Guggal, a threatened species that is endemic to western India, is tapped to extract medicinally important oleo–gum–resin(guggul). However, the plant dies after gum exudation. The indigenous tapping techniques used by local people were examined in Gujarat, India to discover the scientific basis behind these techniques: selection of gum inducer, season of tapping, and plant parts to be tapped. First, the presence of Xanthomonas axonopodis pv. commiphorae(Xac) in the gum suspension used for tapping was established. This bacterium induces gum oozing from the tapped plants and later, causes them to die off. The population of Xac in gum was found to decrease with the age of the gum. With that, fresh gum increased the tapping success. Second, local people preferred tapping during the warm season, which we validated by determining that Xac growth was best at 30 °C. Tapping during September(mean temperature 25.7–30.0 °C) clearly favoured growth of the pathogen and yielded maximum guggul. Multiple tapping on a mature tree ensured maximum gum extraction before its death. Finally, application of indigenous technology under natural plant stands by the local people ensured availability of this important raw drug for consumption. Our study established that the age–old traditional methods have a strong scientific basis. However, it is imperative to formulate strategies based on contemporary scientific understanding to protect this natural resource before it becomes extinct.  相似文献

Mamejvo (Enicostemma axillare Raynal) is a perennial herb with diversified health benefits. It was found to be affected by a witches?? broom disease under the field conditions at Anand, Gujarat, India. Affected plants were 53.33% reduced in height due to shortened internodes, leading to typical witches?? broom symptoms. Flower size was reduced significantly in the affected plants compared with the healthy ones and their petal color gradually turned green (virescence). Affected plants produced 2.5 times more dry herbage than the normal plants but with less active ingredient (swertiamarin). Symptomatic plants died early while root suckers from such plants failed to establish after transplanting. DAPI stained transverse sections of affected shoot tips showed the presence of discrete greenish fluorescence in the phloem cells under UV light. Amplification of 1.2?kb phytoplasma specific rDNA fragment from diseased tissue confirmed the presence of the pathogen. Further characterization of the pathogen through virtual rDNA?CRFLP pattern and rDNA sequence based phylogeny suggested that the pathogen, Enicostemma witches?? broom phytoplasma, belonged to 16SrII?CC group of ??Ca Phytoplasma aurantifolia??.  相似文献

Different laboratory synthesized metal nanoparticles viz. Copper oxide (CuO), Zinc oxide (ZnO) and silver doped titanium dioxide (Ag‐TiO2) were studied for their effect on hatching and survival of larvae and fry of Indian major carp, rohu, Labeo rohita both in direct application in tank water & coated onto tanks. Among these nanoparticles, CuO and ZnO nanoparticles exhibited highest percentage of hatching in both direct addition (78.0 ± 3.1% and 78.05 ± 4.2%, respectively) and coating onto tanks (58.6 ± 2.1% and 61.2 ± 2.7%, respectively) at 1 mg mL?1 while least percentage of hatching was recorded in Ag‐TiO2 nanoparticles irrespective of its concentration & mode of supplementation. Highest survival of L. rohita fry (50.13 ± 2.2%) was observed after 15 days post hatching in CuO coated tanks followed by ZnO coated tanks (38.6 ± 2.8%) while least was recorded in Ag‐TiO2 coated tanks (22.53 ± 3.0%). However in control tanks coated with Poly‐Urethane base with hardener and uncoated control tanks, the survival was 42.4 ± 1.2% and 41.36 ± 1.8% respectively. Further, significantly lower microbial load of water was recorded in CuO nanoparticles coated tanks (1.5 × 1010 CFU L?1) as compared to uncoated control tanks (1.1 × 1016 CFU L?1) without affecting water quality parameters. On the other hand, in Ag‐TiO2 coated tanks, significantly lower microbial load (1.0 × 106 CFU L?1) as compared to uncoated control tanks at 15 days post hatching was recorded. However, Ag‐TiO2 was toxic to L. rohita larvae & fry both in direct application and coating onto tanks. Considering the beneficial effects of CuO nanoparticle application, it has the scope of being used in a more eco‐friendly way in hatchery operations.  相似文献

The contamination of hazardous metal(loid) is one of the serious environmental and human health risks. This study isolated a total of 40 cadmium (Cd)- and arsenic (As)-resistant bacterial isolates from coastal sediments by pour plate technique using tryptic soy agar supplemented with Cd or As (50 mg l?1) for use as metal(loid) bioremediation agents. Out of 40, 4 isolates, RCd3, RCd6, RAs7, and RAs10, showed a relatively higher growth rate in Cd- or As-supplemented culture media which were selected for further study. The selected isolates showed a high minimum inhibitory concentration (60–400 mg l?1 for Cd and 400–2200 mg l?1 for As), which demonstrated their remarkable Cd and As resistance capabilities. The metal(loid) removal efficiencies (0.032–0.268 μg Cd h?1 mg?1 and 0.0003–0.0172 μg As h?1 mg?1 [wet weight cell]) of selected isolates indicated their greater magnitude in absorbing Cd compared to As from water. Phylogenetic analysis of the 16S rDNA sequences revealed that isolates RCd3, RCd6, RAs7, and RAs10 were closely related to Acinetobacter brisouii, Pseudomonas abietaniphila, Exiguobacterium aestuarii, and Planococcus rifietoensis, respectively. Because of high Cd and As resistance and removal efficiency, the selected isolates can survive in a high metal(loid)-contaminated environment and could be a potential tool for bioremediation of high metal(loid)-contaminated effluents to protect the aquatic environment.  相似文献

Guggal (Commiphora wightii (Arnott) Bhandari comb. nov.) is a small tree which is tapped for medicinally important oleo?Cgum?Cresin. Naturally infected plant oozes oleo?Cgum?Cresin from its trunk and primary branches. However, in either case, the plant dies slowly after oozing. A bacterium was established to be responsible for these phenomena. Four isolates of this bacterium were characterised by biochemical tests, Biolog GN2 microplate reaction, rDNA sequencing, which suggested that the pathogen belonged to the genus Xanthomonas. However, phylogenetic analysis based on chaperone protein (dnaK) gene, TonB?Cdependent receptor (fyuA) gene, DNA gyrase B (gyrB) gene and RNA polymerase sigma factor (rpoD) gene sequences placed it as a member of X. axonopodis 9.2 rep?CPCR/DNA?CDNA homology cluster close to X. perforans, X. alfalfae and X. euvesicatoria. Further elucidation of phylogenetic position of the test strains was achieved from a gyrB based tree considering sequences from 71 representative strains. Test strains were confirmed to be members of X. axonopodis. These had very narrow infectivity limited to Commiphora spp. Hence, we propose a novel pathovar, X. axonopodis pv. commiphoreae pv. nov. as the cause of gum oozing in guggal.
