Vaccination of pigs against the helminth nematode Trichinella could be a good alternative to prevent the risk of human infection. In order to develop an efficient and safe vaccine, the choice of the adjuvant is an important issue. In this study, two adjuvants were selected to prepare vaccines based on total soluble Trichinella spiralis muscle larvae (ML) antigen: Montanide ISA 70 water in oil emulsion and Montanide IMS nanoparticles. Aluminium hydroxide was used as a reference adjuvant. The immune response was checked by ELISA of parasite antigen specific IgG1 and IgE. Finally, protection induced in vaccinated mice was measured after a T. spiralis challenge by counting ML burdens. The results clearly showed an impact of adjuvants on the specific IgG1 and IgE antibody responses against T. spiralis. Differences were observed between the rates of protection induced according to the type of formulation, although the three adjuvants tested were able to enhance the humoral immune response. This work demonstrated the need to use an adjuvant to obtain a specific IgG1 and IgE responses directed against the total soluble extract of T. spiralis.  相似文献

To explain the elevated osmium isotope (186Os-187Os) signatures in oceanic basalts, the possibility of material flux from the metallic core into the crust has been invoked. This hypothesis conflicts with theoretical constraints on Earth's thermal and dynamic history. To test the veracity and uniqueness of elevated 186Os-187Os in tracing core-mantle exchange, we present highly siderophile element analyses of pyroxenites, eclogites plus their sulfides, and new 186Os/188Os measurements on pyroxenites and platinum-rich alloys. Modeling shows that involvement in the mantle source of either bulk pyroxenite or, more likely, metasomatic sulfides derived from either pyroxenite or peridotite melts can explain the 186Os-187Os signatures of oceanic basalts. This removes the requirement for core-mantle exchange and provides an effective mechanism for generating Os isotope diversity in basalt source regions.  相似文献

During animal development, several planar cell polarity (PCP) pathways control tissue shape by coordinating collective cell behavior. Here, we characterize by means of multiscale imaging epithelium morphogenesis in the Drosophila dorsal thorax and show how the Fat/Dachsous/Four-jointed PCP pathway controls morphogenesis. We found that the proto-cadherin Dachsous is polarized within a domain of its tissue-wide expression gradient. Furthermore, Dachsous polarizes the myosin Dachs, which in turn promotes anisotropy of junction tension. By combining physical modeling with quantitative image analyses, we determined that this tension anisotropy defines the pattern of local tissue contraction that contributes to shaping the epithelium mainly via oriented cell rearrangements. Our results establish how tissue planar polarization coordinates the local changes of cell mechanical properties to control tissue morphogenesis.  相似文献
