Hyperthermia is a form of a cancer treatment which is frequently applied in combination with radiotherapy (RT) to improve therapy responses and radiosensitivity. The mode of action of hyperthermia is multifactorial; the one hand by altering the amount of the blood circulation in the treated tissue, on the other hand by modulating molecular pathways involved in cell survival processes and immunogenic interactions. One of the most dominant proteins induced by hyperthermia is the major stress-inducible heat shock protein 70 (Hsp70). Hsp70 can be found in the blood either as a free-protein (free HSP70) derived from necrotic cells, or lipid-bound (liposomal Hsp70) when it is actively released in extracellular vesicles (EVs) by living cells. The aim of the study was to evaluate the levels of free and liposomal Hsp70 before and after treatment with RT alone or hyperthermia combined with radiotherapy (HTRT) in dogs and cats to evaluate therapy responses. Peripheral blood was collected from feline and canine patients before and at 2, 4, 6 and 24 h after treatment with RT or HTRT. Hsp70 enzyme-linked immunosorbent assays (ELISAs) were performed to determine the free and liposomal Hsp70 concentrations in the serum. The levels were analysed after the first fraction of radiation to study immediate effects and after all applied fractions to study cumulative effects. The levels of free and liposomal Hsp70 levels in the circulation were not affected by the first singular treatment and cumulative effects of RT in cats however, after finalizing all treatment cycles with HTRT free and liposomal Hsp70 levels significantly increased. In dogs, HTRT, but not treatment with RT alone, significantly affected liposomal Hsp70 levels during the first fraction. Free Hsp70 levels were significantly increased after RT, but not HTRT, during the first fraction in dogs. In dogs, on the other hand, RT alone resulted in a significant increase in liposomal Hsp70, but HTRT did not significantly affect the liposomal Hsp70 when cumulative effects were analysed. Free Hsp70 was significantly induced in dogs after both, RT and HTRT when cumulative effects were analysed. RT and HTRT treatments differentially affect the levels of free and liposomal Hsp70 in dogs and cats. Both forms of Hsp70 could potentially be further investigated as potential liquid biopsy markers to study responses to RT and HTRT treatment in companion animals.  相似文献

In the experimental garden of the Department of Soil Bioengineering and Landscape Construction, University of Applied Life Sciences in Vienna, Austria, coarse root systems of three different brush species were completely excavated and semi-automatically digitized. The species were Lonicera xylosteum, Ligustrum vulgare and Euonymus europaeus. The 3-D root ar-chitectures reveal different growth strategies between species, which are related to ecological characteristics and physical soil proper-ties. The root architecture of Lonicera xylosteum and Ligustrum vulgare, planted in the under layer of the live slope grading, where the soil is very tight and the soil water content and fertility are relatively low, is shallow. However, the root distribution of E. eu-ropaeus, planted in the middle layer, where environmental conditions are better, is deeper. Most of the root biomass of the three spe-cies is concentrated in the 0-30 cm soil layer. A quarter of the root biomass of Ligustrum vulgare is distributed in the upper layer of the plant inlay. E. europaeus has a relatively even distribution in the 30-60 cm and 60-90 cm soil layer.  相似文献

The ability of animals to appropriate sources of food and select among them to obtain a nutritionally adequate diet that is best suited for their physiologic status has been examined in numerous species, but considerable research has addressed self-selection trials with chicks. Observations on diet preference have led to the assumption that birds have an innate ability to regulate their intake of macro- and micronutrients, and that they can do so with a considerable degree of precision (e.g. Holcombe et al. 1975, 1976; Elkin et al. 1985; Harper and Peters 1989; Emmerson et al. 1991; Steinruck et al. 1991a,b; Steinruck and Kirchgessner 1993a,b,c; Shariatmadari and Forbes 1993; Forbes and Shariatmadari 1994, 1996). Experimental evidence also shows that domesticated pigs which are offered a choice between two diets will select a diet containing an adequate amount of protein in preference to one that is deficient in protein, or will select specific proteins among various protein sources (Robinson 1974; Kyriazakis et al. 1990; Kyriazakis and Emmans 1992, 1993; Bradford and Gous 1992; Roberts and Azain 1997). The characteristic amount of a specific protein selected by the animals appeared to be related to the quality of the protein source. This was obvious from a few amino acid selection studies showing that growing-finishing pigs have the ability to select preferentially amino acid-adequate diets over amino acid-deficient diets within a few days (Devilat et al. 1970; Robinson 1975a,b; Davey 1978; Henry 1987, 1993). In contrast, there are also some exceptions to the hypothesis that selection of diet is related to the nutritional quality of the protein fed (Ashley and Anderson 1975). However, some of those studies are not conclusive due to changes in preferences by the animals over time and differences in protein source, palatability and amino acid pattern between the test diets. Since, an investigation of a self-selection behaviour by piglets has rarely been studied in detail, the purpose of this research was to determine whether piglets that are given a choice of diets with high or low lysine contents can regulate the intake of each to optimize lysine intake and performance. The objectives of this study were to determine the preference of piglets for diets varying in lysine content and to determine whether the preferences for these diets change with time. This indispensable amino acid was selected for study because it is first limiting in conventional diets for growing pigs.  相似文献

The nutrient composition of 15 commercially available microalgae powders of Arthrospira platensis, Chlorella pyrenoidosa and vulgaris, Dunaliella salina, Haematococcus pluvialis, Tetraselmis chuii, and Aphanizomenon flos-aquae was analyzed. The Dunaliella salina powders were characterized by a high content of carbohydrates, saturated fatty acids (SFAs), omega-6-polyunsaturated fatty acids (n6-PUFAs), heavy metals, and α-tocopherol, whereas the protein amounts, essential amino acids (EAAs), omega-3-PUFAs (n3-PUFAs), vitamins, and minerals were low. In the powder of Haematococcus pluvialis, ten times higher amounts of carotenoids compared to all other analyzed powders were determined, yet it was low in vitamins D and E, protein, and EAAs, and the n6/n3-PUFAs ratio was comparably high. Vitamin B12, quantified as cobalamin, was below 0.02 mg/100 g dry weight (d.w.) in all studied powders. Based on our analysis, microalgae such as Aphanizomenon and Chlorella may contribute to an adequate intake of critical nutrients such as protein with a high content of EAAs, dietary fibers, n3-PUFAs, Ca, Fe, Mg, and Zn, as well as vitamin D and E. Yet, the nutritional value of Aphanizomenon flos-aquae was slightly decreased by high contents of SFAs. The present data show that microalgae are rich in valuable nutrients, but the macro- and micronutrient profiles differ strongly between and within species.  相似文献

The effect of conjugated linoleic acid (CLA) on energy metabolism was studied in a respiration experiment with 20 growing pigs. From 70 kg liveweight onwards the animals received isoenergetic rations containing 0 or 3% CLA on the basis of metabolic bodyweight. Between 95 and 100 kg liveweight the gas exchange of each animal was measured over 48 h in a respiration chamber. This was preceded by a 5‐day quantitative determination of faeces and urine. The heat production and energy retention of the animals were determined by the carbon–nitrogen balance technique and the RQ method. Additionally several parameters of blood serum were determined. CLA had no effect on digestibility and metabolizability of energy, nor were gas exchange and heat production (HP) of the animals affected by the treatment. Average heat production across all measurements was 628 kJ/kg W0.75 in the control group and 636 kJ/kg W0.75 in the CLA group. No difference in fat deposition was observed (control 386, CLA group 379 kJ/kgW0.75). The N balance on the other hand was 2.2 g/day higher in the CLA‐supplemented animals. This caused retained protein energy, as a proportion of total energy retention, to increase from 24.2 to 25.8%. The ratio of retained protein energy to fat energy was 10% higher in the CLA‐treated animals than in the controls. Body mass increase over the trial as a whole was unaffected (control 27.6, CLA group 28.1 kg, equivalent to daily gains of 931 and 946 g, respectively). Blood serum lipoproteins exhibited few CLA‐induced changes. The greatest effects were observed in triacylglycerols, which were raised in the CLA animals compared with the controls. The ratio of cholesterol‐low‐density lipoprotein to cholesterol‐high‐density lipoprotein rose by 17% as a result of CLA treatment. Serum concentrations of albumin, creatinine, urea and glucose were unchanged. In conclusion, the study showed that supplementation with CLA under isoenergetic feeding conditions with a strongly positive energy balance has no marked effect on total metabolism, although a slightly enhanced deposition of protein was evident.  相似文献

Vitamin D deficiency in humans is widespread, and only a few food items are important natural sources of vitamin D. This study investigated the effect of UVB exposure of laying hens on the vitamin D content in egg yolk. In a two-factorial design, hens fed a vitamin D-deficient (-D) or -adequate (+D) diet were nonexposed or exposed to UVB light over a period of 4 weeks. UVB exposure of the -D group caused nearly normal egg production rate and egg shell quality; exposure of the +D group did not further improve these parameters. UVB exposure tended to improve the concentration of plasma 25-hydroxycholecalciferol (25(OH)D(3)), but had no effect on 1,25-dihydroxycholecalciferol in plasma or on cholecalciferol and 25(OH)D(3) in egg yolk. The present study shows that a short-term exposure of laying hens to UVB light is not an appropriate way to improve the vitamin D content of egg yolk.  相似文献

It has been shown that some dietary plant proteins beneficially influence lipid metabolism in animals. The effect of pea protein in this respect however has not yet been investigated. Therefore, we studied the effect of purified pea protein on the lipid metabolism in rats. Twenty-four rats received diets with either 200 g/kg of casein or purified pea protein for 16 days. Concentrations of triacylglycerols in liver, plasma and lipoproteins did not differ between both groups of rats. However, rats fed the pea protein diet had a lower concentration of total cholesterol in the liver and the very low density lipoproteins (VLDL) fraction than rats fed the casein diet (p < 0.05); cholesterol concentration in plasma, low density lipoproteins (LDL) and high density lipoproteins (HDL) did not differ between both groups. Rats fed pea protein moreover had an increased mRNA concentration of cholesterol-7α-hydroxylase in the liver and an increased amount of bile acids excreted via faeces compared with rats fed casein (p < 0.05). Concomitantly, mRNA concentrations of sterol regulatory element-binding protein (SREBP)-2 and its target genes 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase and LDL receptor in the liver were increased in rats fed pea protein (p < 0.05). The data of this study suggests that pea protein stimulates formation and excretion of bile acids, which leads to a reduced hepatic cholesterol concentration and a reduced secretion of cholesterol via VLDL. An increased gene expression of SREBP-2 and its target genes HMG-CoA reductase and LDL receptor may be a means to compensate for the increased loss of cholesterol for bile acid synthesis.  相似文献

This study was performed to assess the effects of potato protein and fish protein on concentrations of lipids in plasma and lipoproteins and the expression of genes involved in lipid metabolism in pigs used as an animal model. Therefore, 27 young male pigs with an average body weight of 22 kg were fed diets supplemented with protein extracted from potatoes (containing 849 g protein/kg dry matter), Alaska Pollack fillet as a source of fish protein (containing 926 g crude protein/kg dry matter) or casein which was used as control, for 3 weeks. Diets were formulated to supply identical amounts of each protein to the pigs by the three protein sources, namely 116 g/day in first week and 150 g/day in the second and third week. Pigs fed potato protein had lower concentrations of cholesterol in plasma and LDL than pigs fed casein (p < 0.05); no effect was observed on concentrations of HDL cholesterol and triglycerides. Pigs fed fish protein had lower cholesterol concentrations in plasma, LDL and HDL, and lower triglyceride concentrations in triglyceride-rich lipoproteins than pigs fed casein (p < 0.05). mRNA concentrations of genes involved in bile acid synthesis and cholesterol uptake were higher in pigs fed fish protein than in pigs fed casein (p < 0.05); no effect on these genes was observed in pigs fed potato protein. Expression of genes involved in lipogenesis and fatty acid oxidation was not altered by fish protein. In conclusion, this study shows that fish protein and potato protein lower plasma cholesterol concentrations in pigs. The hypocholesterolaemic effect of fish protein might be in part caused by a stimulation of bile acid synthesis; the reason for the hypocholesterolaemic effect of potato protein requires further elucidation.  相似文献
