Cassava roots, a major food in Africa, contain cyanogenic glucosides that may cause toxic effects. Malawian women farmers considered fields of seemingly similar cassava plants to be mixes of both ‘cool’ and ‘bitter’ cultivars. They regard roots from ‘cool’ cultivars as non-toxic. Roots of ‘bitter’ were considered to require extensive traditional processing done by women to be safe for consumption. But curiously, these women farmers preferred ‘bitter’ cultivars since toxicity confers protection against theft, which was a serious threat to the food security of their families. We studied how well these farmers comprehend the effects of genetic variations in cassava when dealing with cyanogenesis in this complex system. Using molecular markers we show that most plants farmers identified as belonging to a particular named cultivar had a genotype typical of that cultivar. Farmers' ethno-classification into ‘cool’ and ‘bitter’ cultivars corresponded to a genetic sub-division of the typical genotypes of the most common cultivars, with four-fold higher cyanogenic glucoside levels in the bitter cultivars. Examining morphology, farmers distinguished genotypes better than did the investigators when using a standard botanical key. Undoubtedly, these women farmers grasp sufficiently the genetic diversity of cassava with regard to cyanogenesis to simultaneously benefit from it and avoid its dangers. Consequently, acyanogenic cassava – the breeding of which is an announced good of some cassava genetic improvement programmes – is not a priority to these farmers. Advances in molecular genetics can help improve food supply in Africa by rapid micropropagation, marker assisted breeding and introduction of transgenic varieties, but can also help to elucidate tropical small-scale farmers' needs and skills. The main objectives of the study were to (a) assess the effect of perimortem stress on blood drainage, (b) compare the efficiency of gill cutting and direct gutting as bleeding methods, and (c) compare pre- and postrigor filleting strategies for presence of residual blood in Atlantic cod fillets. Anesthetized cod had significantly higher drainage of blood compared to stressed fish. Nevertheless, the visual assessments of residual blood were not affected by stress or bleeding method. Some minor, but significant, differences between pre- and postrigor fillets were found. Stressed fish were initially less light in color than those unstressed, but after ice storage there was no noticeable difference. However, initially, the largest difference in fillet color was due to different bleeding methods. Fillets cut from fish subjected to gill cutting were lighter and less red in color than those bled by direct gutting. After 7 and 21 days of storage, the color differences observed in fresh and salted fillets, respectively, were mainly due to the different rigor status when they were filleted, indicating that fillets cut postrigor were superior to fillets cut prerigor. Prerigor filleting resulted in lower water holding capacity after ice storage in anesthetized, direct gutted cod.  相似文献

In implantation trials carried out with Prosopis chilensis and Prosopis flexuosa, the following variables were considered: implantation time (December and March), implantation technique (direct seeding and seedling transplantation) and age of the seedlings at the time of transplantation (60, 105 and 150 days). Survival was evaluated for more than 9 months after implantation and trials were repeated in 3 consecutive years.

Results from both species were similar and they show that survival after transplantation was higher (60–80%) than from direct seeding (35–45%). Survival from transplantation was independent of the size and age of the seedlings and it was less affected by climatic conditions than direct field sowing. Survival from direct seeding was higher in March than in December. The best time for either implantation alternative would be the end of the summer or beginning of fall, when climatic conditions increase the probability of seedling survival.  相似文献

This work was undertaken to investigate the feasibility of using near infrared spectroscopy (NIR) and partial least squares regression (PLS) as a tool to characterize the basic wood properties of Norway Spruce (Picea abies (L.) Karst.). The wood samples originated from a trial located in the province of Västerbotten in Sweden. In this trial, the effects of birch shelterwoods (Betula pendula Roth) of different densities on growth and yield in Norway spruce understorey were examined. All Norway spruce trees in each shelterwood treatment were divided into three growth rate classes based on diameter at breast height (1.3?m) over bark. Five discs were cut from each tree (i.e. from the root stem, and at 20%, 40%, 60%, and 80% of the total height). The discs from 40% tree height were used (i.e., where the largest variations in annual ring widths and wood density were found). A total of 27 discs were selected. The discs were used for measuring annual ring widths, wood density, average fiber length and the fiber length distributions. Milled wood samples prepared from the discs were used for recording NIR spectra. PLS regression was used to generate prediction models for the wood properties (Y-matrix) and NIR spectra (X-matrix) as well as between the wood properties (Y-matrix) and the fiber length distributions (X-matrix). One set of models was generated using untreated spectra and fiber length distributions. For a second set of models the structure in the X-matrix, which was orthogonal to the matrix described by the wood properties, was eliminated using a soft target rotation technique called orthogonal signal correction (OSC). The PLS model obtained using “raw” untreated NIR spectra and fiber length distributions had a poor modeling power as evidenced by the cumulative Q2 values. For the PLS models based on untreated NIR spectra the cumulative Q2 values ranged from a minimum of 16% (wood density) to a maximum of 46% (no. of annual rings). Orthogonal signal correction of the X-matrix (NIR spectra or fiber length distributions) gave PLS models with a modeling power corresponding to cumulative Q2 values well in excess of 70%. The improvement in predictive ability accomplished by the OSC procedure was verified by placing four of the 27 observations in an external test set and comparing RMSEP values for the test set observations without OSC and with OSC.  相似文献

Background

The Swedish control program for salmonella includes restrictions and on-farm control measures when salmonella is detected in a herd. Required control measures are subsidised by the government. This provides an opportunity to study costs for on-farm salmonella control. The aim of this study was to describe the costs for on-farm salmonella control in Swedish cattle herds and to investigate the effects of herd factors on these costs in dairy herds.

Results

During the 15 years studied there had been a total of 124 restriction periods in 118 cattle herds; 89 dairy herds, 28 specialised fattening herds and three suckler herds. The average costs per herd for on-farm salmonella control was 4.60 million SEK with a median of 1.06 million SEK corresponding to approximately 490 000 and 110 000 EUR. The range was 0.01 to 41 million SEK corresponding to 1080 EUR to 4.44 million EUR per farm. The costs cover measures required in herd-specific control plans, generally measures improving herd hygiene. A mixed linear model was used to investigate associations between herd factors and costs for on-farm salmonella control in dairy herds. Herd size and length of the restriction period were both significantly associated with costs for on-farm control of salmonella with larger herds and longer periods of restrictions leading to higher costs. Serotype detected and administrative changes in the Swedish Board of Agriculture aiming at reducing costs were not associated with costs for on-farm salmonella control.

Conclusions

On-farm control of salmonella in Swedish cattle herds incurred high costs but the costs also varied largely between herds. Larger herds and longer restriction periods increased the costs for on-farm control of salmonella in Swedish dairy herds. This causes concern for future costs for the Swedish salmonella control program as herd sizes are increasing.  相似文献

Background

Measurement of plasma concentration of natriuretic peptides (NPs) is suggested to be of value in diagnosis of cardiac disease in dogs, but many factors other than cardiac status may influence their concentrations. Dog breed potentially is 1 such factor.

Objective

To investigate breed variation in plasma concentrations of pro‐atrial natriuretic peptide 31‐67 (proANP 31‐67) and N‐terminal B‐type natriuretic peptide (NT‐proBNP) in healthy dogs.

Animals

535 healthy, privately owned dogs of 9 breeds were examined at 5 centers as part of the European Union (EU) LUPA project.

Methods

Absence of cardiovascular disease or other clinically relevant organ‐related or systemic disease was ensured by thorough clinical investigation. Plasma concentrations of proANP 31‐67 and NT‐proBNP were measured by commercially available ELISA assays.

Results

Overall significant breed differences were found in proANP 31‐67 (P < .0001) and NT‐proBNP (P < .0001) concentrations. Pair‐wise comparisons between breeds differed in approximately 50% of comparisons for proANP 31‐67 as well as NT‐proBNP concentrations, both when including all centers and within each center. Interquartile range was large for many breeds, especially for NT‐proBNP. Among included breeds, Labrador Retrievers and Newfoundlands had highest median NT‐proBNP concentrations with concentrations 3 times as high as those of Dachshunds. German Shepherds and Cavalier King Charles Spaniels had the highest median proANP 31‐67 concentrations, twice the median concentration in Doberman Pinschers.

Conclusions and Clinical Importance

Considerable interbreed variation in plasma NP concentrations was found in healthy dogs. Intrabreed variation was large in several breeds, especially for NT‐proBNP. Additional studies are needed to establish breed‐specific reference ranges.  相似文献

Traditional fisheries management theory supports aggressive exploitation of old and large fish to maximize a stock’s biomass production and yield. Here we present an age-structured fish population model with multidimensional density-dependence to test the hypotheses that protection of large, fecund individuals is beneficial for the population and selected fisheries variables and that effects of maternal size on early survival rate change the resilience and fisheries productivity of a pike population (Esox lucius L.) exploited by recreational angling. We find that, compared to the traditional regulatory approach of management by small minimum-length limits (so that culling of large fish is encouraged), preservation of large and old individuals through harvestable-slot length limits promises considerable benefits for fisheries quality, without compromising the long-term conservation of the population. We also find that ignoring maternal effects on early survival of offspring might overestimate the equilibrium spawning stock abundance by up to 17% and the predicted harvest by up to 11%, potentially putting pike populations at risk from overharvest if size-dependent maternal effects are ignored in fisheries models. If the findings from our simulation study hold for empirical systems, they suggest altered harvest regulations in many of consumptive pike recreational fisheries are needed to protect large individuals to a greater extent that currently pursued.  相似文献

A long-term study was carried out in 11 dairy herds in the Khon Kaen province of northeast Thailand between August 2001 and November 2004. The objective was to investigate seroprevalence dynamics of Neospora caninum infection in the herds and to demonstrate patterns of seroconversion in individual cattle. Each herd was visited once a year, in total four times, and sera from cattle > 3 months of age and farm dogs as well as a sample from the bulk milk were collected. All samples were analysed for presence of specific antibodies by an N. caninum iscom ELISA. The overall percentage of antibody-positive cattle was constant and varied only between 10 and 13% over the 4 years, but the variation in within-herd seroprevalence between herds was substantial. Two herds had > or = 20% seropositive animals at all samplings and consistently high bulk milk OD, whereas two herds had no seropositive animal at the last two samplings and low bulk milk OD. Five herds had a decreasing trend of within-herd seroprevalence, whereas the remaining six herds had a higher portion of test-positive individuals at the end of the study. A total of 424 individuals were sampled more than once; 344 (81%) and 32 (8%) were consistently antibody-negative and antibody-positive, respectively. The proportions of animals that changed from being seronegative to seropositive and from being seropositive to seronegative between the years were 3.9-4.6% and 19-39%, respectively. Apparent vertical and horizontal transmission rates were 58% (95% CI; 44-71%) and 5% (95% CI; 3-7%), respectively. In conclusion, the overall percentage of N. caninum antibody-positive cattle was constant over the years, but the within-herd seroprevalence varied substantially between the herds. Seroconversions were likely to occur in individual cattle although most animals had consistent serological status throughout the study.  相似文献

Neural circuits regulate cytokine production to prevent potentially damaging inflammation. A prototypical vagus nerve circuit, the inflammatory reflex, inhibits tumor necrosis factor-α production in spleen by a mechanism requiring acetylcholine signaling through the α7 nicotinic acetylcholine receptor expressed on cytokine-producing macrophages. Nerve fibers in spleen lack the enzymatic machinery necessary for acetylcholine production; therefore, how does this neural circuit terminate in cholinergic signaling? We identified an acetylcholine-producing, memory phenotype T cell population in mice that is integral to the inflammatory reflex. These acetylcholine-producing T cells are required for inhibition of cytokine production by vagus nerve stimulation. Thus, action potentials originating in the vagus nerve regulate T cells, which in turn produce the neurotransmitter, acetylcholine, required to control innate immune responses.  相似文献

Electrophysiological techniques were used to analyse the effects, on the activity in the sensory nerves of Periplaneta americana and Blabera craniifer, after topical applications of three different pyrethroid insecticides: deltamethrin, (1R)-tetramethrin, and 5-benzyl-3-furylmethyl (E)-(1R)-cis-2,2-dimethyl-3-(2-oxothiolan-3-ylidenemethyl)cyclopropanecarboxylate (RU-15525; ‘Kadethrin’). The insecticides were applied: (a) to the cercus, and the activity was recorded on the cercal nerve and on the central nervous connectives; or (b) to the last segments of the metathoracic leg, and the corresponding activity was recorded on the crural nerve. In both cases, several categories of receptor were affected but the major part of the recorded activity came from the mechanoreceptors, the functional properties of which were monitored throughout the experiments. In most cases, topical application of small amounts of pyrethroid were found to induce an increase in the spontaneous activity, followed by decrease in excitability. Characteristic differences were, however, found between the three compounds and the effects were concentration- and solvent-dependent. The results were consistent with those obtained from experiments on the sense organs of vertebrates but did not support the idea that the effectiveness of pyrethroid insecticides with an α-cyanobenzyl group, compared with those without an α-cyanobenzyl group, originates from a greater effect on the peripheral sensory receptors.  相似文献
