A 20-year-old pony mare with persistent ventricular tachycardia (VT) was examined cardiopathologically. At necropsy, the heart was enlarged and rounded with both ventricular dilatation. In a longitudinal section of the ventricular septum, a large grayish white patchy lesion (5 x 25 mm) was detected in the relatively higher portion. Microscopically, the lesion was extensive myocardial fibrosis located in the vicinity of the proximal part of the left bundle branch. Partially the fibrotic lesion was in contact with the branch. Such a lesion might play an important role in creating a suitable background for the development of VT via automaticity or reentry mechanism. 相似文献

ABSTRACT: To clarify the quantitative changes in the transport of orally intubated protein into the blood circulation as macromolecules in development, immunoglobulin Y (IgY) extracted from chicken eggs was administered orally to juvenile Japanese eel, Anguilla japonica . For the first experiment, which was performed before the commencement of artificial feeding, the oral delivery of 2.0 μg/0.1 g bodyweight of IgY resulted in a rapid increase in plasma IgY to a maximum of 2.30 μg/mL. However, the transport of IgY into the blood decreased significantly in the experiments that followed, which were performed after 12, 25 and 42 days. During this period, bodyweight increased approximately by a factor of eight, and rapid growth of the stomach was observed histologically. Possible contributions for the development of the alimentary canal to the diminishment of intestinal protein assimilation are discussed. 相似文献

Using cDNA from a CRFK cell line as a template, PCR amplification was performed with the Ub1S and poly(dT) primers to isolate feline ubiquitin genes. Sequencing of the 495 bp PCR fragment revealed that the putative amino acids induced by this fragment gave a fusion protein consisting of a ubiquitin polypeptide (76 amino acids) and an extension protein of ribosomal proteins L40 (52 amino acids). The putative amino acid sequence of ubiquitin was identical to those of humans, rats and pigs.The recombinant glutathione S-transferase (GST)–feline ubiquitin fusion proteins were produced in Escherichia coli and purified. The fusion proteins had a molecular weight of about 42 kDa and were detected by immunoblot assay with rabbit anti-ubiquitin antiserum.The mRNAs from heat-shocked and non-heat-shocked cells were subjected to RT-PCR (Ub1S and poly(dT) primers) analysis. The molecular weights of the ubiquitinated proteins in heat-shocked CFRK cells were between 18 kDa and 24 kDa by immunoblot assay.These results suggested that there were more ubiquinated proteins in the heat-shocked CRFK cells than in the pre-heat-shocked cells. 相似文献

It was recently reported that canine parvoviruses (CPV) had entered cat populations and induced disease in infected cats, while they had affected only dogs in the past. It is important to determine whether conventional feline panleukopenia virus (FPLV) vaccines protect against recent CPV infections. In this study, the cross-reactivity of virus-neutralising (VN) and haemagglutinin-inhibition (HI) antibodies in cats induced by FPLV and CPV s were examined. Lower cross-reactivities of VN and HI antibodies against each CPV strain were observed in cats experimentally inoculated with FPLV or vaccinated with an inactivated FPLV vaccine. In addition, we revealed the existence of a novel type of FPLV, which reacted weakly with antibodies induced by the conventional FPLV vaccine. 相似文献

New Rice for Africa (NERICA) is a general name for interspecific rice varieties derived from a cross between the high‐yielding Asian rice (Oryza sativa L.) between locally adapted African rice (Oryza glaberrima Steud.). Eight NERICAs were evaluated for cold tolerance (CT) at the reproductive stage and compared with their O. sativa parents and three Japanese standard rice varieties over 3 years. Cold tolerance was evaluated based on the filled grain ratio (FGR) after cold water irrigation. The FGR was greatly reduced by cold water irrigation. NERICA 1, 2 and 7 had higher FGR (51.9–57.9 %), while NERICA 6, 15 and 16 had lower FGR (6.2–14.5 %). NERICA 1, 2 and 7 were less affected by cold stress, with a 31 % mean reduction in FGR, while NERICA 6, 15 and 16 were greatly affected, with their FGRs being reduced by more than 80 %. NERICA 3 and 4 were moderately affected by cold stress, with about 45 % reduction rate in FGR. FGR significantly influenced the grain weights of the varieties with strong positive correlations (r = 0.83–0.91; P < 0.001), and thus, similar trends in grain weights were observed. Grain weights were reduced by 61.7–96.4 % under cold stress. NERICA 1, 2 and 7 showed significantly better performance than NERICA 3 and 4, while NERICA 6, 15 and 16 performed poorly under cold water irrigation. The Japanese varieties Koshihikari (very tolerant) and Ozora (moderately tolerant) were more affected by cold water irrigation than NERICA 1, 2 and 7. On the basis of the mean reduction rate (%) in FGR under cold stress, the varieties were classified as follows: NERICA 1, 2 and 7 as tolerant; NERICA 3 and 4 as moderately tolerant; and NERICA 6, 15 and 16 as susceptible to cold stress. However, NERICA 7 grain yields were lower under cold stress due to both greatly reduced number of panicles per plant and number of spikelets per panicle. To investigate the reason for the high mortality of cultured juvenile Pacific bluefin tuna (PBT), especially during land‐based culture, we examined the effect of the rate of light‐intensity change during the dawn period. Juvenile PBT kept in a natural light environment (control) were compared with those exposed to an artificially slowed change in light intensity during the dawn period (test group). For the test group, lamps, which were connected to automatic timers and placed above the rearing tank, were switched on from 0345 to 0515 h at 15‐min intervals. After 9 d, the survival rate of the test and control group was identical. Between Day 1 and Day 6, whole‐body cortisol levels were not different between groups. However, whole‐body glucose levels in the test group were significantly higher than those in the control group. Both groups displayed a diel rhythm in plasma cortisol concentration that peaked at 0300–0600 h. These results suggest that slowing the increase in light intensity during the dawn period did not improve survival and that low light intensity itself induces high mortality in cultured PBT juveniles. 相似文献

The degree of intramuscular adipose tissue accumulation is one of the factors affecting meat quality. Accumulation of adipocytes is also observed under the pathological condition of skeletal muscle such as muscular dystrophy and sarcopenia. The origin of adipocytes seen in skeletal muscle is mesenchymal progenitor cells that can give rise to both adipocytes and fibroblasts. In the present study, we demonstrated that siRNA-mediated suppression of MyoD expression in rat skeletal muscle progenitor cell culture, which comprises both myogenic satellite cells and mesenchymal progenitor cells, resulted in diminished myotube formation and an unexpected spontaneous appearance of white adipocytes. Suppressing myomaker expression also resulted in complete absence of myotube formation without reducing MyoD expression, but no adipogenesis was seen in this scenario, indicating that decline in MyoD expression rather than decreased myotube formation is necessary to induce adipogenesis. In addition, spontaneous adipogenesis induced by suppressing MyoD expression in culture was inhibited by the conditioned medium from control culture, indicating that anti-adipogenic factor(s) are secreted from MyoD-positive myogenic cells. These results indicate the presence of regulatory mechanism on adipogenesis by myogenic cells. 相似文献

One-day-old, 2-wk-old, and 4-wk-old call ducks (Anas platyrhyncha var. domestica) inoculated intravenously with the H5N1 highly pathogenic avian influenza virus A/chicken/Yamaguchi/7/2004 isolate (Ck/Yama/7/04) were examined clinically, pathologically, and virologically. Clinically, the birds exhibited mild-to-severe neurologic signs and corneal opacity. All birds in the 1-day-old group and one bird in the 4-wk-old group died within 4 days after the virus inoculation. Histologic changes were characterized by severe nonpurulent encephalitis and necrotic lesions of feather epithelium on day 3 postinoculation (PI) or later. Focal necrosis of myocardial cells, pancreatic acinar cells, skeletal myocytes, and corneal epithelial cells was observed. Viral antigens were detected in association with necrotic changes. Viruses were isolated from all examined organs including the skin with many feathers. Serum antibody against the virus was detected in all surviving birds on day 10 PI by hemagglutination-inhibition tests. These results suggest that Ck/Yama/7/04 has a pathogenicity that causes neurologic sign, nonpurulent encephalitis with mortality, and feather lesions for call ducks. Feather lesions with viral antigens and the virus isolation from the skin suggest that Ck/Yama/ 7/04 has a predilection for feathers in call ducks. 相似文献

Growth hormone (GH) plays a specific role to inhibit apoptosis in the bovine mammary gland through the insulin-like growth factor (IGF)-I system, however, the mechanism of GH action is poorly understood. In this study, we show that GH dramatically inhibits the expression of IGFBP-5, and GH along with IGF-I enhanced the phosphorylation of Akt through the reduction of IGF binding protein (IGFBP)-5. To determine how GH affects Akt through IGF-I in bovine mammary epithelial cells (BMECs), we examined the phosphorylation of Akt in GH treated BMECs and found that IGF-I induced phosphorylation of Akt was significantly enhanced by the treatment with GH. We demonstrated that GH reduces mRNA and protein expression of IGFBP-5 in BMECs, but it does not affect the expression of IGFBP-3. To determine that the enhanced effect of the Akt phosphorylation by the treatment of GH is due to the inhibition of the expression of IGFBP-5, we examined the effect of IGFBP-3 and -5 on the phosphorylation of Akt through IGF-I in the GH-treated BMECs. The phosphorylation of Akt was inhibited in a dose-dependent manner when IGFBP-5 was added at varying concentrations and was also inhibited in the presence of IGFBP-3. The results of this study suggest that GH plays an important role on mammary gland involution in bovine mammary epithelial cells. 相似文献

The present study was conducted to examine the effects of three tannin relatives (tannic acid, TA; gallic acid, GA; and ellagic acid, EA) on antihyaluronidase and reactive oxygen species (ROS) scavenging activity, in vitro fertilization (IVF) parameters, and the acrosome reaction (AR) induced by sperm-zona interaction. Among the three tannin relatives, TA and EA showed the strongest potency for blocking the hyaluronidase activity of boar sperm, with concentration-dependent inhibition over the range of 2-10 microg/ml. In contrast, ROSs were effectively scavenged by TA and GA, but not EA. When cumulus-free oocytes were inseminated in IVF medium containing 5 microg/ml of the tannin relatives, polyspermy was significantly reduced by TA and EA (32 and 29%, respectively) compared with oocytes treated with or without GA (51 and 69%, respectively) under conditions that maintained a high sperm penetration rate (P<0.05). Interestingly, induction of the AR by treatment of preincubated sperm with progesterone was blocked by TA and GA as a result of their higher levels of ROS scavenging activity, while EA, which possessed weak ROS scavenging activity, did not disturb induction of the AR with progesterone. However, the incidence of AR induced by sperm-zona interaction was significantly decreased by the strong antihyaluronidase actions of TA and EA compared with that in the absence of these compounds. Treatment with the compounds caused neither a protective proteolytic modification of the zona pellucida matrix before fertilization nor a reduction in acrosomal proteolytic activity or the number of zona-bound sperm. These findings suggest that the antihyaluronidase action of EA effectively prevents polyspermy by suppression of AR functionality induced by sperm-zona interaction and that hyaluronidase intervention is therefore required during porcine IVF. 相似文献
