The objectives of the present study were to quantify the effects of a biological chronic stressor (lameness) on the duration and frequency of different oestrous behaviours in parallel with milk hormone profiles. Dairy cows 51.8 ± 1.4 days postpartum (n = 59), including 18 non‐lame control cows, were scored for lameness and closely observed for signs of oestrus having had their follicular phases synchronized by administration of gonadotrophin‐releasing‐hormone (GnRH) followed by prostaglandin F (PG) 7 days later. Lameness shortened the period when herd‐mates attempted to mount the lame cows (1.83 ± 0.69 h vs 5.20 ± 1.53 h; p = 0.042) but did not affect the overall duration of total behaviours (lame 12.3 ± 1.3 h vs non‐lame 15.2 ± 1.3 h). Lameness also lowered the intensity of oestrus [1417 ± 206 points (n = 18) vs 2260 ± 307 points (n = 15); p = 0.029]. Throughout the synchronized oestrous period, lame cows mounted the rear of herd‐mates less frequently (p = 0.020) and tended to chin rest less (p = 0.075). Around the period of maximum oestrous intensity, lameness also diminished the proportion of cows mounting the rear of another cow and chin resting (p = 0.048, p = 0.037, respectively). Furthermore, lame cows had lower progesterone values during the 6 days before oestrous (p ≤ 0.05). Fewer lame cows were observed in oestrus following PG (non‐lame 83%, lame 53%; p = 0.030); however, if prior progesterone concentrations were elevated, lame cows were just as likely to be observed in oestrus. In conclusion, following endogenous progesterone exposure, lameness shortens the period when herd‐mates attempt to mount lame cows but does not affect the incidence of oestrous. However, lame cows are mounted less frequently and express oestrus of lower intensity. This is associated with lower progesterone prior to oestrus but not with abnormal oestradiol or cortisol profiles in daily milk samples.  相似文献

Experiments were conducted to determine if diurnal temperature variation could be used to prime cicer milkvetch ( Astragalus cicer L.) seed and thereby shorten the time for seed germination. For untreated cicer miikvetch seed at constant 20°C, 50% of the seeds germinated in 8.2 days and 90% of the seeds germinated in 10.5 days. In contrast, seed that was first primed for 7 days with a diurnal temperature variation (35 (8h)/5 (16h) °C) had values of 1.9 days and 3.3 days, respectively. A second experiment evaluated priming of seed in bulk to test if a procedure might be developed that could be scaled up to prime commercial quantities of seed. A bulk sample of seed was soaked in water for 5h at room temperature and then kept in a high humidity atmosphere during 7 days of priming at 35/5°C. Hydrated seed from the bulk sample had 50% germination in 2.7 days and 90% germination in 5.3 days. When the bulk sample was dried for 8 days at room temperature, the benefit of priming on shortening germination time was retained. It is likely that diurnal temperature priming could be adapted to large volumes of seed.  相似文献

Accelerator mass spectrometer dating of an assemblage of fibrous and leather footwear from Arnold Research Cave in central Missouri documents a long sequence of shoe construction by prehistoric Midwestern peoples, beginning perhaps as early as 8300 calendar years before the present (cal years B.P.). An earlier fibrous sandal form dates from 8325 to 7675 cal years B.P., and later fibrous or leather slip-ons span the period from 5575 to 1070 cal years B.P. The assemblage adds to a growing picture of the highly varied nature of prehistoric footwear production in the United States throughout the Holocene.  相似文献

The anomalous scattering data from five Cd in the native protein were used to determine the crystal structure of cadmium, zinc (Cd,Zn) metallothionein isoform II from rat liver. The structure of a 4-Cd cluster was solved by direct methods. A 2.3 A resolution electron density map was calculated by iterative single-wavelength anomalous scattering. The structure is folded into two domains. The amino terminal domain (beta) of residues 1 to 29 enfolds a three-metal cluster of one Cd and two Zn atoms coordinated by six terminal cysteine thiolate ligands and three bridging cysteine thiolates. The carboxyl terminal domain (alpha) of residues 30 to 61 enfolds a 4-Cd cluster coordinated by six terminal and five bridging cysteine thiolates. All seven metal sites have tetrahedral coordination geometry. The domains are roughly spherical, and the diameter is 15 to 20 A; there is limited contact between domains. The folding of alpha and beta is topologically similar but with opposite chirality. Redundant, short cysteine-containing sequences have similar roles in cluster formation in both alpha and beta.  相似文献

The clinical, gross and histopathological findings in 50 sheep affected with Johne's disease are described. Clinically 90% were emaciated and 20% showed severe diarrhoea. On necropsy there was thickening of the walls of the intestines, particularly of the ileum, caecum and less frequently the jejunum, but in 36% of sheep the changes were only mild. Histologically there was a granulomatous enteritis, typhlitis and colitis, with the most severe changes in the terminal ileum. High numbers of acid-fast organisms were present in the terminal ileum in over 70% of sheep. Mycobacterium paratuberculosis was cultured from only 8% of the sheep examined.  相似文献
