Identification of an H1N1 subtype of swine influenza virus and serological analysis

To investigate the epizootic of swine influenza virus(SIV), 60 nasal swabs were collected from a clinical cases of pig farm in Tai'an City, Shandong Province of China in April 2017. SIV was isolated by inoculating into 10-day-old Special Pathogen Free embryonated eggs and the whole genome was sequenced. An H1N1 subtype SIV was isolated and designated as A/swine/Shandong/TA04/2017(H1N1). Phylogenetic analysis showed that apart from the polymerase A(PA) fragment belonging to the 2009 pandemic H1N1 branch, seven genome segments belonged to avian-like H1N1 influenza virus lineage. The cleavage site sequence of the hemagglutinin(HA) protein was PSIQSR↓G, which is a typical molecular biological characteristic. Five potential N-glycosylation sites(N14, N26, N277, N484 and N543) were found in the HA gene. To further investigate the epidemiology of SIV in this farm, the 995 serum samples were assessed with EAH1N1 2009 pandemic H1N1 and H3 N2 antigens. The results showed that the total positive rate was 65.43%. The positive rates of single virus infection detected by EAH1N1, 2009 pdmH1N1 and H3 N2 for serum HI(Hemagglutination inhibition) were 48.35, 30.85 and 7.47%, respectively. The results showed that SIV in Shandong Province has been reassorted in some segments and the SIV-positive rate was high on the SIV outbreak farm. These data provide evidence of an epizootic of SIV.     

棉花不同时期去叶源对库源关系影响的初步研究

通过对棉花不同时期去主茎叶、叶枝叶、果枝叶和全部叶片的研究,初步探讨了棉花叶源对库的影响.结果表明:无论蕾期还是盛铃期,叶片是棉花光合产物的主要来源,没有叶片棉花就不能正常生长,也很难保证其产量;不同部位的叶片中,主茎叶的作用显著,蕾期去主茎叶后株高日增长量减少0.5 cm以上,干物质积累明显下降,盛铃期则导致棉铃大量脱落,成铃平均减少2个以上;去果枝叶的作用相对较小,去叶枝叶几乎无影响.     

苜蓿褐斑病抗性与几种同工酶的关系

以苜蓿5个品种在褐斑病抗性上存在差异的抗、感单株为材料，研究了接种病菌后苜蓿褐斑病抗性与多酚氧化酶(PPO)、过氧化物酶(POD)和超氧化物歧化酶(SOD)及其酶活性的关系。研究结果表明，褐斑病侵染后，苜蓿抗病株系和感病株系叶片中PPO、POD和SOD同工酶谱带多少和强弱存在明显的差异，即大部分抗病株系PPO、POD和SOD酶活性高于感病株系，并有新的同工酶带出现。PPO、POD和SOD在苜蓿抗褐斑病上可能起到重要作用，其同工酶谱带和酶活性的差异有可能做为鉴定苜蓿抗褐斑病的一种同工酶标记。     

库存本阅览室与外借库的合并管理

我馆在2005年将中文图书库存本阅览室与中文图书外借库合并成中文书库,进行统一管理。本文结合工作实际,分析了书库合并前分开管理的弊端,阐述了二者合并的意义,提出了合并后的具体管理措施。     

Influence of all-trans retinoic acid on proliferation and differentiation of neural stem cells from new born rat striatum

AIM: To study the effect and mechanism of all-trans retinoic acid (atRA) on neural stem cell (NSCs) proliferation and differentiation from new born Sprague-Dawley rat striatum. METHODS: NSCs were isolated from the brains of new born Sprague-Dawley rat striatum, and the features of cells were characterized by immunofluorescence staining. The effects of different culture medium on cell cycle distribution and proliferation of NSCs were determined by flow cytometry (FCM). The effects of atRA on differentiation of NSCs were determined by immunofluorescence staining and classified count of differentiated cells. RESULTS: FCM assay indicated that atRA inhibited the proliferation of NSCs. The percentage of cells in G0/G1 phase in atRA treatment group was significantly higher than that in control, and the proliferation index (PI) was significantly low. The percentage of neurons differentiated from NSCs in atRA group was 2.5 times of the control group after induced by adding 10% FCS in culture medium. CONCLUSION: atRA counteracts the effects of bFGF on the promotion of mitosis and inhibition of differentiation of NSCs. atRA also promotes NSCs to differentiate into neurons in vitro.     

应用热脉冲技术对小美旱杨耗水量的研究

应用热脉冲对河套灌区农田防护小美旱杨（Populus popular’s）日蒸腾耗水量进行了研究。结果表明：在正常生长状态下，小美旱杨的日蒸腾耗水量变化曲线呈宽峰状，随着林龄增加，日蒸腾耗水量逐渐增加，在不同水分条件下,灌水时日蒸腾耗水量：萌生苗日蒸腾耗水量大于扦插日蒸腾耗水量。

中国矮马运铁蛋白遗传多态性的初步研究

采用聚丙烯酰胺凝胶电泳对中国矮马运铁蛋白多态性进行了测定。在运铁蛋白位点共发现5种表型，即F2F2，DF2，F2H2，F2O和F2R，由Tf^D,Tf^F2,Tf^F2,Tf^H2,Tf^O和Tf^R等5个等位基因控制，其基因频率分别为0.0312,0.7188,0.0625,0.0312和0.1563；基因的杂合度（H），个体鉴别概率（Dp)和亲仔关系排除概率（PE）分别为0.4530,0.6875和0.2586。     

猪生殖与呼吸综合征病毒M基因和N基因的表达

将猪生殖与呼吸综合征病毒（PRRSV）的M基因和N基因从重组质粒pMD18-T—M—N中亚克隆至pBV220原核表达栽体上，成功构建了重组表达质粒pBVM—N。将pBVM—N转化大肠埃希氏菌DH5α感受态细胞．重组菌经温度敏感诱导表达，其细菌裂解物经SDS—PAGE可检测到分子质量约为19ku的目的蛋白，与M基因表达产物一致；经蛋白质分析软件Bandscan分析，其表达量可达19．1％；Western-blotting分析表明，该重组蛋白可被兔抗PRRSV血清所识别，与预期的M基因表达产物相一致，而N基因未获表达。     

Study on integrin-mediated albumin microbubbles adherence to activated leukocytes

AIM:To investigate the mechanism responsible for albumin microbubbles adherence to activated leukocytes. METHODS: In vitro studies were performed in which activated or nonactivated leukocytes were incubated with albumin microbubbles and observed under microscopy. The suspensions of leukocytes and microbubbles which contained or absented of integrins were analyzed with flow cytometry.RESULTS: A minimum of 50cells were identified under transillumination. 5 min after microbubbles were incubated with leukocytes, the number of cells interacting with microbubbles was greater for activated cells than for nonactivated cells(20.30±2.67 vs 4.50±1.43, P <0.01).Microbubbles attached to the surface of activated leukocytes were phagocytosed and remained intact for up to 30min. Microbubble attachment was inhibited notably by blocking the leukocyte β₂-integrin Mac-1(P <0.01) and by VLA-4mAb slightly(P <0.05) CONCLUSION: The mechanism of albumin microbubbles attaching to and phagocytosed by leukocytes was due to β₂-integrin and VLA-4 mediation. Phagocytosed microbubbles can remain at the regions of inflammation for15 min, also responsible to ultrasound.
