ABSTRACT:   In the present study, the role of carbonic anhydrase (CA) and Na/K-ATPase in the gill and epidermal tissues in the giant freshwater prawn Macrobrachium rosenbergii was examined as a function of molting stage. CA activity levels in the front and back gills were low at the intermolt stage C0, but increased significantly at premolt stage D3, and then decreased after molting. In the epidermal tissue, activity levels decreased gradually towards premolt to a minimum level at stage D3, but became elevated at postmolt stages A and B. Na/K-ATPase levels in the front and back gills did not change significantly during the molt cycle. CA in the gill is possibly involved in supplying counter-ions for ion uptake, while CA in the epidermal tissue may play a role in mineralizing the exoskeleton after ecdysis. Na/K-ATPase in the gills may function in ion uptake from the ambient medium; however, since its activity was not influenced by the molt cycle, it probably does not have a major role in osmoregulation in the freshwater environment.  相似文献

The effect of seed size on germination and seedling growth was assessed using cultivars of greengram and blackgram. Significant positive correlation was noticed between seed size and germination, seedling height, root length, primary leaf size and seedling weight. The possible advantages of choosing cultivars with heavier seeds for attaining better crop stand especially under rainfed situation are discussed.  相似文献

Summary Response of anthers in in vitro culture was examined in the indica-japonica hybrids of rice (Oryza sativa L.). Significant genotypic differences were observed for callus induction and regeneration among the different interracial hybrids of indica-japonica races. Induction frequency of haploids ranged from 57.7 to 72.9 per cent and doubled haploid androgenic lines ranged from 27.1 to 42.3 per cent in the anther culture of the different hybrids. The indica-japonica hybrids recorded partial pollen grain and spikelet fertility in F1 (29.9 to 41.5% and 19.4 to 48.7% respectively) as well as in F2 (42.7 to 50.6% and 37.1 to 54.4% respectively). In contrast, the androgenic doubled haploid lines recorded significant increase and the pollen grain and spikelet fertility was 76.3 and 78.6 per cent respecitively. Protoplast fusion can be used to produce somatic hybrids of species that cannot be obtained by sexual hybridization. The possibility to introgress genes from Solanum species into the cultivated tomato species Lycopersicon esculentum, and to obtain novel cytoplasm-nucleus combinations (cybrids) was considered as an important strategy to extend the genetic variation available for tomato breeding. Somatic hybrids between L. esculentum and other Lycopersicon species, as well as between L. esculentum and Solanum or Nicotiana species, have been produced. Specific mutants, genotypes with antibiotic resistances, and metabolic inhibition by iodoacetate or iodoacetamide and irradiation were used for the selection of hybrids. In addition, the improvement of protoplast culture techniques and the use of the favourable tissue culture traits derived from species such as L. peruvianum, which have been introduced into tomato by classical breeding, allowed the efficient recovery of somatic hybrids. However, the occurrence of somatic incongruity in fusion combinations of L. esculentum and Solanum and even more in L. esculentum and Nicotiana, did not allow the production of true cybrids and/or fertile hybrids, indicating the importance of both cytoplasm-nucleus and nucleus-nucleus interactions in somatic incongruity. Another problem with fusions between distantly related species is the strongly reduced fertility of the hybrids and the very limited homoeologous recombination between chromosomes of the parental species. Partial genome transfer from donor to recipient through microprotoplast (+) protoplast fusion, and the production of monosomic or disomic chromosome addition lines, light overcome some of these problems. In symmetric somatic hybrids between L. esculentum and S. tuberosum the occurrence of limited somatic and meiotic recombination was demonstrated. Fertile progeny plants could be obtained, though at a low frequency, when embryo rescue was performed on a large scale after backcrossing hexaploid somatic tomato (+) potato hybrids with a tetraploid potato genotype. The potential value of genomic in situ hybridization (GISH) and RFLPs for the analysis of the genome/chromosome composition of the hybrids has been demonstrated for intergeneric somatic hybrids between Lycopersicon and Solanum.Abbreviations cpDNA chloroplast DNA - mtDNA mitochondrial DNA  相似文献

A field study was conducted to determine if differences in grain filling rate and duration existed in two early cultivars of rice. There is no cultivar difference in duration of grain filling and grain matured in about 30 days from anthesis in both the cultivars. The effective grain filling period in rice is only 10 days between 3rd and 13th day from anthesis. There is significant difference in grain filling rate and is positively related to grain weight. Grain filling rate is more important than duration of filling for achieving higher grain size and thus yield in rice.  相似文献

The batch bioreduction of Cr(VI) by the cells of newly isolated chromium-resistant Acinetobacter sp. bacteria, immobilized on glass beads and Ca-alginate beads, was investigated. The rate of reduction and percentage reduction of Cr(VI) decrease with the increase in initial Cr(VI) concentration, indicating the inhibitory effect of Cr(VI). Efficiency of bioreduction can be improved by increasing the bioparticle loading or the initial biomass loading. Glass bioparticles have shown better performance as compared to Ca-alginate bioparticles in terms of batch Cr(VI) reduction achieved and the rate of reduction. Glass beads may be considered as better cell carrier particles for immobilization as compared to Ca-alginate beads. Around 90% reduction of 80 ppm Cr(VI) could be achieved after 24 h with initial biomass loading of 14.6 mg on glass beads. Artificial neural network-based models are developed for prediction of batch Cr(VI) bioreduction using the cells immobilized on glass and Ca-alginate beads.  相似文献

Utilization of industrial solid wastes for the treatment of wastewater from another industry could help environmental pollution abatement, in solving both solid waste disposal as well as liquid waste problems. Red mud (RM) is a waste product in the production of alumina and it poses serious pollution hazard. The present paper focuses on the possibility of utilization of RM as an adsorbent for removal of Remazol Brilliant Blue dye (RBB), a reactive dye from dye-contaminated water. Adsorption of RBB, from dye-contaminated water was studied by adsorption on powdered sulfuric acid-treated RM. The effect of initial dye concentration, contact time, initial pH, and adsorbent dosage were studied. Langmuir isotherm model has been found to represent the equilibrium data for RBB?CRM adsorption system better than Freundlich model. The adsorption capacity of RM was found to be 27.8?mg dye/g of adsorbent at 40?°C. Thermodynamic analysis showed that adsorption of RBB on acid-treated RM is an endothermic reaction with ?H 0 of 28.38?kJ/mol. The adsorption kinetics is represented by second-order kinetic model and the kinetic constant was estimated to be 0.0105?±?0.005?g/mg?min. Validity of intra-particle diffusion kinetic model suggested that among the mass transfer processes during the dye adsorption process, pore diffusion is the controlling step and not the film diffusion. The process can serve dual purposes of utilization of an industrial solid waste and the treatment of liquid waste.  相似文献

The union of potato monoploid genotypes (2n=1x =12) through protoplast fusion may result in vigorous somatic hybrids due to a reduction of the “genetic load” normally present in this highly heterozygous tetraploid (2n=4x=48) crop. More than 100 androgenic monoploids derived from diploid (2n=2x=24)Solanum phureja Juz. & Buk. andS. chacoense Bitt. xS. phureja clones were evaluated in field trials during 1996 and 1997 to identify the most promising genotypes for protoplast fusion experiments. Compared to the total population, the 1996 selected genotypes had higher means for tuber number (30.1 vs 11.2 tubers/plant), average tuber weight (3.0 vs 1.8 g/tuber) and total yield (66.1 vs 20.4 g/plant). Similarly, the 1997 selected genotypes had higher means for tuber number (42.8 vs 25.4 tubers/plant), average tuber weight (3.6 vs 2.5 g/tuber) and total yield (114.0 vs 63.4 g/plant) compared to the total population. The 31 selected monoploid genotypes from 1996-97 varied in their response to protoplast isolation and culture from no growth (9), cell enlargement (5), limited cellular divisions (8), callus formation (5) to plant regeneration from callus (4). Chemical fusion and electrofusion produced three groups of intermonoploid somatic hybrids. Polymorphic simple sequence repeat (SSR) loci enabled distinction of somatic hybrids from parental somaclones. Rapid DNA extraction with SSR analysis enabled screening of calluses to identify somatic hybrid tissue prior to plant regeneration. The somatic hybrids were highly polyploid, mostly hexaploid (2n=6x=72), possibly due to fusion of endopolyploid protoplasts and/or chromosomal doubling during plant regeneration.  相似文献
