Reproductive, metabolic, and endocrine responses to feed restriction and GnRH treatment in primiparous, lactating sows

The current experiment was carried out to determine whether exogenous GnRH treatment in primiparous, lactating sows undergoing feed restriction would improve reproductive performance after weaning. Sows were allocated to one of three treatments: AA sows (n = 8) were fed to appetite throughout a 28-d lactation, AR (n = 12) and AR + GnRH (n = 12) sows were fed as AA sows from farrowing to d 21 of lactation, and feed intake was reduced to 50% of the ad libitum intakes from d 22 to 28. The AR + GnRH sows received 800 ng of GnRH i.v. every 6 h from d 22 to 28 of lactation, and AA and AR sows received saline. Sow weight, backfat, and litter weight were recorded weekly. Within 2 d after farrowing, litter size was standardized to 8 to 10. At d 17 of lactation, an indwelling jugular catheter was surgically implanted in each sow. Blood samples were taken for characterization of plasma LH, FSH, insulin, IGF-I, and leptin by RIA at d 21 and before and after weaning on d 28 of lactation. After weaning, all sows were given ad libitum access to feed, checked for onset of standing estrus twice daily with mature vasectomized boars, and inseminated 12 and 24 h after onset of standing estrus with pooled semen from the same fertile boars (3 x 10(9) sperm/AI). After breeding, feed allowance was reduced to NRC (1988) requirements for gestation. At d 28 +/- 3 of gestation, sows were killed and ovulation rate and embryo survival were determined. Restricted sows lost more weight during lactation than AA sows (P < .02). During the period of feed restriction, plasma IGF-I and postprandial insulin and leptin in AR and AR + GnRH sows, and LH pulse frequency in AR sows, were lower than those in AA sows (P < .04). Associations (P < .004) between plasma insulin and leptin and between leptin and mean LH concentrations were established. The LH pulse frequency in AR + GnRH sows did not differ from that in AA sows before weaning. After weaning, maximum, mean, and minimum LH concentrations in the AA and AR sows, and FSH concentrations in AR sows, increased (P < .05) in response to weaning. Paradoxically, GnRH treatment in lactation seemed to suppress the expected LH and FSH responses to weaning. Ovulation rate and embryo survival were not different among the three groups. In conclusion, although exogenous GnRH therapy restored LH secretion in feed-restricted sows, it did not improve overall reproductive performance.     

Single agent gemcitabine chemotherapy in dogs with spontaneously occurring lymphoma

BACKGROUND: Gemcitabine has been shown to be effective as a single agent in a variety of tumors including nonHodgkin's lymphoma. Its use in veterinary medicine has been limited and to date this drug has not been used as a first-line therapy in dogs with lymphoma. HYPOTHESIS: Gemcitabine as a single agent may be efficacious in dogs presented for the first time with lymphoma. ANIMALS: Twenty-four dogs with spontaneously occurring lymphoma. METHODS: All dogs were clinically staged and given gemcitabine at 400 mg/m(2) over a 30-minute intravenous infusion weekly for 3 weeks and then given 1 week off treatment before starting a second cycle. RESULTS: A single dose of gemcitabine lowered both neutrophil count (decrease in mean neutrophil count from 10,640 cells/ microL to 3,140 cells/microL) and platelet count (decrease in mean platelet count from 201,290 cells/microL to 139,190 cells/microL) 7 days after administration. Consequently gemcitabine dosage was reduced at the second treatment in 8 of 21 dogs or a dose delay of 1-7 days and a reduction of dosage was used in 7 of 21 dogs. Seven dogs completed the assigned 4-week cycle. Two of these dogs had progressive disease and 5 had stable disease. No objective responses were seen in dogs treated with a second cycle of gemcitabine. CONCLUSIONS AND CLINICAL IMPORTANCE: Gemcitabine administration as a single agent resulted in hematologic toxicity and did not reduce lymphoma burden. If gemcitabine is to be used in veterinary medicine, additional prospective pharmacologic studies should be done to determine the appropriate dosage, regimen, and schedule of use before it can be recommended for use in the treatment of dogs with lymphoma as a single agent.     

The status of the freshwater pearl mussel Margaritifera margaritifera Linn. in Scotland

1. One hundred and sixty rivers in Scotland with historical records of freshwater pearl mussel Margaritifera margaritifera were surveyed between 1996 and 1999 for the presence of the species. 2. M. margaritifera populations were classed as either ‘extinct’ (no mussels remaining), ‘not currently viable’ (ranging from only dead shells present to no juveniles present, regardless of the number of adults found), or ‘functional’ (at least one juvenile was found, regardless of the overall numbers of adults present). 3. M. margaritifera populations could be classed as ‘functional’ in only 52 rivers (34% of total surveyed) and in only 17 of these were juveniles below 20 mm (5 yrs old) present. Furthermore, in only 10 of these functional rivers were mussels also still considered either to be common or abundant and these included both large easterly‐flowing rivers and small western rivers and streams. 4. In approximately two‐thirds (101) of the 155 rivers occupied 100 years ago, M. margaritifera is now extinct or is about to become extinct. Furthermore, analysis indicates that the rate of mussel population extinction has accelerated since 1970, with a recent average of two mussel river extinctions per year. 5. The predominant threat to the mussel populations has undoubtedly been pearl fishing. This has been occurring at every river, even in the most remote northwest areas, where most of the ‘functional’ populations remain. Recent legislation has provided full protection to M. margaritifera, so that all pearl harvesting is now illegal. 6. Locally, river engineering and host salmonid stock decline poses a serious threat and eutrophication has already eliminated populations in southern and eastern Scotland. 7. In every part of its global range, M. margaritifera has declined substantially and is now either threatened with extinction or is highly vulnerable. Based on recent estimates from across the species range, it appears that Scotland now probably holds at least half of the world's remaining functional M. margaritifera populations. Thirty thousand years of human colonization in tasmania: new pleistocene dates

Basal dates of 30,420 years before present (BP) from a limestone cave in the Florentine River valley and 30,840 BP from a sandstone rockshelter in the Shannon River valley on the edge of the central Tasmanian highlands indicate colonization of Tasmania 8,000 years earlier than previously thought. These data indicate that people arrived before the Bassian Bridge was exposed about 23,000 years ago and support evidence that Tasmania and Australia may have been connected intermittently during the past 50,000 years. The dates support earlier suggestions that the Tasmanian inland was an important focus for systematic occupation and exploitation by human groups and dispel a belief that the Aboriginal economy 30,000 years ago was based on littoral, lacustral, and riverine resources. The absence of megafauna at both sites points to their extinction by 30,000 years ago in Tasmania. The people inhabiting Tasmania at this time, together with those at Monte Verde in Chile, were the most southerly humans on Earth.     

An outbreak of Equid herpesvirus abortion in New South Wales.

Thirty-three of the 44 mares on a Thoroughbred stud in New South Wales aborted or lost foals within one day of birth. Gross pathological and histological changes were in keeping with Equid herpesvirus I (EHV-1) abortion. In the six foals that underwent virological examination, EHV was isolated and typed as EHV-1 by restriction endonuclease analysis. EHV-1 abortion had not occurred previously on this stud and the source of the infection was not identified.
