Transplantation of fetal hematopoietic stem cells in utero: the creation of hematopoietic chimeras

Transplantation of normal, immature, fetal hematopoietic cells into a preimmune fetal recipient with a congenital hemoglobinopathy may allow partial reconstitution of normal hemoglobin production without the complications associated with postnatal bone marrow transplantation (immunosuppression and the occurrence of graft versus host disease). In order to test this hypothesis the naturally occurring polymorphism at the beta-hemoglobin locus of the sheep was used as a marker for engraftment and hematopoietic chimerism. Intraperitoneal injection of allogeneic fetal stem cells into normal fetal lambs resulted in hematopoietic chimerism in three of four surviving recipients. This chimerism has been sustained for 6 months after birth and 9 months after engraftment, without evidence of graft versus host disease, and without the use of immunosuppressive therapy.     

Tumor-Associated Erythrocytosis in a Dog With Nasal Fibrosarcoma

Erythrocytosis (hematocrit, 79%) was diagnosed in an 8-year-old, neutered female, mixed-breed dog with an intranasal fibrosarcoma. Both serum and tumor erythropoietin (Ep) activities were elevated, as determined by the polycythemic exhypoxic mouse model, and the Ep activity was neutralized in that model by rabbit anti-Ep antibodies. Tumor resection normalized the hematocrit.     

Towards Well-aligned electrospun PAN/MWCNTs composite nanofibers: Design,fabrication, and development

A proper collector is designed and examined in electrospinning process to produce electrospun nanofibers with favored mechanical propertied. The quality of product was controlled by changing and optimizing the process variables, namely electrospinning time, gap distance, and collector rotating speed in a manner that well-aligned yarns were fabricated from polyacrylonitrile (PAN) dilute solutions. It was found that the tensile characteristics of fabricated yarns are greatly dependent on the process variables. Incorporation of multi-walled carbon nanotubes (MWCNTs) into the polymer solution revealed improvement to the yarn strength because of enhancement in alignment of the filaments. The state of fiber alignment and dispersion of MWCNTs were detected by means of scanning electron microscopy. It was illustrated that combination of nanofibers and microfibers gives PAN/MWCNTs composite nanofibers with high surface area and high porosity to satisfy sophisticated users.     

Paroxetine Attenuates the Development and Existing Pain in a Rat Model of Neurophatic Pain

Background: P2X4 receptor (P2X4R), a purinoceptor expressed in activated spinal microglia, plays a key role in the pathogenesis of neuropathic pain. Spinal nerve injury induces up-regulation of P2X4R on activated microglia in the spinal cord, and blockade of this receptor can reduce neuropathic pain. The present study was undertaken to determine whether paroxetine, an inhibitor of P2X4R, could attenuate allodynia and hyperalgesia in chronic constriction injury (CCI) model of neuropathic pain when used preemptively or after the sciatic nerve injury. Methods: Male Wistar rats (150-200 g, n = 6) were divided into 3 different groups: 1- CCI vehicle-treated group, 2- Sham group, and 3- CCI paroxetine-treated group. Paroxetine (10 mg/kg, i.p.) was administered 1 h before surgery and continued daily until day 14. In other part of the study, paroxetine (10 mg/kg, i.p.) was administered at day 7 post injury and continued daily until day 14. von Frey filaments for mechanical allodynia and analgesia meter for thermal hyperalgesia were used to assay pain behavior. Results: In a preventive paradigm, paroxetine significantly attenuated both mechanical allodynia and thermal hyperalgesia (P<0.001). A significant decrease in pain behavior was seen with paroxetine on existing allodynia (P<0.001) and hyperalgesia (P<0.01) when initiated at day 7 post injury. Conclusion: It seems that paroxetine can attenuate pain behavior when administered before and also after sciatic nerve injury in CCI model of neuropathic pain. Identification of molecular markers linked to adult plant leaf rust resistance gene <Emphasis Type="Italic">Lr48</Emphasis> in wheat and detection of <Emphasis Type="Italic">Lr48</Emphasis> in the Thatcher near-isogenic line with gene <Emphasis Type="Italic">Lr25</Emphasis>

The recessive adult plant resistance (APR) gene Lr48 in wheat was tagged with flanking random amplified polymorphic DNA (RAPD) markers. Markers S336₇₇₅ in coupling and S3₄₅₀ in repulsion with Lr48 were identified in wheat line CSP44. Tests of these markers on available Thatcher near-isogenic lines (NILs) detected the likely presence of Lr48 in TcLr25. A test of allelism of APR involving the cross TcLr25 × CSP44 indicated that Lr48 was present in both lines. A separate experiment on inheritance of resistance in an F₂ population of TcLr25 × Agra Local confirmed the presence of a dominant seedling resistance gene (Lr25) and a recessive APR gene (Lr48) in TcLr25. This study demonstrated the value of molecular markers in identifying the presence of masked genes in genetic stocks where direct phenotyping failed to detect their presence.     

Renal erythropoietic factor: role of ions and vasoactive agents in erythropoietin formation

Evidence is provided for the existence of a renal erythropoietic factor, devoid of vasopressor activity, which upon interaction in vitro with normal serum yields erythropoietin. When undialyzed serum is used, erythropoietin inactivation develops in the incubation mixtures, and this inactivation appears to be dependent on an enzymatic component in preparations of the factor and on ions in serum.     

Changes in Biogenic Amines and Bacteria of Tiger-Toothed Croaker (Otolithes ruber) during Ice Storage

Changes in the content of biogenic amines (BA; e.g., putrescine, tyramine, and histamine) and the mesophilic and psychrotrophic bacterial counts of tiger-tooth croaker (Otolithes ruber) were monitored during ice storage for 18 days. BA and bacterial loads gradually increased (p < 0.05) with storage time. On the first day of storage, no amines were detected, and no histamine was detected up to 6 days. The best linear regressions between BA and bacterial counts were found between putrescine and psychrotrophic bacteria. Furthermore, it was found that psychrotrophs were the dominant microorganisms during storage and showed significant (p < 0.05) differences with storage time. The initial concentration of putrescine was 1.27 μg g^?1 and reached 18.8 μg g^?1 when the psychrotrophic bacterial load was over 10⁸ CFU g^?1. The results suggest that the level of putrescine is a suitable indicator of freshness for O. ruber, and the shelf life of O. ruber stored in ice as determined by BA content and microbiological data was 10 days.     

Enzyme inhibition and protein-binding action of the procyanidin-rich french maritime pine bark extract, pycnogenol: effect on xanthine oxidase

Pycnogenol, an extract from French maritime pine bark (PBE), is a complex mixture of bioflavonoids with reported protective effects against disease. PBE is an effective scavenger of reactive oxygen species, and its main constituents are procyanidins of various chain lengths. To find out the biochemical basis of action of PBE on enzyme activity, involvement of its redox activity and direct binding to the enzyme in its subsequent action on enzyme activity have been investigated. PBE dose-dependently inhibited the activities of xanthine oxidase, xanthine dehydrogenase, horseradish peroxidase, and lipoxygenase, but it did not affect the activities of glucose oxidase, ascorbate oxidase, or elastase. To characterize the mechanism of PBE action, studies were focused on xanthine oxidase and glucose oxidase. Under non-denaturing conditions, PBE changed the electrophoretic mobility of xanthine oxidase but not of glucose oxidase. Gel filtration chromatography confirmed higher molecular weight complexes of xanthine oxidase and xanthine dehydrogenase in the presence of PBE. It was found that hydrophobic bonding might be the dominant mode of interaction between PBE and xanthine oxidase. The importance of the binding in the effect of PBE on enzyme activity was supported by the observation that PBE binds to and inhibits catalase, but not superoxide dismutase. However, no correlation was found between superoxide/hydroxyl radical scavenging activity and the inhibitory effect on xanthine oxidase activity of PBE, various purified flavonoids, or other complex mixtures of bioflavonoids. The results indicate that PBE selectively inhibits xanthine oxidase through binding to the enzyme rather than by the redox activity.
