Three slow-growing legume trees used for desert reforestation and urban gardening in the Sonoran Desert of Northwestern Mexico
and the Southwestern USA were evaluated whether their growth can be promoted by inoculation with plant growth-promoting bacteria
(Azospirillum brasilense and Bacillus pumilus), unidentified arbuscular mycorrhizal (AM) fungi (mainly Glomus sp.), and supplementation with common compost under regular screenhouse cultivation common to these trees in nurseries. Mesquite
amargo (Prosopis articulata) and yellow palo verde (Parkinsonia microphylla) had different positive responses to several of the parameters tested while blue palo verde (Parkinsonia florida) did not respond. Survival of all tree species was over 80% and survival of mesquite was almost 100% after 10 months of cultivation.
Inoculation with growth-promoting microorganisms induced significant effects on the leaf gas exchange of these trees, measured
as transpiration and diffusive resistance, when these trees were cultivated without water restrictions. 相似文献
Chocolate is often labeled with percent cocoa solids content. It is assumed that higher cocoa solids contents are indicative of higher polyphenol concentrations, which have potential health benefits. However, cocoa solids include polyphenol-free cocoa butter and polyphenol-rich nonfat cocoa solids (NFCS). In this study the strength of the relationship between NFCS content (estimated by theobromine as a proxy) and polyphenol content was tested in chocolate samples with labeled cocoa solids contents in the range of 20-100%, grouped as dark (n = 46), milk (n = 8), and those chocolates containing inclusions such as wafers or nuts (n = 15). The relationship was calculated with regard to both total polyphenol content and individual polyphenols. In dark chocolates, NFCS is linearly related to total polyphenols (r2 = 0.73). Total polyphenol content appears to be systematically slightly higher for milk chocolates than estimated by the dark chocolate model, whereas for chocolates containing other ingredients, the estimates fall close to or slightly below the model results. This shows that extra components such as milk, wafers, or nuts might influence the measurements of both theobromine and polyphenol contents. For each of the six main polyphenols (as well as their sum), the relationship with the estimated NFCS was much lower than for total polyphenols (r2 < 0.40), but these relationships were independent of the nature of the chocolate type, indicating that they might still have some predictive capabilities. 相似文献
Long-term potentiation (LTP) of synaptic strength, the most established cellular model of information storage in the brain, is expressed by an increase in the number of postsynaptic AMPA receptors. However, the source of AMPA receptors mobilized during LTP is unknown. We report that AMPA receptors are transported from recycling endosomes to the plasma membrane for LTP. Stimuli that triggered LTP promoted not only AMPA receptor insertion but also generalized recycling of cargo and membrane from endocytic compartments. Thus, recycling endosomes supply AMPA receptors for LTP and provide a mechanistic link between synaptic potentiation and membrane remodeling during synapse modification. 相似文献
Mammalian sex chromosomes have undergone profound changes since evolving from ancestral autosomes. By examining retroposed genes in the human and mouse genomes, we demonstrate that, during evolution, the mammalian X chromosome has generated and recruited a disproportionately high number of functional retroposed genes, whereas the autosomes experienced lower gene turnover. Most autosomal copies originating from X-linked genes exhibited testis-biased expression. Such export is incompatible with mutational bias and is likely driven by natural selection to attain male germline function. However, the excess recruitment is consistent with a combination of both natural selection and mutational bias. 相似文献
Manzamine A, a member of the manzamine alkaloids, was originally isolated from marine sponges of the genus Haliclona. It was recently shown to have activity against pancreatic cancer cells, but the precise mechanism of action remained unclear. To further our understanding of the mechanism of action of manzamine A, chemogenomic profiling in the yeast S. cerevisiae was performed, suggesting that manzamine A is an uncoupler of vacuolar ATPases. Fluorescence microscopy confirmed this effect on yeast vacuoles, where manzamine A produced a phenotype very similar to that of the established v-ATPase inhibitor bafilomycin A1. In pancreatic cancer cells, 10 µM manzamine A affected vacuolar ATPase activity and significantly increased the level of autophagosome marker LC3-II and p62/SQSTM1 as observed by western blot analysis. Treatment with manzamine A in combination with bafilomycin A1 (inhibitor of autophagosome-lysosome fusion) did not change the levels of LC3-II when compared to cells treated with bafilomycin A1 alone, suggesting that manzamine A is a potential inhibitor of autophagy by preventing autophagosome turnover. As autophagy is essential for pancreatic tumor growth, blocking this pathway with manzamine A suggests a promising strategy for the treatment of pancreatic cancer. 相似文献
Timber use in central Europe is expected to increase in the future, in line with forest policy goals to strengthen local wood supply for CO2-neutral energy production, construction and other uses. Growing stocks in low-elevation forests in Switzerland are currently high as exemplified by the Swiss canton of Aargau, for which an average volume of 346 ± 16 m3 ha−1 was measured in the 3rd Swiss National forest inventory (NFI) in 2004–2006. While this may justify a reduction of growing stocks through increased timber harvesting, we asked whether such a strategy may conflict with the sustainability of timber production and conservation goals. We evaluated a range of operationally relevant forest management scenarios that varied with respect to rotation length, growing stock targets and the promotion of conifers in the regeneration. The scenarios aimed at increased production of softwood, energy wood, the retention of potential habitat trees (PHTs) and the conversion to a continuous cover management system. They were used to drive the inventory-based forest simulator MASSIMO for 100 years starting in 2007 using the NFI sampling plots in Aargau. We analyzed model outputs with respect to projected future growing stock, growth, timber and energy yield and harvesting costs. We found growing stock to drop to 192 m3 ha−1 in 2106 if business-as-usual (BAU as observed between the 2nd and 3rd NFI) timber volumes were set as harvesting targets for the whole simulation period. The promotion of conifers and a reduction of rotation lengths in a softwood scenario yielded 25% more timber over the whole simulation period than BAU. An energy wood scenario that reduced growing stock to 200 m3 ha−1 by 2056 and promoted the natural broadleaved regeneration yielded 9% more timber than BAU before 2056 and 30% less thereafter due to decreasing increments. The softwood scenario resulted in higher energy yield than the energy wood scenario despite the lower energy content of softwood. Retaining PHT resulted in a reduction of timber harvest (0.055 m3 ha−1 yr−1 per habitat tree) and higher harvesting costs. Continuous cover management yielded moderate timber amounts throughout the simulation period, yet sustainably. Considering climate change, we discuss the risks associated with favoring drought- and disturbance-susceptible conifers at low elevations and emphasize that continuous cover management must allow for the regeneration of drought-adapted tree species. In conclusion, our simulations show potential for short-term increases in timber mobilization but also that such increases need to be carefully balanced with future forest productivity and other forest ecosystem services.
‘Blanquilla’ pears processed as fresh-cut products are highly sensitive to browning and softening. Common postharvest methods, such as the use of antibrowning compounds and/or modified atmosphere packaging, fail to preserve ‘Blanquilla’ pear slices long enough to be marketable. However, treatment with 1-MCP before cutting and peeling considerably improved their textural properties (9.2 N vs. 1.1 N with and without 1-MCP treatment, respectively) and color (a* values of 1 vs. 5 after 15 d at 4 °C, for slices pear treated with 1-MCP and without treatment, respectively). These positive changes were closely related to a decrease in respiratory activity determined on whole pears after 3 months of storage in air at 0 ± 1 °C and 95% R.H. (0.40 ± 0.05 mmol CO2 kg−1 h−1vs. 0.77 ± 0.04 mmol CO2 kg−1 h−1 with and without 1-MCP treatment, respectively) and ethylene production (1.18 ± 0.36 nmol C2H4 kg−1 h−1vs. 5.751 ± 1.12 nmol C2H4 kg−1 h−1 for samples treated with and without 1-MCP, respectively). The use of 1-MCP allows fresh-cut ‘Blanquilla’ pears to be sold up to about 5 d after processing. Treatment with 1-MCP could be a viable alternative to common technologies for extending the shelf-life of ‘Blanquilla’ pears as a fresh-cut product. 相似文献
Flavobacterium psychrophilum is an important fish pathogen, responsible for Cold Water Disease, with a significant economic impact on salmonid farms worldwide. In spite of this, little is known about the bacterial physiology and pathogenesis mechanisms, maybe because it is difficult to manipulate, being considered a fastidious microorganism. Mutants obtained using a Tn4351 transposon were screened in order to identify those with alteration in colony morphology, colony spreading and extracellular proteolytic activity, amongst other phenotypes. A F. psychrophilum mutant lacking gliding motility showed interruption of the FP1638 locus that encodes a putative type-2 glycosyltransferase (from here on referred to as fpgA gene, Flavobacterium psychrophilum glycosyltransferase). Additionally, the mutant also showed a decrease in the extracellular proteolytic activity as a consequence of down regulation in the fpgA mutant background of the fpp2-fpp1 operon promoter, responsible for the major extracellular proteolytic activity of the bacterium. The protein glycosylation profile of the parental strain showed the presence of a 22 kDa glycosylated protein which is lost in the mutant. Complementation with the fpgA gene led to the recovery of the wild-type phenotype. LD50 experiments in the rainbow trout infection model show that the mutant was highly attenuated. The pleiotropic phenotype of the mutant demonstrated the importance of this glycosyltranferase in the physiology and virulence of the bacterium. Moreover, the fpgA mutant strain could be considered a good candidate for the design of an attenuated vaccine. 相似文献
