OBJECTIVE: To purify canine carbonic anhydrase isoenzyme III (CA-III) and determine plasma, serum, and tissue concentrations of CA-III in healthy dogs and dogs with experimentally induced muscle damage. ANIMALS: 121 healthy Beagles. PROCEDURE: Muscle was obtained from 2 Beagles after euthanasia, and CA-III was purified and characterized by use of column chromatography and electrophoresis, respectively. A CA-III-specific ELISA was developed to determine concentrations of CA-III in plasma of 116 dogs and tissues of 1 dog. Serum creatine kinase (CK) activity and CA-III concentration were also determined before and after induction of muscle damage by IM injection of 2 ml of 10% lidocaine to 2 dogs. RESULTS: Canine CA-III had a molecular weight of 28 kd and an isoelectric point of 8.2. Mean (+/- SD) concentration of CA-III in plasma of healthy dogs was 16.91 +/- 9.55 ng/ml. The highest tissue concentration of CA-III was detected in skeletal muscle. Serum concentration of CA-III increased and peaked within the first 2 to 3 hours after induction of muscle damage. The increase in CA-III concentration was more rapid than that of CK activity, and concentration reached its maximum and returned to baseline sooner than did CK activity. CONCLUSIONS AND CLINICAL RELEVANCE: The CA-III ELISA we developed was a sensitive method for determining CA-III concentrations in plasma, serum samples, and tissue specimens of dogs. Use of this ELISA requires only a small volume of serum and may enable the study of changes in CA isoenzyme concentrations associated with muscle disorders in dogs. 相似文献

The growing condition of afforestation trees and damage to lower-story trees caused by felling and hauling of upper-story
trees in a multi-storied forest in Peninsular Malaysia was surveyed. Seventy months have passed since the planting ofAcacia mangium upperstory trees, which have grown 233 m3/ha in volume. The growth ofShorea leprosula thirty-five months after planting of lower-story trees have reached 5.54 m in height. However, as the ratio of height to
basal diameter exceeds 100:1, trees are tapering to a considerable degree. A backhoe was installed in order to fell the upper-story
trees. From the results of logging, the damage ratio estimation of the lower-story trees was simulated by using the Monte
Carlo method. The damage ratio differed considerably depending on planting patterns, height of upper-story trees and felling
skills. The results of this survey proved that the felling and hauling of upper-story trees would be technically feasible
in the future, except when one row upper-story trees and one row lower-story trees method were employed. 相似文献

This study examines a method for designing an optimum profile of a forest road in a short matter of time. In this method,
a genetic algorithm (GA) searches for points where the gradient changes and dynamic programing (DP) designs the longitudinal
slope. This method can be applied to the ground heights of an actual forest road. The evaluation function of the DP assesses
the total expenditure. The inverse of this function is the fitness function. Crossover and mutation are operators of an evolution
simulation of a GA. They are each repeated 20 times and 420 profiles are designed and assessed, all within about 10 min. A
comparison of the profile designed by the GA with that of an actual forest road shows only one section where the formation
level differed substantially. Overall, both profiles seem to show a high degree of correspondence. From this result, we believe
that the profile designed by the GA is best, or the one of the best. A conventional method takes about 65 h to calculate all
changing points of gradients. However, by using the GA, the calculation time was reduced to about 1/389 that of the coventional
method, showing that the method which uses the GA through the profile design is very useful. 相似文献

Lymph drainage routes from the abdominal and pelvic cavities in beagle dogs were observed serially by following the time course of India ink administered intraperitoneally. Four systems of lymph drainage routes from the peritoneal cavity were observed in this study. The earliest drainage returned to the cranial mediastinal lymph nodes via the sternal lymph vessels; subsequently, the sternal lymph nodes located along the internal thoracic artery became involved. Then, a drainage route via the lymph vessel along the left vagus nerve was observed. The final drainage route flowed into the lateral lymph vessel through the thoracic duct located on the vertebra. These results show that India ink is absorbed from the peritoneal cavity, and that the lymph drainage first flows mainly towards the cranial mediastinal lymph nodes through the ventral lymphatic channels. Our serial observations suggest that, over time, the lymph drainage routes changed from the ventral abdominal to the dorsal thoracic lymphatic channels in the thorax. 相似文献

Current‐year Fagus crenata seedlings survive damping‐off in the forest‐edge plot compared to those in the closed‐stand plot. Greater illumination found in the forest‐edge plot is likely to influence the defense of seedlings against pathogenic attack, by increasing the amount of phenolic compounds and forming periderm. This study aimed to clarify the chemical defense system by (i) isolation of antifungal compounds from seedling hypocotyls in the forest‐edge plot, (ii) quantification of antifungal compounds in hypocotyls in the forest‐edge and closed‐stand plots, (iii) estimation of their antifungal activities against Cylindrocarpon sp. and Colletotrichum dematium and (iv) clarification of localization of phenolics in the hypocotyl tissues. Catechin and epicatechin were isolated as antifungal compounds showing inhibition of microconidia germination of Cylindrocarpon sp. and mycelial growth of Cylindrocarpon sp. and C. dematium. In the beginning of July when damping‐off was the most severe in the closed‐stand plot, the total amount of catechin and epicatechin in the forest‐edge seedlings was approximately three times greater than that in the closed‐stand seedlings. Flavan‐3‐ols, including catechin and epicatechin, were distributed mainly in the epidermal tissue on June 1 and in cortical tissues on June 15 and July 6, all from plants harvested from the forest‐edge plot. We conclude that F. crenata could survive damping‐off through chemical defense via catechin and epicatechin production and its distribution in seedling hypocotyls. 相似文献

The distribution of cortical resin canals and periderm formation in the cortex of Pinus thunbergii was studied in relation to early invasion of the pinewood nematode, Bursaphelenchus xylophilus. Nematode invasion was restricted in stem cuttings of P. thunbergii in which periderm closed cortical resin canals. Early invasion of the nematodes was also restricted in stem cuttings where wound periderm had formed in response to prior nematode inoculation. It was concluded that early invasion of pinewood nematodes in living bark tissue is restricted by periderm in mature stems and by wound periderm that had formed as the result of previous nematode infection. 相似文献

In order to study the changes in ectomycorrhizal development during symptom expression of pine wilt disease, root window observations were conducted concurrent with measurements of leaf water potential as well as photosynthetic and transpiration rates of 5‐year‐old Pinus thunbergii trees that were inoculated with the pinewood nematode (PWN) Bursaphelenchus xylophilus. Infected trees were compared with girdled and uninfected control trees. Ectomycorrhizas developed constantly during the experimental period in control trees but did not develop in the girdled trees. Ectomycorrhizal development ceased within 2 weeks in those trees that finally died after PWN infection. In the trees that survived PWN infection, ectomycorrhizal development ceased within 1–4 weeks of inoculation but was resumed thereafter within 3–6 weeks. Ectomycorrhizal development ceased prior to a decrease in both photosynthetic rate and leaf water potential in the inoculated trees. 相似文献

Equine carbonic anhydrase isozymes (CA-I and CA-II) were purified from erythrocytes by several column chromatography. Polyclonal anti-CA-I and anti-CA-II sera were produced in rabbits. Sensitive competitive enzyme-linked immunosorbent assays (ELISA) were established to determine the developmental changes in CA-I and CA-II levels in equine erythrocytes. Concentrations of CA-I and CA-II in erythrocytes from 150 clinically normal thoroughbreds (123 racehorses and 27 riding horses) were determined by ELISA. Mean (+/- SD) concentrations of CA-I and CA-II in racehorses were 1.70 +/- 0.48 and 0.94 +/- 0.13 mg/g hemoglobin (Hb), respectively. Mean concentrations of CA-I and CA-II in riding horses were 2.34 +/- 0.52 and 0.76 +/- 0.08 mg/g Hb, respectively. When the CA levels in racehorses and riding horses were compared, the CA-I level in riding horses was higher than that in racehorses (p=0.01). The CA-II level in racehorses was higher than that in riding horses (p=0.02). These data suggest that the levels of CA isozymes in erythrocytes of racehorses were influenced by chronic physical stress. The CA-I concentration in erythrocytes of 2-month-old horses was approximately 0.25 mg/g Hb. The CA-I level noticeably increased during the first year of life and approached normal adult levels by 2 years. Dietary habits must be considered as one of the major potential factors resulting in acquired malocclusions in rabbits. Although the dentition of the wild rabbit and the domesticated laboratory rabbit are basically identical, dietary habits are noticeably different. Therefore, the prevalence of tooth problems between these lagomorph species were investigated anatomically and radiographically. Mean measurements of the skull and dental arches suggested that wild rabbits have slightly shorter and wider skulls and dental arches compared with domestic laboratory rabbits. Root elongation of incisors and check teeth, and periodontal disease were more frequently observed in domestic laboratory rabbits. Diagnostic radiographs from domestic pet rabbits showed relatively higher crowns, severe root elongation, and advanced periodontitis. These results do not provide definitive evidence that dietary habits cause malocclusions, however they suggest that diet is a major factor in the initiation of malocclusions in rabbits. 相似文献
