Cells use transporters of different affinities to regulate nutrient influx. When nutrients are depleted, low-affinity transporters are replaced by high-affinity ones. High-affinity transporters are helpful when concentrations of nutrients are low, but the advantage of reducing their abundance when nutrients are abundant is less clear. When we eliminated such reduced production of the Saccharomyces cerevisiae high-affinity transporters for phosphate and zinc, the elapsed time from the initiation of the starvation program until the lack of nutrients limited growth was shortened, and recovery from starvation was delayed. The latter phenotype was rescued by constitutive activation of the starvation program. Dual-transporter systems appear to prolong preparation for starvation and to facilitate subsequent recovery, which may optimize sensing of nutrient depletion by integrating internal and external information about nutrient availability.  相似文献

Perennial plants are known to be one of the most influential parameters in desert ecosystems affecting microbial activity. In this study, we examined the importance of these perennial shrubs and attempted to determine the most influential factor that contributes the most to the ecosystem by separating the physical part and the organic contribution of perennial plants. The study site is located in the northern Negev Desert, Israel, where 50 Hammada scoparia shrubs and 50 artificial plants were randomly marked to be used as a tool for the above objectives. Soil samples were collected monthly in the vicinity of the canopies of both shrubs while control samples were collected from the open areas between the shrubs. All samples were collected from the upper (0–10 cm) and the deeper (10–20 cm) soil layers. The contribution to microbial activity was measured by evaluation of the microbial community functions in soil. The results of the research showed a trend of a strong influence of the perennial H. scoparia shrubs on microbial community function. The functional aspects of the microbial community that were measured were CO2 evolution, microbial biomass, microbial functional diversity, and the physiological profile of the community. High values for all parameters were observed under the vicinity of the H. scoparia shrubs, while the artificial plants were found to have a weaker effect on the community according to soil depth. The upper-soil layer at both locations (artificial plant canopy and control samples) showed higher values of the functioning parameters than that of the deeper soil layer. The results indicate the importance of the organic contribution (plant litter) in comparison to the physical part of desert shrubs.  相似文献

The Mediterranean ecosystem of the Carmel Mountain ridge in Israel is subjected to an increasing number of forest fires of various extents and severities due to intense human activities in the region. On 8 April 2005, a low-moderate severity forest fire occurred at the northwestern part of the ridge and burned more than 150 ha of natural vegetation. Soil water repellency (WR) is a property usually modified by the litter and soil organic matter combustion as a consequence of fire, which has implications for the hydrological balance in the affected soils. A field study was conducted with the following objectives: 1) to investigate in situ WR changes at three soil depths as a consequence of the fire, 2) to evaluate the short-term evolution of WR under field conditions, and 3) to study the relationship between pre-fire vegetation type and slope aspect on the persistence of WR in the burned area. Soil WR was measured by the Water Drop Penetration Time (WDPT) test. Measurements were conducted monthly at 31 field sites within the burned area over a period of seven months (April 2005–November 2005), and compared to adjacent unburned areas. Soil WR measurements included more than 3400 WDPT tests at soil surface and at 5 and 10 cm depths. The results indicate that fire induced WR in previously wettable soils exhibited high levels of persistence at the soil surface during the first six weeks after the fire, while at 5 cm depth WR persistence was lower. At 10 cm depth soil was mostly wettable. After six weeks the frequency of WR occurrence diminished at the soil surface and at 5 cm depth. In addition, WR was found to be highly related with the pre-fire vegetation type and with slope aspect.  相似文献

Cells remove proteins by two processes: degradation and dilution due to cell growth. The balance between these basic processes is poorly understood. We addressed this by developing an accurate and noninvasive method for measuring protein half-lives, called "bleach-chase," that is applicable to fluorescently tagged proteins. Assaying 100 proteins in living human cancer cells showed half-lives that ranged between 45 minutes and 22.5 hours. A variety of stresses that stop cell division showed the same general effect: Long-lived proteins became longer-lived, whereas short-lived proteins remained largely unaffected. This effect is due to the relative strengths of degradation and dilution and suggests a mechanism for differential killing of rapidly growing cells by growth-arresting drugs. This approach opens a way to understand proteome half-life dynamics in living cells.  相似文献

The Mediterranean ecosystem of Mt. Carmel is subjected to increasing number of forest fires at various extents and severities due to increasing human activities. Accordingly, we tested whether in areas exposed to different fire histories vegetation regeneration is different in north versus south facing slopes, and the potential impact on erosion processes. Using remote sensing techniques we evaluated the Enhanced Vegetation Index (EVI) to monitor vegetation recovery following a single fire and three successive fires, using a series of Landsat images taken between 1985–2002. Following a single fire, vegetation cover reached pre-disturbance values within less than 5 years. Repeated fires caused further reduction of EVI values, especially at south facing slopes (SFS). The effects of three successive fires within 10 years, followed by a three year recovery period, however, are negligible when considering vegetation cover values. This was deduced as north facing slope EVI values returned to pre-disturbance conditions at the end of the 3 years and SFS EVI values to 80% of the pre-disturbance conditions. Our results indicate that Mediterranean eco-geomorphic systems are quite resilient, showing quick response, at least in terms of return to pre-disturbance states of vegetation cover, and hence of soil erosion rates. This is true not only in response to a disturbance caused by a single fire, but also for repetitive fire incidents.  相似文献

The gram-negative bacterium Acidovorax citrulli causes bacterial fruit blotch (BFB) disease of cucurbits, which represents a serious threat to melon and watermelon production worldwide. To date, there are no efficient means to manage the disease, and reliable resistance sources for cucurbit germplasm are lacking. Mineral nutrition markedly affects plant diseases. Recently, we reported that disease severity on melon foliage and A. citrulli growth in the leaf tissue were significantly influenced by the form of nitrogen supply. In the present study, we investigated the influence of potassium nutrition on BFB severity and A. citrulli establishment in the foliage of melon plants. Fertilization with relatively low concentrations of potassium increased these variables compared with higher potassium concentrations. Since establishment of A. citrulli during the growing season is assumed to increase the incidence of fruit infection, the fact that mineral nutrition influences BFB incidence in the plant foliage is of particular importance.  相似文献

Biological soil crusts (BSCs) and the soils directly below crusts (SDBCs) (0–5 mm) were collected in the Negev Desert (Israel) during the wet and dry seasons of 2007 and 2008, gently separated, and microbial basal respiration, microbial biomass carbon (Cmic), carbon (C) source utilization rates, and catabolic diversity were analyzed using MicroRespTM plates. The seasonal-change patterns of these parameters were similar to those of soil organic C (Corg) in the BSCs, i.e., increases were observed during the dry seasons relative to the wet seasons. Few seasonal variations in qCO2 and Cmic/Corg in the BSCs indicated that the increases in crustal organism basal respiration and C source utilization rates can be attributed to microbial propagation as a result of the increases in available C during the dry seasons. High frequency of rain events, with precipitation higher than 0.1 mm during spring, can enable crustal organisms to maintain photosynthetic activity and can facilitate microbial propagation and Corg accumulation in the BSCs. The seasonal dynamics of the four biotic parameters in the SDBCs were the opposite of those of the BSCs, and C source utilization rates and catabolic diversity were higher than in the BSCs during the wet seasons. Downward migration of exopolysaccharides, crustal organism cell contents, and intracellular solutes with water infiltration can increase C and nutrient availability and enhance microbial catabolic activities and propagation in the SDBCs.  相似文献

Roots of the herb Panax ginseng are known to contain high levels of bioactive saponins. Here, we isolated saponins from ginseng root powder and studied their inhibitory effect on the absorption of dietary fat in male Balb/c mice. Consumption of ginseng saponins suppressed the expected increase in body weight and plasma triacylglycerols, following a high-fat diet and observed higher intake. Consumption of ginseng saponins had no effect on the concentration of the total plasma cholesterol in both chow and high-fat diets in mice. The mode by which saponins from ginseng inhibit lipid metabolism was assessed as the in vitro inhibition of pancreatic lipase. Ginseng saponin inhibited pancreatic lipase with an apparent IC50 value of 500 mug/mL. Our results suggest that the anti-obesity and hypolipidemic effects of Ginseng in high-fat diet-treated mice were attributed to the isolated saponin fraction. These metabolic effects of the ginseng saponins may be mediated by inhibition of pancreatic lipase activity.  相似文献
