Monoclonal antibodies against hog cholera and bovine viral diarrhoea viruses were assayed on organ tissue sections of experimentally infected animals. The animals had been infected simultaneously with both viruses. The antibodies were tested using an indirect immunofluorescence test and an indirect enzyme immunoassay with a biotin/streptavidin/peroxidase detection system. A polyclonal hyperimmune serum was used as a control in direct immunofluorescence tests. Both techniques based on monoclonal antibodies were more sensitive and more specific than the conventional test, the enzyme immunoassay being more sensitive than the immunofluorescence test. Small amounts of BVD viral antigen were demonstrable with monoclonal antibodies in most organ tissues.  相似文献

In the Norddeich orphanage seals about four to five months of age were exposed intranasally and by contact to a phocine herpesvirus which originated from a young seal in this station. Virus excretion by the nasal route was observed in two animals on day 2 and 15 post inoculation, respectively. Production of neutralizing antibodies was detected one week after exposure. The antibody titre range was approximately tenfold higher in the inoculated seals than in animals of the contact group. The seals did not become seriously sick.  相似文献

Isolates of non-cytopathogenic bovine viral diarrhoea (BVD) virus from 18 persistently infected calves from one herd were compared by using monoclonal antibodies directed against the major viral glycoprotein gp53. All the isolates displayed an almost identical reaction pattern. Based on this antigenic analysis three cytopathogenic BVD and three non-cytopathogenic BVD viruses closely related to the non-cytopathogenic BVD herd isolate were selected. Six of the persistently infected calves were inoculated with a pool of the three closely related cytopathogenic BVD viruses and two with a pool of the three non-cytopathogenic BVD viruses. In addition three animals were infected with one closely related cytopathogenic BVD strain (Indiana) and two animals with the antigenetically different cytopathogenic BVD viral strain A1138/69. Regardless of the inoculation route all the animals superinfected with closely related cytopathogenic BVD viruses developed the characteristic lesions of mucosal disease within 14 days of infection. Animals which were inoculated with non-cytopathogenic BVD viruses which closely resembled the herd isolate, or with cytopathogenic BVD viruses which did not resemble the herd isolate did not develop any signs of disease. However, the latter group produced antibodies to the superinfecting virus.  相似文献

Eight harbour seals (Phoca vitulina), two of them seronegative, six seropositive against PDV and a seronegative grey seal (Halichoerus grypus) were exposed to a low doses of a cell culture-propagated phocine distemper virus isolate (PDV 2558/Han 88). An intranasal route of inoculation was chosen. Clinical signs, resembling those of 1988's seal disease and seroconversion were observed in both seronegative harbour seals. One of them succumbed to the infection. The virus was not transmitted to another susceptible harbour seal which served as in-contact animal. Virus could be recovered from leucocytes of the diseased seals. Viremia was also present in a seropositive harbour seal that developed mild clinical signs; other seropositive seals were protected from clinical disease. The grey seal showed seroconversion upon inoculation, but did not develop any signs of disease. The humoral immune response of the seals plainly discriminated between homologous (PDV) and heterologous (canine distemper virus, CDV) virus as shown by virus neutralization tests and an antibody-binding assay (PLA).  相似文献

Eight groups of altogether 25 goats without neutralizing antibodies against BVD virus, were inoculated either intranasally or intranasally and subcutaneously with two different BVD virus isolates during different stages of gestation. In all 18 goats inoculated within the first 78 days of gestation an abortion and foetal death rate of approximately 100% occurred. Only one goat gave birth to a clinically healthy kid. The other seven goats which were inoculated after the 78th day of gestation showed also a high foetal death rate. Only two of them gave birth to clinically healthy kids. Neutralizing antibodies against BVD virus could be detected in blood samples drawn from 14 kids born at normal term including stillborn and non-viable offsprings. BVD virus was reisolated from different organs taken from seven foetuses. It was not possible to isolate BVD virus from any of the normal offsprings.  相似文献

The characterization of runoff-related pesticide input and the identification of areas of concern in the field are essential for a comprehensive assessment of risk and the planning of mitigation measures. To this end, the agriculturally-derived aqueous and particle-associated pesticide contamination of the Lourens River and its tributaries was measured in a comprehensive design. Sampling was performed in the beginning of April prior to the first rainfall of the wet season and in the middle of April during high water conditions following the first rainfall of 9.6 mm d-1. Pre-runoff samples indicated contamination with endosulfan at levels up to 0.06 μg L-1, while no other pesticides were detectable. Rainfall-induced runoff resulted in an increase in the levels of the pesticides endosulfan, deltamethrin, azinphos-methyl, chlorpyrifos, and procymidone up to levels of 0.35, 1.4, 0.6, 0.19 and 9 μg L-1, respectively in water samples. Endosulfan, azinphos-methyl, and chlorpyrifos were detected at maximum concentrations of 273, 152, and 245 μg kg-1 in suspended sediments. No increased pesticide levels were detected at a control site upstream of the agricultural area. The Lourens River received its contamination via the tributaries discharging the surrounding farming area. Contamination of the six tributaries depended on landuse and slope characteristics and enabled the identification of target sites for risk reduction approaches. Transient pesticide levels exceeded the target water quality limit proposed by the South African Department of Water Affairs and Forestry (DWAF). The Lourens River site downstream of the farming area has been identified as a site where potential toxic conditions could arise.  相似文献
