An estrogen-binding protein and endogenous ligand in Saccharomyces cerevisiae: possible hormone receptor system 总被引:10,自引:0,他引:10
A protein macromolecule in the cytosol of the unicellular eukaryotic yeast Saccharomyces cerevisiae selectively binds the vertebrate estrogen hormone 17 beta-estradiol with high affinity. Lipid extracts of the yeast cells or the conditioned growth medium yield a substance that can bind competitively to the tritiated estradiol-binding sites in the yeast and to mammalian estrogen receptors. These findings suggest that the binding protein may be a primitive hormone receptor and that the lipid-extractable substance represents the endogenous ligand. 相似文献

Interest of mussel hatchery development has increased during the last years as natural seed collection does not always satisfy the demand of producers. Success on larval bivalve culture depends on several factors such as diet, temperature or stocking density. In this study, the effect of larval density in mussel culture success has been assessed testing three larval densities: high (50 larvae mL?1), medium (25 larvae mL?1) and low (5 larvae mL?1). Results showed higher growth and survival at low density but the amount of postlarvae was low. The highest number of settled larvae was observed at high density but mortality was also high and a delay on the larval development was detected producing a prolongation on the culture length. However, medium density resulted in a good survival and growth and also in a considerable amount of postlarvae, as this density could be adequate for massive mussel seed production. Cortisol content on the larvae and postlarvae was also analysed and significant differences were obtained among groups probably due to the stress associated to settlement and metamorphosis. 相似文献

At 25 °C, metamorphosis in leopard grouper Mycteroperca rosacea larvae took 60 days after hatching. The total length at day 1 was 1.95±0.22 mm and juveniles reached a length of 30.64±0.23 mm at day 60; the increase was approximately linear. We describe eight stages of development during this period. Larvae with the yolk sac attached occur from days 1 to 4 (Stages 1 and 2). The preflexion occurs on days 5–20 (Stages 3 and 4). Bending notochord occurred at day 25 (Stage 5). The other morphological changes that precede the juvenile phase occurred progressively until day 60 (Stages 6–8). Allometric growth in the height and length of the head, trunk length, height and length of the tail and the diameter of the eye compared with the total length showed two distinct stages of growth. Inflexion point, where growth is positive, occurred when larvae reached between 18.75 and 21.59 mm, which corresponds to larvae at days 35–40. 相似文献

Lectins are proteins or glycoproteins with the ability to link to carbohydrates at the cell surface in a specific and reversible manner. Studies have showed that lectins are demonstrate immunomodulatory and antitumor agents. This study aims to evaluate the effect of lectin extracted from the seeds of Canavalia brasiliensis (ConBr) on murine melanoma B16F10 cells by analyzing cell viability, apoptosis index, cell migration, production of cytokines and nitric oxide (NO). Results showed that ConBr was able to reduce cell viability and thwart apoptosis, which could be observed by decrease in cell migration. ConBr also induced NO and IL-12 synthesis. Altogether, these data demonstrate the potential of ConBr as a therapeutic agent for melanoma. 相似文献

The effect on mycelial growth of the fungus Botrytis cinerea of a set of structurally related tricyclic hydroquinones [9,10-dihydroxy-4,4-dimethyl-2,3,5,8-tetrahydroantracen-1(4H)-one and 9,10-dihydroxy-4,4-dimethyl-5,8-dihydroanthracen-1(4H)-one derivatives] and tricyclic quinones [4,4-dimethylanthracen-1,9,10(4H)-trione derivatives] was studied. In general, the anthraquinones presented higher activity than the anthrahydroquinones. Anthraquinone and anthrahydroquinone derivatives with methyl groups on the A ring showed higher antifungal activity than the unsubstituted ones, 4,4,6,7-tetramethyl-(4H)-anthracene-1,9,10-trione being the most active compound of this set. The presence of a polar group such as hydroxymethyl reduced the activity. The effect of two anthrahydroquinones and two anthraquinones on the conidia germination of the fungus was also determined. Anthrahydroquinones did not affect the germination. The most active compound was 4,4-dimethylanthracene-1,9,10(4H)-trione, with 100% inhibition of germination at 7 h of incubation. These results again suggest that the structure of the anthraquinones is important in exerting an antifungal effect on B. cinerea. Furthermore, possible mechanisms of action of compound 4,4-dimethylanthracene-1,9,10(4H)-trione were studied. This compound did not produce lipoperoxidation of membrane and did not induce the formation of oxygen reactive species, but it was able to permeabilize the plasmatic membrane of B. cinerea, increasing the phosphorus concentration in the intracellular medium. 相似文献

Soil microorganisms with phytase- and 1-aminocyclopropane-1-carboxylate (ACC) deaminase activities are widely studied as plant growth-promoting rhizobacteria (PGPR). Here, we explored the bacterial community structure and occurrence of putative PGPR in plants grown in agro-ecosystems and undisturbed ecosystems from northern, central, and southern Chile. Total rhizobacterial community structure was evaluated by denaturing gradient gel electrophoresis, and dominant bands present in diverse ecosystems were sequenced. Significant differences in total bacterial communities were shown with some bacterial orders (Enterobacteriales, Actinomycetales, and Rhizobiales) being highly similar to both ecosystems. Twenty-nine putative PGPR, showing phytate- and ACC-degrading activities and production of auxin, were selected from across the sites. Based on 16S rRNA gene sequencing, the putative PGPR were characterized as Enterobacteriales (Enterobacter, Serratia, Pantoea, Rahnella, Leclercia), Pseudomonas, and Bacillus, consistent with previously reported PGPR and endophytic bacteria. Beta-propeller phytase genes with similarity to Bacillus were also identified. PGPR from agro-ecosystems appeared to show higher auxin production compared to those from undisturbed ecosystems. This study demonstrates that putative PGPR are widely distributed across Chilean soils. Further understanding of their contribution to the growth and adaptation of plant hosts to local soil conditions may provide opportunity for development of new PGPR in Chilean agriculture. 相似文献

Management of soils to facilitate plant beneficial microbial interactions requires basic knowledge of the species composition and microbial community structures in the plant rhizosphere. Here, we examined composition of bacterial communities associated with rhizosphere microsites located at the root tips and mature root zones of Lolium perenne when grown in Chilean ash-derived volcanic soils (Andisols: Freire and Piedras Negras soil series). Community structures were analyzed by pyrosequencing of 16S ribosomal RNA (rRNA) genes followed by in silico analysis for phylogenetic assignments (MOTHUR and Visualization tool for Taxonomic Compositions of Microbial Community (VITCOMIC)). Analysis of the community structure revealed significant differences in community structures in relation to the soil series, which differed particularly in the relative abundance of Cyanobacteria and Firmicutes. However, no significant differences were observed with respect to root microsite location in the same Andisol series. Predominant taxa included members of the Proteobacteria, Actinobacteria, and Acidobacteria. Analysis by VITCOMIC showed that dominant bacterial groups comprised only 5 to 10 % of the total bacterial community and the remaining majority of bacteria included low-abundant taxa (Fusobacteria, Thermotogae, Lentisphaerae, Tenericutes, Deferribacteres Spirochaetes, Planctomycetes, Thermotogae, and Deinococcus-Thermus), most of which have not been previously reported or associated with the plant rhizosphere according to GenBank database. The results indicate that most of bacteria in the Chilean Andisols have not been described to the rhizosphere plants and their functional traits are still largely unknown. 相似文献

A surface energy balance model (SEB) was extended by Lagos et al. Irrig Sci 28:51–64 (2009) to estimate evapotranspiration (ET) from variable canopy cover and evaporation from residue-covered or bare soil systems. The model estimates latent, sensible, and soil heat fluxes and provides a method to partition evapotranspiration into soil/residue evaporation and plant transpiration. The objective of this work was to perform a sensitivity analysis of model parameters and evaluate the performance of the proposed model to estimate ET during the growing and non-growing season of maize (Zea Mays L.) and soybeans (Glycine max) in eastern Nebraska. Results were compared with measured data from three eddy covariance systems under irrigated and rain-fed conditions. Sensitivity analysis of model parameters showed that simulated ET was most sensitive to changes in surface canopy resistance, soil surface resistance, and residue surface resistance. Comparison between hourly estimated ET and measurements made in soybean and maize fields provided support for the validity of the surface energy balance model. For growing season’s estimates, Nash–Sutcliffe coefficients ranged from 0.81 to 0.92 and the root mean square error (RMSE) varied from 33.0 to 48.3 W m?2. After canopy closure (i.e., after leaf area index (LAI = 4) until harvest), Nash–Sutcliffe coefficients ranged from 0.86 to 0.95 and RMSE varied from 22.6 to 40.5 W m?2. Performance prior to canopy closure was less accurate. Overall, the evaluation of the SEB model during this study was satisfactory. 相似文献
