The lactic dehydrogenase and acetylcholinesterase enzymes of the boll weevil, Anthonomous grandis, have been shown to be inactivated by dye-sensitized photooxidation mediated by substituted xanthenes. The efficiency of the photooxidation reaction was correlated with the degree of halogenation of the dye molecule, the efficiency of the dye in singlet oxygen formation, and the strength of binding to lactic dehydrogenase. Changes in the in vivo levels of these enzymes due to ingestion of rose bengal by adult weevils are not further modified in the presence of light.  相似文献

Summary Surface additions of (15NH4)2SO4 were used to measure the immobilization and subsequent movement of exogenous N added to two litter types of contrasting quality (Cornus florida and Quercus prinus). Litterbaskets were used to measure the litter mass loss and N dynamics and to follow the movement of the 15N label through litter, F layer, and soil pools. Half of the litterbaskets of each species were treated with naphthalene to reduce microarthropod densities. The faster decomposing C. florida litter maintained a higher excess atom % 15N, and a greater relative concentration of the labeled input (g 15N g–1) than did Q. prinus litter. In both litter types the excess atom % 15N, relative concentration (g 15N g–1), and absolute amount of label recovered in the litter declined over time. This occurred during a period of net accumulation of total litter N, implying simultaneous release of the initial input and immobilization of N from other sources. The concentration of 15N in the soil increased over time, while the F layer apparently acted as an intermediary in the transfer of 15N from litter to soil. Naphthalene effectively reduced microarthropod numbers in all horizons of the litterbaskets and significantly reduced the decay rates of Q. prinus, but not C. florida litter. Naphthalene did not appear to affect total N dynamics in the litter. However, with all horizons taken together, the naphthalene-treated litterbaskets retained more total 15N than the control litterbaskets. Naphthalene also changed the vertical distribution of 15N within litterbaskets, so that the litter retained less of the 15N-labeled input and the F layer and soil horizons retained more of the labeled input than in control litterbaskets. Our major conclusions are: (1) the N pool of decomposing litter is dynamic, with simultaneous N release and immobilization activating N turnover even during the net accumulation phase; (2) litter quality is an important determinant of immobilization and retention of exogenous N inputs and, therefore, turnover of the litter N pool; and (3) microarthropod activity can significantly affect the incorporation and retention of exogenous N inputs in decomposing litter, although these changes are apparently not reflected in net N accumulation or release during the 1st year of decomposition. However, the naphthalene may have affected microbially mediated N dynamics and this possibility needs to be considered in interpreting the results.  相似文献

This study addressed differences between Diplocardia spp. (a native North American earthworm) and Octolasion tyrtaeum (an introduced European species), with respect to behavior, influence on soil microbial biomass, and plant uptake of N in tallgrass prairie soils. We manipulated earthworms in PVC-encased soil cores (20 cm diameter) over a 45-day period under field conditions. Treatments included: (1) control with no earthworms, (2) Diplocardia spp. only, and (3) O. tyrtaeum only. Prior to addition of earthworms, seedlings of Andropogon gerardii (a dominant tallgrass) were established in each core, and a dilute solution of 13C-labeled glucose and 15N-labeled (NH4)2SO4 was added to the soil to facilitate examination of earthworm/microbe/plant interactions. We found that Diplocardia spp. were significantly more active than O. tyrtaeum, and quickly assimilated 13C and 15N from the tracer. Individuals of Diplocardia spp. were present at shallower soil depths than O. tyrtaeum throughout the study. Contrary to expectation, this greater activity of Diplocardia spp. did not result in increased plant productivity. Rather, the activity of Diplocardia spp. was associated with less plant growth and smaller amounts of N acquired by A. gerardii seedlings compared to controls or O. tyrtaeum treatments. We observed few significant influences of earthworm treatments on microbial biomass C or N pool sizes, but the microbial C/N ratio was consistently greater in the presence of Diplocardia spp. relative to O. tyrtaeum. Results of this study indicate that activity of earthworms may enhance competition for N between microbes and plants during the growing season in tallgrass prairie.  相似文献

The responses of tallgrass prairie plant communities and ecosystem processes to fire and grazing are well characterized. However, responses of invertebrate consumer groups, and particularly soil-dwelling organisms, to these disturbances are not well known. At Konza Prairie Biological Station, we sampled soil macroinvertebrates in 1994 and 1999 as part of a long-term experiment designed to examine the effects and interactions of annual fire, mowing, and fertilization (N and P) on prairie soil communities and processes. For nearly all taxa, in both years, responses were characterized by significant treatment interactions, but some general patterns were evident. Introduced European earthworms (Aporrectodea spp. and Octolasion spp.) were most abundant in plots where fire was excluded, and the proportion of the total earthworm community consisting of introduced earthworms was greater in unburned, unmowed, and fertilized plots. Nymphs of two Cicada genera were collected (Cicadetta spp. and Tibicen spp.). Cicadetta nymphs were more abundant in burned plots, but mowing reduced their abundance. Tibicen nymphs were collected almost exclusively from unburned plots. Treatment effects on herbivorous beetle larvae (Scarabaeidae, Elateridae, and Curculionidae) were variable, but nutrient additions (N or P) usually resulted in greater densities, whereas mowing usually resulted in lower densities. Our results suggest that departures from historical disturbance regimes (i.e. frequent fire and grazing) may render soils more susceptible to increased numbers of European earthworms, and that interactions between fire, aboveground biomass removal, and vegetation responses affect the structure and composition of invertebrate communities in tallgrass prairie soils.  相似文献

Prescribed fires are likely to re-emit atmospherically deposited mercury (Hg), and comparison of soil Hg storage in areas affected by prescribed fire to that in similar unburned areas may provide cross-validating estimates of atmospheric Hg deposition. Prescribed fires are common in the southeastern United States (US), a region of relatively high Hg deposition compared to the rest of the US, and are thus a potentially significant source of re-emitted atmospheric Hg. Accordingly, Hg was determined in soil layers of a prescribed fire experiment in a Florida longleaf pine forest. The Hg deficit in the annually burned forest floor relative to the forest floor unburned for 46 years (0.180 g ha?1?yr?1) agreed to within 5% of an independent estimate of Hg deposition for this site based on a regional monitoring network and computer model (0.171 g ha?1 yr?1). Consideration of other potential inputs and outputs of Hg suggested that atmospheric deposition was the primary input of Hg to the site. If extrapolated, these results suggest that prescribed fires in the southeastern US mainly re-emit atmospherically deposited Hg and that this re-emission is less than 1% of total US anthropogenic emissions. However, emissions at other sites may vary due to the possible presence of Hg in underlying geological strata and differences in fire regime and levels of atmospheric Hg deposition.  相似文献

The soil actinomycete causing formation of nitrogen-fixing symbiotic nodules on roots of the woody angiosperm Comptonia peregrina (L.) Coult. (Myricaceae) has been isolated from surface-sterilized root nodules after incubation and enzyme maceration. The filamentous bacterium grows slowly in pure culture on a yeast extract medium, producing sporogenous bodies which form large numbers of ovoid spores. Reinfection of sand-grown or aeroponically grown seedlings of Comptonia was achieved repeatedly with inocula prepared from suspensions of the Comptonia isolate. The same actinomycete has been reisolated from these seedling nodules. The induced nodules are highly active in the acetylene-reduction assay, and plants grow vigorously without an exogenous supply of fixed nitrogen.  相似文献

Plant-derived carbon compounds enter soils in a number of forms; two of the most abundant being leaf litter and rhizodeposition. Our knowledge concerning the predominant controls on the cycling of leaf litter far outweighs that for rhizodeposition even though the constituents of rhizodeposits includes a cocktail of low molecular weight organic compounds which represent a rapidly cycling source of carbon, readily available to soil microbes. We determined the mineralization dynamics of a major rhizodeposit, glucose, and its relationship to land-use, microbial community and edaphic characteristics across a landscape in the southeastern United States. The landscape consists of cultivated, pasture, pine plantation, and hardwood forest sites (n = 3). Mineralization dynamics were resolved in both winter and summer using an in situ13C-glucose pulse-chase approach. Mineralization rates of the labeled glucose decline exponentially across the 72 h measurement periods. This pattern and absolute mineralization rates are consistent across seasons. An information-theoretic approach reveals that land-use is a moderately strong predictor of cumulative glucose mineralization. Measures assessing the size, activity, and/or composition of the microbial community were poor predictors of glucose mineralization. The strongest predictor of glucose mineralization was soil-extractable phosphorus. It was positively related to glucose mineralization across seasons and explained 60% and 48% of variation in cumulative glucose mineralization in the summer and winter, respectively. We discuss potential mechanisms underlying the relationship between soil phosphorus and glucose mineralization. Our results suggest that specific soil characteristics often related to land-use and/or land-management decisions may be strong predictors of glucose mineralization rates across a landscape. We emphasize the need for future research into the role of soil phosphorus availability and land-use history in determining soil organic carbon dynamics.  相似文献
