Size and composition of the weed seedbank was assessed after 12 years of application of four tillage systems in two crop rotations. Mouldboard and chisel ploughing at 45 cm, minimum tillage at 15 cm and no tillage were compared in continuous winter wheat and a pigeon bean/winter wheat 2-year rotation. Weed control was based upon post-emergence herbicide application. Weed seedling emergence from soil samples taken at 0–15, 15–30 and 30–45 cm depths was assessed in a non-heated glasshouse for 12 months. The tillage system influenced weed seedbank size and composition to a much greater extent than crop rotation. Total weed seedling density was higher in no tillage, minimum tillage and chisel ploughing plots in the 0–15, 15–30 and 30–45 cm layers respectively. Density in the whole (0–45 cm) layer did not differ significantly among tillage systems. With no tillage, more than 60% of the total seedlings emerged from the surface layer, compared with an average 43% in the other tillage systems. Crop rotation did not influence either weed seedbank size or seedling distribution among soil layers, and only had a small influence on major species abundance. The weed seedbank was dominated (>66%) by Conyza canadensis (L.) Cronq. and Amaranthus retroflexus (L.), which thrived in chisel ploughing and no tillage respectively. Results suggested that crop rotation and substitution of mouldboard ploughing by non-inversion tillage (especially by minimum tillage) would not result in increased weed problems, whereas use of no tillage might increase weed infestations because of higher seedling recruitment from the topsoil.  相似文献

Plasma cortisol is the most commonly used indicator of stress in fish but, as the blood sampling procedure itself can be a source of stress, it would be helpful to measure cortisol using less invasive matrices. It is also necessary to find alternative matrices as stress indicators in dead fish in which blood sampling is impossible. In the present study, we investigated transport stress in three aquaculture species, European sea bass (Dicentrarchus labrax L.), common carp (Cyprinus carpio L.) and rainbow trout (Oncorhynchus mykiss Walbaum), by cortisol determination (radioimmunoassay) in plasma and other matrices (skin mucus, gut content, lateral muscle and caudal fin). Cortisol significantly increased after transport in all species and matrices, except in the sea bass gut content, where it remained unchanged. The three species responded to transport stress by producing different cortisol levels. In conclusion, the significant correlation found between plasma cortisol and most of the other matrices opens up the possibility of using them to evaluate stress in fish: mucus sampling is a less invasive method than blood sampling, and in addition to muscle and fin sampling, it can be used in postmortem fish.  相似文献

Membrane transporters that use energy stored in sodium gradients to drive nutrients into cells constitute a major class of proteins. We report the crystal structure of a member of the solute sodium symporters (SSS), the Vibrio parahaemolyticus sodium/galactose symporter (vSGLT). The approximately 3.0 angstrom structure contains 14 transmembrane (TM) helices in an inward-facing conformation with a core structure of inverted repeats of 5 TM helices (TM2 to TM6 and TM7 to TM11). Galactose is bound in the center of the core, occluded from the outside solutions by hydrophobic residues. Surprisingly, the architecture of the core is similar to that of the leucine transporter (LeuT) from a different gene family. Modeling the outward-facing conformation based on the LeuT structure, in conjunction with biophysical data, provides insight into structural rearrangements for active transport.  相似文献

Amyloid diseases, including Alzheimer's, Parkinson's, and the prion conditions, are each associated with a particular protein in fibrillar form. These amyloid fibrils were long suspected to be the disease agents, but evidence suggests that smaller, often transient and polymorphic oligomers are the toxic entities. Here, we identify a segment of the amyloid-forming protein αB crystallin, which forms an oligomeric complex exhibiting properties of other amyloid oligomers: β-sheet-rich structure, cytotoxicity, and recognition by an oligomer-specific antibody. The x-ray-derived atomic structure of the oligomer reveals a cylindrical barrel, formed from six antiparallel protein strands, that we term a cylindrin. The cylindrin structure is compatible with a sequence segment from the β-amyloid protein of Alzheimer's disease. Cylindrins offer models for the hitherto elusive structures of amyloid oligomers.  相似文献

The three-dimensional structure of ribulose-1,5-biphosphate carboxylase-oxygenase (RuBisCO), has been determined at 2.6 A resolution. This enzyme initiates photosynthesis by combining carbon dioxide with ribulose bisphosphate to form two molecules of 3-phosphoglycerate. In plants, RuBisCO is built from eight large (L) and eight small (S) polypeptide chains, or subunits. Both S chains and the NH2-terminal domain (N) of L are antiparallel beta, "open-face-sandwich" domains with four-stranded beta sheets and flanking alpha helices. The main domain (B) of L is an alpha/beta barrel containing most of the catalytic residues. The active site is in a pocket at the opening of the barrel that is partly covered by the N domain of a neighboring L chain. The domain contacts of the molecule and its conserved residues are discussed in terms of this structure.  相似文献

Abnormally high spiking activity can damage neurons. Signaling systems to protect neurons from the consequences of abnormal discharge activity have been postulated. We generated conditional mutant mice that lack expression of the cannabinoid receptor type 1 in principal forebrain neurons but not in adjacent inhibitory interneurons. In mutant mice,the excitotoxin kainic acid (KA) induced excessive seizures in vivo. The threshold to KA-induced neuronal excitation in vitro was severely reduced in hippocampal pyramidal neurons of mutants. KA administration rapidly raised hippocampal levels of anandamide and induced protective mechanisms in wild-type principal hippocampal neurons. These protective mechanisms could not be triggered in mutant mice. The endogenous cannabinoid system thus provides on-demand protection against acute excitotoxicity in central nervous system neurons.  相似文献

Despite possible agronomic and environmental benefits, the diffusion of soil conservation tillage systems in Italy is currently rather low. The aim of this study was to compare the performance of different soil tillage techniques, in an effort to identify suitable soil management options for irrigated crops in Central Italy. An experiment was carried out on maize and soybean from April to October in two consecutive years (1993 and 1994) in Maccarese (a coastal location near Rome). The systems compared were: conventional mouldboard ploughing (CT), minimum tillage, ridge tillage and no-tillage (NT). In 1993, actual crop evapotranspiration was measured throughout the growing season on NT and CT soybean, using a micrometeorological technique.
No significant differences due to soil tillage were found for grain yield and yield irrigation water use efficiency (IWUEy), except for soybean in 1994, in which yields and IWUEy were 59 % higher on conservation tillage treatments compared with CT. In 1994 soybean yield water use efficiency was 10.1 and 9.5 kg ha−1 mm−1 for NT and CT respectively. The results suggest that the adoption of soil conservation tillage is feasible, for the specific cropping system, with equivalent or better performances as conventional tillage.  相似文献
