Prediction of grain protein content in winter wheat through leaf color measurements using a chlorophyll meter

A field trial was conducted over a 3-year period at the Hokkaido Kitami Agricultural Experiment Station to examine whether the grain protein content (GPC) of a winter wheat cultivar (Triticum aestivum L. cv. Chihokukomugi) suitable for Japanese noodle-making could be predicted before harvest. The prediction of the GPC was accurate based on the color of the second leaf (just below the flag leaf) at the end of the emergence of the inflorescence, when nitrogen application was graded. In order to evaluate the reliability of this test, a survey of 95 wheat fields in the eastern part of Hokkaido was also carried out during a 3-year period. The prediction of the GPC for this cultivar based on the color of the second leaf was less accurate across many sites. The results of this survey, however, suggested that the leaf color could be used as an index for ranking the GPC as low or high in relation to processing requirements. When the leaf color value of the second leaf measured with a chlorophyll meter at the end of the emergence of the inflorescence was less than 40, it was predicted that the GPC would be lower than the processing requirement. This index could be applied to the cultivars grown in the eastern part of Hokkaido, except for those grown on peat soils.     

Comparative external morphology of cetacean spermatozoa

SUMMARY: To compare size and morphology of spermatozoa in cetaceans, sperm and epididymis samples were collected from 10 species in four families and spermatozoa were observed with phase-contrast and scanning electron microscopes. According to the average total length of the spermatozoa, the 10 species examined were classified into the following four groups in order of increasing size: (i) Baird's beaked (Ziphiidae) and Bryde's whales (Balaenopteridae); (ii) Dall's and finless porpoises (Phocoenidae); (iii) common, bottlenose, and Pacific white-sided dolphins (Delphinidae); and (iv) killer and short-finned pilot whales, and Risso's dolphin (Delphinidae). Spermatozoa head length of Bryde's whale and the finless porpoise were shorter than those of the other species. Spermatozoa head width was widest in the killer whale and thinnest in the Baird's beaked whale. The lateral aspects of sperm heads from the 10 species were characterized as the 'anterior region of the sperm head is thin and flat while the posterior region is thick.' The dorsal aspects of sperm heads were 'paddle-shaped' in Bryde's whales, 'bowling pin-shaped' in Baird's beaked whales, 'Japanese fan-shaped' in killer whales, an 'elongated ellipsoid shape' in Delphinidae except for killer whales, and 'ellipsoid shaped' in Phocoenidae. Size and morphology of the spermatozoa showed interspecific differences among the 10 species examined, which correspond to cetacean taxonomic classification.     

Comparison of environmental conditions in two representative oyster farming areas: Hiroshima Bay,western Japan and Oginoham a Bay (a branch of Ishinomaki Bay), northern Japan

ABSTRACT:   Sea water environmental conditions over annual cycles were investigated and compared between two oyster farming areas, western Hiroshima Bay and Oginohama Bay (a branch of Ishinomaki Bay) in Miyagi Prefecture, to appropriately manage oyster culture or more efficiently utilize farming areas. The environmental parameters of temperature, salinity, nutrient concentrations (NO₂–N, NO₃–N, NH₄–N, PO₄–P, and SiO₂–Si) and size-fractionated chlorophyll- a (<0.2, 2–20, >20µm), and abundances of microzooplankton were measured in each bay at the surface, and 2 and 5 m depth layers. Differences in the annual mean values and results with monthly paired Student's t -tests showed that salinity was lower, and temperature, nutrient (especially PO₄–P) and chlorophyll- a concentrations, and abundance of microzooplankton, were higher in Hiroshima Bay than in Oginohama Bay. Differences in environmental conditions between inshore and offshore areas of each bay suggest that inflows of river water in western Hiroshima Bay and sea water from offshore had the most significant effects on the environmental conditions. It is concluded that such oceanographic and biological differences strongly affect the oyster farming system, especially regarding the optimum usage of offshore areas in Summer under clean, cold and stable seawater conditions, rather than food quantity in Hiroshima Bay, and under more abundant food conditions in Oginohama Bay.

Regulated fast nucleocytoplasmic shuttling observed by reversible protein highlighting

The observation of the regulation of fast protein dynamics in a cellular context requires the development of reliable technologies. Here, a signal regulation cascade reliant on the stimulus-dependent acceleration of the bidirectional flow of mitogen-activated protein kinase (extracellular signal-regulated kinase) across the nuclear envelope was visualized by reversible protein highlighting. Light-induced conversion between the bright and dark states of a monomeric fluorescent protein engineered from a novel coral protein was employed. Because of its photochromic properties, the protein could be highlighted, erased, and highlighted again in a nondestructive manner, allowing direct observation of regulated fast nucleocytoplasmic shuttling of key signaling molecules.     

Oxygen isotopic compositions of asteroidal materials returned from Itokawa by the Hayabusa mission

Meteorite studies suggest that each solar system object has a unique oxygen isotopic composition. Chondrites, the most primitive of meteorites, have been believed to be derived from asteroids, but oxygen isotopic compositions of asteroids themselves have not been established. We measured, using secondary ion mass spectrometry, oxygen isotopic compositions of rock particles from asteroid 25143 Itokawa returned by the Hayabusa spacecraft. Compositions of the particles are depleted in (16)O relative to terrestrial materials and indicate that Itokawa, an S-type asteroid, is one of the sources of the LL or L group of equilibrated ordinary chondrites. This is a direct oxygen-isotope link between chondrites and their parent asteroid.     

Furazolidone induces the activity of microsomal enzymes that metabolize furazolidone in chickens

The nitrofuran antibacterial agent furazolidone (FZ) is still used in veterinary medicine in some countries in the Middle and Far East. The present study aimed to show the effect of FZ on the activity of microsomal enzymes that metabolize FZ, and to identify the enzyme that contributes to FZ metabolism in chickens. Wistar rats and White Leghorn chickens were administered FZ once a day for four consecutive days. FZ metabolism was accelerated by FZ administration in chickens, but not in rats. The elevation of FZ metabolism coincided with the induction of NADPH cytochrome P450 reductase (CPR) activity in chickens, but such induction was not observed in rats. FZ metabolizing activities were inhibited in the presence of a CPR inhibitor (diphenylene iodonium chloride) but not by the addition of archetypal cytochrome P450 inhibitors (CO or n-octylamine). The preset study concluded that FZ accelerated its own metabolism in the chicken by induction of the activity of CPR.
