选用32头经产和16头初产荷斯坦奶牛进行不完全8×8拉丁方设计试验(4周为期,共12周),进行评估粗蛋白(CP)、可消化瘤胃非降解蛋白(RUP)和瘤胃保护蛋氨酸(RPM,饲喂的是美国乔治亚州肯尼索德固赛公司的产品Mepron)。日粮舍(按干物质(DM)计)苜蓿青贮21%、玉米青贮34%、高水分玉米22%-26%、豆粕10%-14%、大豆皮4%、添加脂肪2%、矿物质和维生素13%、中性洗涤纤维27%-28%。添加或不添加压榨大豆粕的瘤胃降解蛋白(RUP),配制为15.8%或17.1%日粮粗蛋白,0或9g RPM/d的2×2×2因子设计。结果表明,二向或三向交互作用都不显著。高日粮CP增加表明采食量1.1kg·d^-1 DM、产奶量1.7kg·d^-1、3.5%乳脂校正乳(FCM)2.2kg·d^-1、乳脂量0.10kg·d^-1、真蛋白0.05kg·d^-1,改善了表观氮平衡及干物质与纤维的消化率。然而,母牛采食较高日粮CP时,乳中尿素氮和估计尿液尿素氮和总氮排泄也增加,表观氮效率(牛奶中氮量,采食氮量)从33%下降到30%。饲喂较多RUP的积极效果在于提高饲料转化率和乳脂肪含量(FCM增加1.8kg·d^-1,乳脂量增加0.08kg·d^-1),但是乳蛋白含量下降,乳中尿素氮和尿液尿素排泄增加..添加RPM增加DM采食量0.7kg·d^-1,FCM增加1.4kg·d^-1,乳脂量增加0.06kg·d^-1,并有提高乳脂率、产奶量和乳蛋白产量的趋势。  相似文献

Two ruminally cannulated cows were fed a diet containing 65% alfalfa haylage and 35% concentrate every 12 h. During two collection periods, whole ruminal contents were sampled before feeding, when feed not eaten was removed (1 h after initiation of feeding) and 1, 2, 3, 4, 6, 8, 10 and 12 h after removal. At each sampling, fluid-associated microorganisms were obtained by straining whole ruminal contents through eight layers of cheesecloth. A portion of the particle-associated population was obtained by chilling squeezed particles before seven successive extractions with saline solution. Microbial ash concentrations ranged from 9.9 to 16.6% of dry matter, with differences due to population (P less than .01) and to time after feeding for both fluid- (P less than .05) and particle-associated (P less than .01) microorganisms. Within 1 h after initiation of feeding, N decreased (P less than .05) from a pre-feeding concentration of 9.5% of organic matter to 7.7% for fluid-associated microorganisms, and from 9.2 to 6.7% for particle-associated microorganisms. Both populations did not return to pre-feeding concentrations until 6 h after feed removal. Nucleic acid concentrations decreased (P less than .05) in both populations after feeding, and did not return to pre-feeding concentrations until 6 h for fluid-associated and 4 h for particle-associated microorganisms. Marine turtles are a taxon of world-wide conservation concern. Effective long-term monitoring is hampered by the fact that populations are widely dispersed except during the breeding season. Thus most monitoring programmes focus on nesting beaches, necessitating resource intensive studies, often over months, that could conceivably, be less comprehensive and focus on more parsimonious sampling. We analyse 11 years of exhaustive monitoring data for two species of Mediterranean marine turtles (Chelonia mydas and Caretta caretta). We resample using a variety of plausible sub-sampling regimens to estimate the total annual nesting population. We project our dataset into the future applying a range of population change rates to explore how adopting a monitoring programme based on sub-sampling would, for example, affect our ability to detect population decline. We show that accurate annual population estimates can be achieved with as few as 14 days of survey effort providing monitoring spans the peak of nesting. Furthermore, the modelled impact of sampling-based monitoring suggests that the duration to detect population change is not increased greatly. Our findings have implications for all marine turtle monitoring and have applicability to other animal groups. It is often considered desirable to perform exhaustive monitoring, with aversion of basing policy recommendations on partial data. However, comprehensive long-term monitoring programmes, particularly in developing nations, although presenting a number of advantages, are often impossible. Accurate total annual censuses can be achieved through a variety of sub-sampling regimens without sacrificing the ability to detect changes in the population trends over time. In this example, a solid block of at least 3 weeks sampling that encompasses the peak of the nesting season is advised.  相似文献

The scombrid Scomberomorus semifasciatus is an important component of inshore fisheries in tropical Australia. Data on the parasite fauna of 593 fish from areas off northern and eastern Australia were examined for evidence of discrete fish populations. The parasites used were juveniles of Pterobothrium pearsoni, Callitetrarhynchus gracilis, Anisakis simplex (sensu latu) and Terranova sp. Tukey Kramer pairwise comparisons gave significant differences in the abundances of two or more parasites between fish from the east coast, the eastern Gulf of Carpentaria and the remainder of northern Australia. Multivariate analysis gave further evidence of differences and the results suggest that at least 4 populations or stocks of grey mackerel occur along the northern and eastern coastline of Australia.  相似文献

The study of magnetic phase transitions in insulating molecular solids provides new insights into mechanisms of magnetic coupling in the solid state and into critical phenomena associated with these transitions. Only a few such materials are known to display cooperative magnetic properties. The use of high-spin molecular components would enhance intermolecular spin-spin interactions and thus a series of chargetransfer (CT) salts have been synthesized that utilize the spin S = 1 molecular cation, [Mn(C(5)(CH(3))(5))(2)](+) (decamethylmanganocenium). The structure and cooperative magnetic behavior of [Mn(C(5)(CH(3))(5))(2)](+)[TCNQ(-) (decamethylmanganocenium 7,7,8,8-tetracyano-p-quinodimethanide) are reported. This salt is a bulk molecular ferromagnet with the highest critical (Curie) temperature (T(c) = 6.2 K) and coercive field (3.6 x 10(3) gauss), yet reported for such a material.  相似文献

Ruminal degradation of eight soluble proteins and 10 protein meals was determined using three methods: 1) an inhibitor in vitro system (IIV), to which inhibitors are added to prevent metabolism of protein degradation products, 2) in situ incubations in nylon bags and 3) in vitro NH3 production in typical ruminal inoculum. In vitro NH3 production rate from different proteins was not related to in situ or IIV degradation rate. Degradation rates for soluble proteins by IIV ranged from .103 to .813/h, yielding estimated extents of degradation that ranged from 73 to 94% (assuming ruminal passage of .05/h). For seven of the protein meals, degradation rates measured by IIV were threefold greater than in situ rates. However, mean degraded fractions estimated from zero-time intercepts were twofold greater using the in situ method, and calculated extents of degradation averaged 83% of IIV values. Extents of degradation estimated (assuming ruminal passage of .05/h) for fish meal, soybean meal, linseed meal, sunflower meal, rapeseed meal, copra meal and meat and bone meal were, respectively; 55, 79, 84, 59, 75, 54 and 58% (IIV) and 46, 63, 69, 51, 52, 43 and 55% (in situ). These values were generally similar to those reported in the literature. The extent of degradation of feather meal was 28% by in situ determination. Neither the IIV nor in situ method gave significant regressions for blood meal; neither method yields reliable data for very slowly degraded proteins. The IIV procedure has the advantage over the in situ method because it can yield degradation estimates for soluble as well as insoluble proteins.  相似文献

A method is described for simultaneous recording of in-vivo nerve activity and toxicological symptoms from 6th-instar larvae of Spodoptera littoralis poisoned with insecticide. Topically applied doses (9 ng-180 μg insect?1) of cypermethrin induced hyperactivity, repetitive activity and bursting in the ventral nerve cord; these activities were associated with different stages of poisoning in the whole animal and are consistent with pharmacokinetic studies. Nerve activity continued long after the larvae appeared to be dead (i.e. up to 5-6 days), although microscopic examination of these larvae revealed that they were still capable of small movements. At the lower doses (up to the LD95) time to death was inversely proportional to dose. Death of the organism may be related to the number of neurones which are affected by the insecticide.  相似文献

A laboratory-scale experiment was conducted with lucerne (Medicago sativa) to determine the effects of acid treatment on proteolysis during ensiling and during subsequent in vitro ruminal protein incubations. Lucerne [300 g dry matter (DM) kg?1 forage] was either untreated (control) or treated with sulphuric, formic or trichloroacetic acid (a protein precipitant that stops enzyme activity) at levels sufficient to adjust immediately forage pH to 4·0, then conserved as either silage or hay. Time-course data indicated that non-protein nitrogen (N) formation was 70–90% complete after 1 d of fermentation in the silo. Non-protein N concentrations (g kg?1 total N) were 177 at ensiling and increased to 567 (control), 426 (sulphuric), 398 (formic) and 263 (trichloroacetic) after 60 d of ensiling. Because non-protein N in silage treated with formic and sulphuric acids was nearly three times greater than that in silage treated with trichloroacetic acid, it is clear that the typical acid treatments only slow proteolysis and do not destroy protease activity during ensiling. The ruminal protein degradation rate of conserved forages was slower than that of fresh-cut forage that was preserved with dry ice immediately after cutting. The degradation rate of all acid-treated forages was similar, indicating a consistent effect on ruminal degradation regardless of method of preservation. There was a clear effect of acid treatment on reducing the rate and extent of ruminal degradation of protein in lucerne hay.  相似文献
