Genotype evaluation,muscle histopathology and ultrastructure in a Duchenne muscular dystrophy animal model

AIM: To evaluate the genotype , muscle histopathology and ultrastructure in dko mice. METHODS: Dystrophin/Utrophin-deficient double knockouts (dko) mice were obtained from university of Oxford, UK. Genotype of filial generation of heterozygote was evaluated by PCR-SSP. HE staining and fluorescent immunohistochemistry by SABC-Cy3 were used to detect striated muscle of dko mouse, and the muscle ultrastructure was observed by transmission electron microscope(TEM). RESULTS: In 112 filial generation mice, there were 28 mdx (25.0%), 26 dko (23.2%) and 58 heterozygote (51.8%), which coincided with the law of Mendelian inheritance. HE staining showed that the myocytes were not very uniform, there were phenomenon of round outline, centrally nucleated fibers, widening interspace, inflammatory cell infiltration and connective tissue proliferation in dko mice. There were no any immunofluorescent expression of dystrophin and utrophin in sarcolemma in dko mice. TEM showed sarcolemma breakage, separation and edema, and loose myofibril texture, inflammatory cell infiltration and connective tissue proliferation in dko mice. CONCLUSION: PCR-SSP is a very quick and accurate way for genotype evaluation of filial generation. The pathophysiology of dko mouse was very similar to Duchenne muscular dystrophy (DMD), and dko mouse is an ideal animal model for study of DMD clinical therapy.     

影响体细胞核移植效率的因素

哺乳动物体细胞核移植 (克隆 )是近年来迅速发展起来的一项新技术 ,对濒危动物保护、优良种畜快速扩群以及核质关系的研究、药物试验等诸多领域都具有重大意义。但体细胞核移植是一项程序繁杂的工作 ,每一步操作所采用的试剂、材料和方案等都会影响到核移植效率 ,因而其影响因素十分众多。文章根据目前已有资料 ,论述了卵母细胞来源、卵母细胞与供体细胞的胞质容量、卵母细胞的去核方案、体细胞供体动物的年龄、供体细胞的组织来源与传代次数及供体细胞的冷藏、冷冻和所处的细胞周期阶段对核移植效率的影响。另外 ,对重组胚的融合 -激活方案、核移植方案及重构胚的培养方案对核移植效率的影响也作了简要论述。     

湖南马尾松遗传改良现状与策略

在叙述湖南马尾松20多年来于马尾松群体遗传改良、马尾松优良家系定向选择、马尾松基因资源收集与种子园营建等方面所取得成绩的基础上，探讨了目前马尾松遗传改良所存在的问题，提出了今后一段时期湖南马尾松遗传改良策略。     

Portable ECG Monitor Front- ended Design with C8051 F020

The electrocardiogram (ECG) signal is a recording of the electrical activity on the body surface generated by the heart frequency of which is weak and low. Many researchers devote themselves to get more perfect ECG in many ways. This article introduces the characterize of an ECG signal , then discusses the requirements for the sampling circuit of an ECG. Since there are some shortcoming in common design means, the authors give an effective circuit. The system is composed of a low-cost , low-power and high-performance MCU C8051F020 , amplifier AD620 , right-leg drive circuit ,communication circuit and power management circuit. Then software design is given which is used to sampling and filtering ECG signal. Levkov filter arithmetic and smooth filter arithmetics are used in this design. In the end , the authors advance a distributing and networking ECG monitor system.     

Carrying Capacity of Stretching-bending Rectifier

In accordance with the deficient elongation of the Stretching-bending Rectifier, as well as the severe wear and slipping of V-belt , the driving system velocity ratio of rectifier and the calculation formula of the theoretical elongation are deduced, which are based on the analysis of its structure and reeling principle. The carrying capacity of the driving system is also analyzed and it reaches the main reason for the elongation of the stretching-bending rectifier that cannot reach request, which is caused by the step less torsion of fixed output of the continuously variable transmission. The improvement measure which can be achieved by increasing the stiffness of the spring to increase the output torsion of the continuously variable transmission is proposed. Thus it makes the actual coefficient of elongation,which can satisfiy the producing request.     

田园管理机悬架部分的建模及仿真

主要介绍了虚拟制造技术在田园管理机悬架上的应用,并对田园管理机械的应用及特点作了介绍。针对目前的田园管理机的悬架存在的问题进行了研究,利用大型的三维建模软件Pro/E建立了悬架的三维模型,采用Pro/E与Adams之间的接口程序mech/pro将模型从Pro/E导入Adams,再进行动力学分析,模拟悬架在工作时各个部分的受力及运动状态,为进一步优化分析提供了依据。     

3个草莓品种在辽阳地区温室栽培比较试验

为优化辽阳地区温室栽培草莓品种,笔者于2018年8月在辽阳县亚新农业合作社进行'香野'红颜'甜查理'温室栽培试验.结果表明,新品种'香野'果实甘甜爽口、香味浓郁、口感好、可溶性固形物含量高、抗病性强,是一个早熟、高产、优质的草莓新品种,可作为'甜查理'的替代品种在辽宁辽阳地区种植.     

农村妇女能力开发在发展贵州草地畜牧业标准化中的作用

着眼于农业从传统方式向现代农业演变的历史性进程,采用实证分析方法,具体分析了贵州省农村妇女的普遍性别特点及其在草地畜牧业标准化中的重要作用,指出进一步加强对贵州农村妇女能力这一人力资源的开发,不但对发展草地畜牧业标准化本身,而且对于发挥农村妇女的巨大潜力,促进贫困山区的开发式脱贫,推动贵州省现代农业发展进程,都具有十分重要的意义。最后就如何加强农村妇女能力开发,以及要注重创新对这一特殊群体的培训方式等方面提出了建议。     

蚯蚓密度与土壤化学肥力主要参数之间的定量关系

为研究蚯蚓密度与土壤化学肥力主要参数之间的定量关系,利用数据分析蚯蚓密度与pH值、电导率（EC）、有机质、全氮、碱解氮、全磷、有效磷、全钾、有效钾、有效钙、有效镁及阳离子交换量（CEC）之间的定量关系。结果显示,蚯蚓密度与pH值呈极显著的负相关关系（P<0.001）,与有机质、全氮和全磷含量呈极显著的正相关关系（P<0.002）,与其他指标的相关性比较弱。研究表明,根据这种关系可以将蚯蚓密度与土壤化学肥力主要指标的定量关系分为5个等级,可为准确地评价蚯蚓对土壤化学肥力的影响提供参考。     

污泥热碱液与氮肥配施对番茄产量、品质及氮转化的影响

随着生活污泥产量逐年增高,其无害化利用得到广泛关注。通过碱性热水解可以有效提取污泥中蛋白质、 多肽、氨基酸等营养物质得到污泥碱性热解液(简称热碱液),但还需进一步探索热碱液在农业中的应用。以番茄 为供试作物,设热碱液氮量分别为 0%(M0)、10%(M1)、20%(M2)、30%(M3)、40%(M4)、60%(M6)及 不施氮(N0)7 个处理,研究不同热碱液与尿素的配施比例对番茄产量、品质及氮转化的影响。结果表明,M3 处 理的产量和单果重均达到各处理最高,且分别比 M0 增加了 17.88% 和 15.19%。番茄的还原糖、可溶性糖、有机酸、 维生素 C、可溶性固形物含量及糖酸比在施入 20% ～ 30% 热碱液后均得到显著提升,逼近理想解排序法(TOPSIS 法)分析结果显示热碱液替代 20% ～ 30% 尿素的产量、品质综合水平最优。从养分吸收角度来说,20% ～ 30% 热 碱液的施入最有利于番茄对氮、磷、钾的吸收。M0 处理番茄叶中的硝酸还原酶活性最高[19.67 μmol/(g·h)], 其次为 M2 处理[13.61 μmol/(g·h)]。M2 处理番茄叶中的谷氨酰胺合成酶活性最高[14.40 μmol/(g·h)],其次 为 M3 处理[14.38 μmol/(g·h)],二者之间无显著性差异。污泥热碱液的合理施用有利于提高作物的氮转化酶活 性,提升番茄的产量及品质,降低果实的硝酸盐含量。
