Lysosomal storage diseases (LSDs) are a group of heterogeneous disorders caused by defects in lysosomal enzymes or transporters, resulting in accumulation of undegraded macromolecules or metabolites. Macrophage numbers are expanded in several LSDs, leading to histiocytosis of unknown pathophysiology. Here, we found that mice lacking the equilibrative nucleoside transporter 3 (ENT3) developed a spontaneous and progressive macrophage-dominated histiocytosis. In the absence of ENT3, defective apoptotic cell clearance led to lysosomal nucleoside buildup, elevated intralysosomal pH, and altered macrophage function. The macrophage accumulation was partly due to increased macrophage colony-stimulating factor and receptor expression and signaling secondary to the lysosomal defects. These studies suggest a cellular and molecular basis for the development of histiocytosis in several human syndromes associated with ENT3 mutations and potentially other LSDs.  相似文献

Summary Field and pot experiments to determine whether the tuber size of volunteer potatoes affected their susceptibility to foliar applications of metoxuron are reported. The reduction in haulm weight of plants from small tubers (2–3 cm diameter) was greater than that of plants from large ones (4–5 cm diameter). This greater control was related to the lower vigour at spraying of the plants from the small tubers. It is suggested that these results could explain the variability in the level of control of volunteer potatoes achieved by metoxuron in carrot crops.  相似文献

The ‘within-season’ module of the Weed Manager decision support system (DSS) predicts the effect of twelve UK arable weeds on winter wheat yields and profitability. The model and decision algorithm that underpin the DSS are described and their performance discussed. The model comprises: (i) seedling germination and emergence, (ii) early growth, (iii) phenological development, (iv) herbicide and cultivation effects and (v) crop yield loss. Crop and weed emergence are predicted from the timing and method of cultivation, species biology, and the weather. Wheat and weeds compete for resources, and yield losses are predicted from their relative leaf area at canopy closure. Herbicides and cultural control methods reduce weed green area index, improving crop yield. A decision algorithm identifies economically successful weed management strategies based on model output. The output of the Weed Manager model and decision algorithm was extensively validated by experts, who confirmed the predicted responses to herbicide application were sufficiently accurate for practical use. Limited independent data were also used in the validation. The development of the module required integrating novel and existing approaches for simulating weed seedling establishment, plant development and decision algorithm design. Combining these within Weed Manager created a framework suitable for commercial use.  相似文献

Prolonged imbibition under conditions of water stress or oxygen deficiency can lead to the induction of secondary dormancy in rape seeds. During imbibition in darkness seeds develop light sensitivity. The percentage of seeds not germinating in the dark depends on various factors prevailing during and after the stress treatment, as well as on the intrinsic susceptibility of the seeds to these factors.

In experiments on the effect of water stress it was found that the percentage of secondary dormant seeds could be increased in the range of 0–75%, by greater water suction and duration of imbibition of the seeds. Additionally, there were indications that increasing treatment temperature resulted in increasing percentages of dormant seeds. However, fewer secondary dormant seeds were produced when treatment temperature and test temperature differed greatly from each other.

Experiments on the effect of oxygen deficiency also revealed a positive effect of treatment duration. The general degree of dormancy induction, however, in this series of experiments was low. At 20 °C almost no dormancy induction was observed, whilst at 12 °C there were between 0 and 30% dormant seeds.

Genotypic differences previously assessed in model experiments with buried seeds were confirmed in both studies. Oilseed rape cultivars Bienvenu and Liglandor were shown to be able to build up high percentages of secondary dormant seeds, whereas cultivars Jet Neuf and Falcon had a rather low susceptibility towards the dormancy imposing factors tested. Cultivar Rubin was intermediate in response. There was often much variation between experiments with the same experimental design. The reasons for this are unclear.  相似文献

Persistence of oilseed rapeseed in soil can result in weed problems but also reduce oil quality of following rape crops or result in unwanted gene escape which is particularly relevant in the context of genetically modified oilseed rape. In this paper data from 13 field experiments at sites in England, Austria and Germany are presented where tillage operations were tested that potentially reduce the build-up of a seed bank. In the majority of experiments seed losses were artificially simulated by broadcasting ca. 10,000 freshly ripened rapeseed m−2 onto cereal stubbles. Oilseed rapeseedlings in autumn, the seed bank in winter–spring and yields of the following crop winter wheat were assessed as a function of tillage regime. During summer and autumn 19–70% of the seeds germinated and emerged. This part of the population was killed by following tillage operations or herbicide applications. However, 0–29%, in moist years 0–5%, of the initially broadcasted seeds developed dormancy and remained ungerminated in the soil until the following winter–spring.

Delaying incorporation of the seeds by leaving the stubble untouched for up to 4 weeks resulted in a reduced seed bank in almost every case. Also, repeated stubble tillage compared to an early single stubble tillage operation resulted in a smaller seed bank. The type of primary tillage (ploughing versus non-inversion cultivation) had no clear effect. No relation was found between the number of seedlings in autumn and the size of the seed bank the following winter–spring. Grain yield of the following crop winter wheat was not adversely affected by delayed stubble tillage.

The results indicate that stubble tillage aiming at a reduced seed bank of oilseed rape should focus on conditions avoiding induction of secondary dormancy rather than improving germination conditions. This means that, under the climatic conditions of central and western Europe, the stubble should be left untouched for several weeks after harvest before starting the usual tillage sequence with stubble tillage and ploughing or a non-inversion tillage sequence.  相似文献

Summary Two experiments have investigated the persistence of 16 arable, annual broad-leaved weed species over 6 years in a silty loam and a clay soil. Small plastic beads were included as an 'inert' comparison. Seeds were broadcast in October on to plots at the start of the experiment, and these were either tine cultivated or ploughed annually thereafter. Plots were sown with either spring or winter wheat. As far as possible, weed seeding was prevented each year. For some species, the seed decline appeared to be slower on the ploughed plots than on the tined plots and in the winter wheat compared with the spring wheat. Seed decline also tended to be slower on the clay soil at Rothamsted than on the silty loam at Long Ashton. Some species declined rapidly (e.g. Brassica napus , Chrysanthemum segetum , Galium aparine , Galeopsis tetrahit ), with annual decline rates in excess of 58%, whereas others declined very little (e.g. Papaver rhoeas decline rate 9%). Most of the other species had decline rates between 20% and 40%. The results are discussed in relation to the development of population dynamics models to predict long-term consequences of alternative weed management strategies.  相似文献

Chlorpropham, propyzamide and trifluralin were assessed in several trials, to test their potential for the control of groundkeeper potatoes (Solanum tuberosum L.). Chlorpropham was insufficiently active, but trifluralin and propyzamide at doses of 4 kg a.i./ha, or more, reduced and delayed the emergence of potato plants in the summer, after autumn/winter or spring applications. The highest dose (12 kg a.i./ha) prevented many tubers producing plants. Greatest activity was attained when the herbicides completely surrounded the tubers when active growth started in the spring. However, both residue and crop tolerance data indicated that the doses of trifluralin and propyzamide required to control the potatoes were so high that appreciable residues remained in the following summer. These damaged vegetable crops drilled in April and May, and affected cereals planted 12 months after application. Thus there is insufficient margin of selectivity between the doses required to control the potatoes and those tolerated by subsequent crops.  相似文献

This study reviews 52 field experiments, mostly from the UK, studying the effects of cultivation techniques, sowing date, crop density and cultivar choice on Alopecurus myosuroides infestations in cereal crops. Where possible, a statistical meta‐analysis has been used to calculate average responses to the various cultural practices and to estimate their variability. In 25 experiments, mouldboard ploughing prior to sowing winter cereals reduced A. myosuroides populations by an average of 69%, compared with non‐inversion tillage. Delaying drilling from September to the end of October decreased weed plant densities by approximately 50%. Sowing wheat in spring achieved an 88% reduction in A. myosuroides plant densities compared with autumn sowing. Increasing winter wheat crop density above 100 plants m?2 had no effect on weed plant numbers, but reduced the number of heads m?2 by 15% for every additional increase in 100 crop plants, up to the highest density tested (350 wheat plants m?2). Choosing more competitive cultivars could decrease A. myosuroides heads m?2 by 22%. With all cultural practices, outcomes were highly variable and effects inconsistent. Farmers are more likely to adopt cultural measures and so reduce their reliance on herbicides, if there were better predictions of likely outcomes at the individual field level.  相似文献
