Effects of β‐cyclodextrin diallyl maleate (CD‐M) on methane production, ruminal fermentation and digestibility were studied both in vitro and in vivo. In in vitro study, diluted ruminal fluid (30 mL) was incubated anaerobically at 38°C for 6 and 24 h with or without CD‐M using hay plus concentrate (1.5:1) as a substrate. The CD‐M was added at different concentrations (0, 1.25, 2.5, 5.0 and 7.5 g/L). The pH of the medium and numbers of protozoa were not affected by the addition of CD‐M. Total volatile fatty acids were increased and ammonia‐N was decreased, molar proportion of acetate was decreased and propionate was increased (P < 0.05) by CD‐M. Methane was inhibited (P < 0.05) by 14–76%. The effect of CD‐M on methane production and ruminal fermentation was further investigated in vivo using four Holstein steers in a cross‐over design. The steers were fed Sudangrass hay and concentrate mixture (1.5:1) with or without CD‐M (2% of feed dry matter) as a supplement. Ruminal proportion of acetate tended to decrease and that of propionate was increased (P < 0.05) 2 h after CD‐M dosing. Total viable counts, cellulolytic, sulfate reducing, acetogenic bacteria and protozoa were unaffected while methanogenic bacteria were decreased (P < 0.05) by CD‐M. The plasma concentration of glucose was increased, whereas that of urea‐N was decreased (P < 0.05). Methane was inhibited (P < 0.05) from 36.4 to 30.1 L/kg dry matter intake by the addition of CD‐M. Apparent digestibilities of dry matter and neutral detergent fiber were not affected while that of crude protein was increased (P < 0.05) in the medicated steers. These data suggested that dietary supplementation of CD‐M decreased methane production and improved nutrient use. 相似文献

Farm managers' decision to cull dairy cows is based on the cows' milk production, history of disorder(s), and reproductive performance, each of which affects dairy cows' lifetime (herd life and productive lifespan). We investigated the relationships among the incidence of clinical mastitis (CM), the reproductive performance, and the culling rate. We also assessed the effects of these relationships on the lifetimes of dairy cows, using the records made before and after the introduction of an automatic milking system (AMS) at Hiroshima University Farm. Milk yield, CM incidence density, and culling rate of dairy cows increased after the AMS introduction. The CM incidence was associated with an elongation of the calving interval in cows with the same parity. CM in the 1st parity might have caused the reductions of the cows' lifetime and their parity at culling. A higher age at first calving (AFC) was associated with an increase in culling rate but did not lead to a significant decrease in lifetime. Investigations of the factors mediating CM in the 1st parity or AFC with CM incidence or culling rate in the later stages might contribute to the control of lifetime of dairy cows. 相似文献

Strains of Xanthomonas campestris pv. vesicatoria Dye 1978 (Xcv), the causal agent of bacterial spot, have been classified into two groups based on their ability to hydrolyze starch. Three monoclonal antibodies (MAbs), 7AH10, 5HB3, and 4AD2, were produced immunized against the living bacteria and were specific to and could distinguish Xcv strains able or unable to hydrolyze starch (Amy+ or Amy–). The MAb 7AH10, obtained against strain UPB141(Amy–) reacted in an enzyme-linked immunosorbent assay with all the Amy– strains (n = 19) and 1 of 11 Amy+ strains. Against Xcv 2625, an Amy– unusual phenotype strain, MAb 5HB3, recognized 97% of our worldwide collection of Xcvs (n = 30). Also against that strain, the MAb 4AD2 reacted with none of the homologous Amy– phenotypes and with 90% (n = 11) of the heterologous Amy+ phenotypes. For all the MAbs, cross reactions with other pathovars or species were less than 4% (n = 67). By assaying a Japanese collection of strains against the three MAbs, the Amy+ strains were distinguished from the Amy– strains, and their relation with other world strains could be demonstrated. To investigate seasonal variations in the digestive functions of sika deer, five female sika deer were provided with an amount of alfalfa hay cubes equivalent to voluntary food intake during winter. We measured the rate at which the food passed through the digestive tract, digestibility and rumen fermentation during the summer (August), autumn (November), winter (February) and spring (May). Total mean retention time in the digestive tract during summer and autumn was numerically longer than that in winter and spring, but the difference did not reach significance. Organic matter and fiber were less digestible in summer and autumn than in winter and spring (P < 0.05), whereas the digestibility of the dry matter tended to vary with the seasons (P < 0.1). Ruminal pH values seasonally changed (P < 0.01), and were the lowest in autumn. The concentration of ruminal ammonia‐nitrogen differed significantly among the seasons (P < 0.05), increasing in winter and decreasing during spring and summer. The numbers of protozoa changed significantly among the seasons (P < 0.05), being higher in autumn than in winter and spring, and intermediate in summer. The concentration of total volatile fatty acids was not seasonally affected, but the molar percentages of propionic acid and butyric acid significantly changed according to season (P < 0.05 and P < 0.01, respectively), and the ratio of acetic to propionic acid tended to change with the seasons (P < 0.1). The results of this study suggested that the digestive functions in sika deer, fed a commercial diet at a restricted level, differed notably among the seasons and these variations might partially be due to environmental effects. 相似文献

Using soft X-ray photography, two-dimensional observations were made of the water distribution within the green stem ofCryptomeria japonica D. Don. Variations in the distribution pattern of the wet area in the horizontal plane of the lower stem were described. The distribution patterns showed extreme variation among individual trees, with broad wet-area distribution types appearing, namely, the regularly distributed wet area and the irregularly distributed wet area. We defined five basic types of wet-area distribution patterns on the basis of their regularity or irregularity. It was concluded that the between-tree variation in the wet-area distribution causes the between-tree variation in the mean moisture content of the heartwood. In this study we examined the effects of 3.3% Garcinia cambogia extract on 10% sucrose loading in mice for 4 weeks. Treatment was found to have no effect on body weight, fat pad weight or serum glucose level. On the other hand, serum total cholesterol, triglycerides, NEFA were observed. Levels of serum insulin and leptin, as well as the leptin/WAT ratio, were lower in the treated mice than in the control. These findings suggested that G. cambogia extract efficiently improved glucose metabolism and displayed leptin-like activity. 相似文献

The objectives of this study were to examine widely divergent fiber sources for their efficacy as ingredients in a meat-based dog diet and to determine the effects of these fibers on fecal excretion responses and mean retention time of marked fiber in the gastrointestinal tract of the dog. Fiber sources tested included beet pulp (BP), tomato pomace (TP), peanut hulls (PH), wheat bran (WB) and alkaline hydrogen peroxide-treated wheat straw (AHPWS). Diets were isonitrogenous (5.3% N) and iso-total dietary fiber (TDF; 12.5%). Thirty female English Pointers (five/treatment) were used in the experiment. Intakes of DM and OM were similar among treatments. The highest intakes of ether extract (EE) occurred on the TP, PH and WB treatments. Dogs fed PH ingested the most crude fiber (23.6 g/d), NDF (53.5 g/d), ADF (34.3 g/d) and TDF (59.7 g/d). Digestibilities of DM and OM for all fiber treatments were lower than the control (87.6 vs 81.8% for DM; 90.2 vs 85.4% for OM), but values were similar among fiber sources. The highest EE and N digestibilities occurred on the control and AHPWS treatments. No differences were noted among exogenous fiber-containing treatments in fiber component digestibility. Digestible energy and ME values generally were similar among treatments. Among fiber sources, BP resulted in the greatest amount of wet feces excreted (270 g/d) and the lowest fecal DM (30.3%). No differences among fiber sources were noted in frequency of defecation or mean retention time. Iso-TDF diets (containing, on average, 12.5% TDF) appear to be utilized similarly, regardless of the diversity in sources of fiber tested. 相似文献
