Aciculosporium take (Ascomycota; Clavicipitaceae) is a causal agent of witches' broom of
bamboo plants. The symptoms of this disease are believed to be induced by plant
hormones, particularly auxins. Indole-3-acetic acid (IAA) was identified in
cultures of this fungus in an l-tryptophan-supplemented liquid medium.
IAA production was confirmed on 30 isolates of A. take from various
hosts and locations at levels up to 1 mg/l. The biosynthetic pathway of
IAA in A. take culture was examined by analyzing intermediate products
and by feeding experiments. The results showed that the indole-3-pyruvic acid
pathway (l-tryptophan → indole-3-pyruvic acid → indole
acetaldehyde → IAA) was the dominant pathway in A. take.

A competitive enzyme-linked immunosorbent assay (C-ELISA) using neutralizing monoclonal antibodies (MAbs) against Akabane virus (AKAV) was developed to detect antibodies to AKAV in cattle sera. The performance of the test using 7 different competitor MAbs was evaluated in sequential serum samples and sera from cattle infected with various bovine arboviruses. The dynamics of the antibody response expressed by percentage of inhibition (PI) in C-ELISA coincided with those of neutralizing antibody titers in sequential serum samples from 2 cattle experimentally infected with AKAV. The value of PI in C-ELISA for convalescent sera from cattle infected with arboviruses correlated with the neutralizing antibody titer to AKAV but was unaffected by the antibodies to other arboviruses. In the validation experiment of C-ELISA using 286 bovine sera previously examined for the AKAV antibody by serum neutralization (SN) test, the relative specificity of C-ELISA was more than 98%, whereas the relative sensitivities of individual MAbs ranged from 49% to 82.2%. Overall agreement between C-ELISA and the SN test varied from 72% to 90% depending on the MAb. These results suggest that the C-ELISA is acceptable as a rapid and specific method for detecting antibodies to AKAV and is a potential alternative to the SN test. 相似文献

The genes TAWAWA1 (TAW1) and ABERRANT PANICLE ORGANIZATION1 (APO1) increase the number of spikelets per panicle (SN). In the present study, we examined the effects of these genes on morphological traits, yield, and yield-related traits including yield components using the near-isogenic lines (NILs) in the genetic background of a japonica rice variety, Koshihikari – NIL-taw1, NIL-apo1-D3, and NIL-apo1-D4 – in a field experiment. The SN and total number of spikelets per area of the three NILs were larger than those of Koshihikari. However, the yield of the three NILs did not exceed that of Koshihikari due to their low filling ability. Interestingly, our field experiments indicated that TAW1 did not affect the diameter of internodes and the PN, whereas APO1 decreased the PN and increased the diameter of internodes. These results suggest that TAW1 and APO1 differently affect yield-related traits. 相似文献

Programmed cell death (PCD) in animals depends on caspase protease activity. Plants also exhibit PCD, for example as a response to pathogens, although a plant caspase remains elusive. Here we show that vacuolar processing enzyme (VPE) is a protease essential for a virus-induced hypersensitive response that involves PCD. VPE deficiency prevented virus-induced hypersensitive cell death in tobacco plants. VPE is structurally unrelated to caspases, although VPE has a caspase-1 activity. Thus, plants have evolved a regulated cellular suicide strategy that, unlike PCD of animals, is mediated by VPE and the cellular vacuole. 相似文献

Diploid gametes generated with tetraploid animals are a stepping stone to improving chromosome manipulation techniques. However,
artificially induced tetraploid individuals generally die soon after hatching. Diploid gametes could be induced by in vivo
cultures of tetraploid primordial germ cells (PGCs) through germ-line chimera. In the present study, characteristics of PGCs
were studied in inviable tetraploid masu salmon, Oncorhynchus masou. Histological observation of tetraploid embryos revealed that the same or smaller numbers of PGCs were observed and they
migrate into the genital ridges as did diploid PGCs during gonadogenesis. By whole-mount in situ hybridization using vasa messenger RNA (mRNA), 4–35 vasa-positive signals were detected in a pair of genital ridges of tetraploids. By cytological
observation of genital ridge cell suspensions, several large round cells were observed, some of which extended pseudopodia.
They also contained large nuclei and round granules in their cytoplasm, characteristics of PGCs. As the results suggest that
inviable artificial tetraploids have PGCs, we expect to achieve diploid gamete production through surrogate propagation and

We reported that meat chicks have either a greater capability to acclimatize to novel environments, or a blunted hypothalamic‐pituitary‐adrenal response to novel environments compared with layer chicks in a commercial base. The present study compared the differences in behavior and plasma corticosterone concentrations under isolation‐induced stress between neonatal meat and layer Nagoya chicks which had been separated from the same population. Both types of neonatal chicks reared in groups were individually separated and their spontaneous activity and distress‐induced vocalizations were monitored for 10 min. The responses of the two types were remarkably different, with the meat chicks being less active than the layer chicks. Distress‐induced vocalizations were fewer in the meat than in the layer chicks. The meat chicks spent more time in a sleeping posture during isolation‐induced stress. Plasma corticosterone concentrations measured at the end of the test tended to be higher in the layer chicks than in meat ones, but not significantly. In conclusion, the selection of Nagoya chickens for meat and layer may have trends similar to those observed in commercial chickens in relation to stress susceptibility. 相似文献

We investigated the optimal timing of simeconazole (RS-2-(4-fluorophenyl)-1-(1H-1,2,4-triazol-1-yl)-3-trimethylsilylpropan-2-ol) application for controlling rice kernel smut in field trials in Miyagi Prefecture,
Japan, using formulations of simeconazole (1.5% granules). The field tests revealed that a submerged application of simeconazole
granules (450–600 g ai/ha) at 1–5 weeks before heading was highly effective against kernel smut, with treatments 1–2 weeks
before heading being the most effective. Submerged application of the fungicide at 2–5 weeks before heading was also highly
effective against false smut, with treatment 3 weeks before heading being the most effective. These periods overlap the timing
for optimal application of simeconazole to control rice sheath blight and ear blight. Consequently, we concluded that treatment
with simeconazole 2–3 weeks before heading can be a useful tool for controlling all four diseases. 相似文献

G protein-coupled receptor (GPR) 120 is an unsaturated fatty acid receptor, which is
associated with various physiological functions. It is reported that the genetic variant
of GPR120, p.Arg270His, is detected more in obese people, and this genetic variation
functionally relates to obesity in humans. Obesity is a common nutritional disorder also
in dogs, but the genetic factors have not ever been identified in dogs. In this study, we
investigated the molecular structure of canine GPR120 and searched for candidate genetic
variants which may relate to obesity in dogs. Canine GPR120 was highly homologous to those
of other species, and seven transmembrane domains and two N-glycosylation sites were
conserved. GPR120 mRNA was expressed in lung, jejunum, ileum, colon, hypothalamus,
hippocampus, spinal cord, bone marrow, dermis and white adipose tissues in dogs, as those
in mice and humans. Genetic variants of GPR120 were explored in client-owned 141 dogs,
resulting in that 5 synonymous and 4 non-synonymous variants were found. The variant
c.595C>A (p.Pro199Thr) was found in 40 dogs, and the gene frequency was significantly
higher in dogs with higher body condition scores, i.e. 0.320 in BCS4–5 dogs, 0.175 in BCS3
dogs and 0.000 in BCS2 dogs. We conclude that c.595C>A (p.Pro199Thr) is a candidate
variant relating to obesity, which may be helpful for nutritional management of dogs. 相似文献

The genetic contribution of 51 broodstock, comprising 29 females and 22 males, reared at Hiroshima City Marine Products Promotion
Center for the production of stocked black sea bream was monitored during two consecutive years using seven microsatellite
DNA loci. The high discrimination ability of these markers was reflected in the polymorphic identification content (PIC=0.831), the exclusion probability (Q≈1), and the low probability of identity index (I=3.635−10). The total number of breeders contributing to the mating process was estimated at 32 (62.7%) in 2000 and 30 (58.8%) in 2001.
On pedigree reconstruction, 69.3% of the offspring were successfully assigned to a single broodstock pair. Loss of alleles
accounted for 16.9% during seed production; nevertheless, 90.9% of males and 69.0% of females participated in the mating process.
Based on microsatellite genetic tagging, 58.9% of the fish sampled during the two months after release were identified as
hatchery stock, presenting no significant differences from wild conspecifics in either fork length or body weight. 相似文献
