Red beetroot has been ranked among the 10 most potent antioxidant vegetables, although only extraction-based methods have been used to evaluate its total antioxidant capacity. Therefore, the present study aims at comparing the traditional extraction-based method with two more recent approaches (QUENCHER -QUick, Easy, New, CHEap and Reproducible- and GAR -global antioxidant response method), in order to establish their suitability in the case of beetroot. Our results indicate that the total antioxidant capacity of beetroot would be underestimated when using extraction-based procedures, since both QUENCHER and GAR methods resulted in a higher total antioxidant capacity. The effect of a thermal treatment on the total antioxidant capacity of beetroot varies among the methods evaluated and our findings suggest different compounds responsible for the total antioxidant capacity detected in each pre-processing method. Remarkably, the present study demonstrates that the traditional extraction-based method seems useful to screen for (changes in) the “bioavailable” antioxidant potential of the root. 相似文献

Thermal denaturation of recombinant human lactoferrin from transgenic rice with different degrees of iron saturation has been studied by differential scanning calorimetry. The maximum temperature, enthalpy change, and activation energy of denaturation were higher when recombinant lactoferrin was more saturated with iron, indicating an increase in the stability of the protein structure. Maximum temperature and activation energy values for apo- and holo-lactoferrins were practically identical to those reported for the same forms of lactoferrin from human milk, which indicates a similar thermal stability. However, the value of enthalpy change for denaturation of the recombinant lactoferrin was 2.5-3-fold lower than that found for the human milk protein. This finding may reflect the influence that the different glycosylation pattern may have in the relationship between lactoferrin domains. Denaturation of recombinant lactoferrin in milk was compared with denaturation in phosphate buffer, and results indicated that the protein was more heat-sensitive when treated in milk than in buffer. 相似文献

The influence of two organic amendments consisting of an urban waste compost (SUW) and a commercial amendment from olive mill wastes (OW) was assessed on the sorption properties and leaching of the ionizable herbicide imazaquin on four soils with different physicochemical characteristics. A loamy sand soil (CR), a loam soil (P44), a silt loam soil (AL), and a clay soil (TM), with low-medium organic matter contents, were chosen. Sorption-desorption experiments were performed on the original soils and on a mixture of these soils with the organic amendments at a rate of 6.25% (w/w). These mixtures were used just after preparation and after aging for 3 months. Imazaquin adsorption was higher on AL soil because of its high content of amorphous iron oxides, whereas it was related to the soils' organic matter (OM) contents on TM and CR soils and to acid pH on P44 soil. Addition of exogenous OM to soils caused a decrease in the adsorption of the herbicide with the only exception of CR soil, due to blocking of adsorptive surfaces and/or equilibrium pH rise. The extent of this decrease was dependent only on the nature of the added amendment on AL soil. The adsorbed amounts of imazaquin on aged organic fertilized soils were usually fairly close to that on original soils. Results of soil column experiments indicate that addition of exogenous organic matter cannot be considered as a regular practice for retarded movement of imazaquin. 相似文献

Purpose
This work analyzes polychlorinated biphenyl (PCB) and heavy metal contamination in fluvial sediments and soils in an urban catchment, according to the geo-accumulation index and to soil and sediment quality guidelines. The catchment is located in Coimbra, Portugal, being affected by frequent flooding, and its main stream is a tributary to one of the major rivers in Portugal (Mondego). Given the presence of industrial activities over time, some inputs of pollutants are expected, but so far, the legacy of historic pollution in this catchment has not yet been investigated.Materials and methods
Twenty-five samples were collected from nine sampling sites at the depths of 0–20, 20–40 and 40–60 cm (to provide a historic perspective) along longitudinal profiles (streamlines) and in soils downstream of pollution sources. These samples were analyzed for six heavy metals (Cu, Cr, Pb, Cd, Zn, Ni), organic carbon, pH and ten PCBs (IUPAC numbers 28, 30, 52, 101, 138, 153, 166, 180, 204, 209).Results and discussion
Total PCB concentrations ranged 0.47–5.3 ng g?1 dry weight (dw), and levels increase from the bottom to the top layers, suggesting an increased input over the last 100 years. PCB congener distribution shows the dominance of hexachlorobiphenyls, especially PCB138, suggesting the existence of local sources. PCB levels did not exceed sediment quality levels, placing sediments/soil under class 1 (not contaminated) or class 2 (trace contamination) with respect to PCB. All six metals exceeded the lowest effect level for sediment quality criteria, and three (Cd, Pb and Zn) largely exceeded the clean levels for dredged materials, placing sediments in class 5 (heavily contaminated). Sampling site S1 presented the highest concentrations of Zn, Pb and Cd, and historic vehicle traffic was identified as the most likely source, given the vertical and horizontal profiles.Conclusions
High levels of Pb, Cd and Zn were found in fluvial sediments at some locations of the Loreto catchment, likely from historic traffic sources. This urban area is frequently affected by flooding events and is currently being subject to urban redevelopment. During these events/actions, historic pollutants in the sediments might surface and be redistributed, impacting the downstream ecosystem of the major Mondego River or increasing the risk of exposure of the urban population.6.

BACKGROUND: Metribuzin is a widely used herbicide that has been identified as a groundwater contaminant. In this study, slow‐release formulations of metribuzin were designed by encapsulating the active ingredient in phosphatidylcholine (PC) vesicles and adsorbing the vesicles onto montmorillonite. RESULTS: The maximum active ingredient content in the slow‐release formulations was 246 g kg?1. Infrared spectroscopy results revealed that the hydrophobic interactions between metribuzin and the alkyl chains on PC were necessary for encapsulation. In addition, water bridges connecting the herbicide and the PC headgroup enhanced the solubility of metribuzin in PC. Adsorption experiments in soils were performed to evaluate the relationship between sorption and leaching. Funnel experiments in a sandy soil revealed that the herbicide was not irreversibly retained in the formulation matrix. In soil column experiments, PC–clay formulations enhanced herbicide accumulation and biological activity in the top soil layer relative to a commercial formulation. PC–clay formulations also reduced the dissipation of metribuzin by a factor of 1.6–2.5. CONCLUSIONS: A reduction in the recommended dose of metribuzin can be achieved by employing PC–clay formulations, which reduces the environmental risk associated with herbicide applications. The effect of beta-cyclodextrin (beta-CD) on the removal of the herbicide norflurazon (NFL) from soils has been investigated. The interaction of NFL with beta-CD in solution yielded the formation of a water-soluble inclusion complex at 1:1 stoichiometric ratio, which gave an increase in NFL solubility. Desorption studies of NFL previously adsorbed on six soils of different characteristics have been performed in the presence of 0.01 M beta-CD or 0.01 M Ca(NO(3))(2) as extractant solutions. Positive hysteresis was observed in all soils when 0.01 M Ca(NO(3))(2) solution was used, indicating that desorption of NFL from these soils was not completely reversible. On the contrary, the application of beta-CD solutions to soils where NFL had been previously adsorbed increased very much its desorption, and a negative hysteresis was obtained for all soils studied; that is, more NFL was desorbed with respect to NFL adsorption isotherm. A clear relationship was observed between the physicochemical characteristics of the soils and the beta-CD concentration necessary to remove the herbicide, the percentages of desorption observed for each soil being inversely related to the values obtained for the Freundlich sorption capacity parameter of the herbicide, K(f). In general, high desorption yields can be obtained with very low beta-CD concentrations, which is an important advantage from an economic point of view, although in those soils that present an extremely high NFL adsorption, higher amounts of beta-CD should be used. The results obtained indicate the high extracting power of beta-CD toward the herbicide previously adsorbed on the soils and the potential use of beta-CD for in situ remediation of pesticide-contaminated soils. 相似文献
