK rates, recommended and 50% recommended rate, as the main plot treatments and five biocontrol agents and biofertilizers as subplot treatments. The results clearly indicated that use of a bioinoculants consortium significantly reduced root rot infection/disease incidence over uninoculated controls. Azospirillum significantly improved the yield of cassava at 50% of the recommended rate of NPK. NPK rates had no significant impact on harvest index of cassava and Trichoderma and vesicular–arbuscular mycorrhizal fungi resulted in a higher harvest index even at 50% of the recommended NPK rate. 1. Coir dust was evaluated as broiler litter in comparison with sawdust and rice husk using 135 commercial broilers. Forty-five broiler chicks were reared to 42 d on a 50 mm layer of each of these litters. 2. Birds reared on coir dust showed no difference in food consumption, body weight gain, food conversion efficiency production number and survivability in comparison to those reared on saw dust and rice husk. 3. The in vivo faecal egg count reduction test (FECRT) is the most commonly used test to detect anthelmintic resistance (AR) in gastrointestinal nematodes (GIN) of ruminants in pasture based systems. However, there are several variations on the method, some more appropriate than others in specific circumstances. While in some cases labour and time can be saved by just collecting post-drench faecal worm egg counts (FEC) of treatment groups with controls, or pre- and post-drench FEC of a treatment group with no controls, there are circumstances when pre- and post-drench FEC of an untreated control group as well as from the treatment groups are necessary. Computer simulation techniques were used to determine the most appropriate of several methods for calculating AR when there is continuing larval development during the testing period, as often occurs when anthelmintic treatments against genera of GIN with high biotic potential or high re-infection rates, such as Haemonchus contortus of sheep and Cooperia punctata of cattle, are less than 100% efficacious. Three field FECRT experimental designs were investigated: (I) post-drench FEC of treatment and controls groups, (II) pre- and post-drench FEC of a treatment group only and (III) pre- and post-drench FEC of treatment and control groups.To investigate the performance of methods of indicating AR for each of these designs, simulated animal FEC were generated from negative binominal distributions with subsequent sampling from the binomial distributions to account for drench effect, with varying parameters for worm burden, larval development and drench resistance. Calculations of percent reductions and confidence limits were based on those of the Standing Committee for Agriculture (SCA) guidelines. For the two field methods with pre-drench FEC, confidence limits were also determined from cumulative inverse Beta distributions of FEC, for eggs per gram (epg) and the number of eggs counted at detection levels of 50 and 25. Two rules for determining AR: (1) %reduction (%R) < 95% and lower confidence limit <90%; and (2) upper confidence limit <95%, were also assessed. For each combination of worm burden, larval development and drench resistance parameters, 1000 simulations were run to determine the number of times the theoretical percent reduction fell within the estimated confidence limits and the number of times resistance would have been declared.When continuing larval development occurs during the testing period of the FECRT, the simulations showed AR should be calculated from pre- and post-drench worm egg counts of an untreated control group as well as from the treatment group. If the widely used resistance rule 1 is used to assess resistance, rule 2 should also be applied, especially when %R is in the range 90 to 95% and resistance is suspected. 相似文献
A seven-year-old standardbred gelding developed marked signs of colic associated with an acute small intestinal obstruction. Surgical exploration revealed three intramural, circumferential constricting lesions in the small intestine, the two most severe of which were in the jejunum and were resected. One approach to decrease the environmental impact of crop production and reduce costs is to optimize agronomic practices and genotypes so that nutrients are used more efficiently. In this study the effects of agronomic practices (rotations, crop protection, fertilization) on yields, nitrogen use efficiency (NUE) and associated parameters were studied in an experiment using two winter wheat genotypes (Cordiale and Scaro) in one season and two potato genotypes (Sarpo Mira and Sante) in two seasons. The wheat showed no varietal differences in yield and NUE; instead the fertilization regime was the main factor affecting yield and NUE with higher values observed when conventional fertilization was used. The exception was for wheat grown after three years grass/clover ley when there was no added yield benefit from conventional fertilization of the organically bred variety (Scaro). This demonstrates the potential for N fixing crops to provide sufficient N to high yielding cereals if grown for long enough prior to planting. The greatest gains in NUE were achieved by combining an N efficient genotype with conventional crop management in an organic rotation. Fertilization and genotypic variation were the main factors affecting potato tuber yield and NUE, with the late maturing Sarpo Mira displaying elevated yields and NUE compared with the early maturing Sante. The use of organic fertility sources resulted in lower NUE, but N release from organic sources may increase NUE of future crops. This highlights the need for long-term nutrient balance and modelling studies to assess NUE at the crop rotation scale. 相似文献
