We have developed an antibody detection enzyme-linked immunosorbent assay (ELISA) for the identification of animals infected by feline immunodeficiency virus (FIV). The ELISA solid-phase antigen consists of recombinant FIV gag proteins expressed in bacteria. The proteins are purified from bacterial lysates as insoluble inclusion bodies. In the case of bacterially expressed p24gag, it is shown that all of the linear, sequential epitopes presented by viral p24 during infection are retained. Purified preparations can be substituted for solid-phase whole virus in the IDEXX PetChektm immunoassay. The antibody ELISA duplicates the sensitivity and specificity of the whole virus based PetChek plate assay.  相似文献

The spotted-wing drosophila, Drosophila suzukii Matsumura, is an invasive pest causing significant damage to soft skinned fruits. Control of D. suzukii is critical since there is no tolerance for infested fruit in the market. While most insecticides control one or more D. suzukii life-stages (e.g., egg, larvae, and adult), the impact of insecticides that are toxic to immature stages  is unclear on the subsequent generation of a field population. Insecticides were applied at field recommended rates on cherries and blueberries in the laboratory to determine immature D. suzukii mortality. Spinetoram, cyantraniliprole, malathion, methomyl, spinosad, and phosmet resulted in relatively high mortality of all immature life stages. Zeta-cypermethrin, cyclaniliprole, and fenpropathrin resulted in lower mortality of egg and all larval instars. Malathion was also applied to lowbush blueberries with different fruit sizes (small, medium, and large) in the laboratory and there was no statistical difference in mortality rates depending on fruit sizes. Mortality data from the laboratory experiments were used to parameterize a refined D. suzukii population model. The model revealed that the timing and order of different insecticide classes are important to control D. suzukii population. Model runs that included early applications of more effective insecticides resulted in high immature mortality and greater reduction of D. suzukii populations compared to treatments applied later.

A feeding trial on yellow tail cichlid Pseudotropheus acei (Regan 1922) was undertaken to asses the effect of dietary Spirulina meal as a natural carotenoid source. Four experimental diets were formulated to progressively replace 0% (C), 2.5% (SP2.5), 5% (SP5) and 10% (SP10) of fish meal weight. Ten fish per tank (initial weight 3.75?±?0.02?g) were randomly distributed into twelve 80?l fiberglass tanks connected to a closed recirculation system (temperature 26.7?±?0.06°C). The diets were tested in triplicate for 12?weeks. The specific growth rate of fish fed all Spirulina diets were significantly higher compared to diet C. Feed intake (FI) tended to increase with dietary Spirulina level, and fish fed diet SP10 had significantly higher FI values compared to diet C. No significant differences in feed conversion ratio were observed among these groups. Although the protein efficiency ratio of fish fed diet C was lower than that of all Spirulina diets, no significant differences were observed among these groups (P?>?0.05). The total egg production and hatching rate (%) of eggs derived from all fish fed with Spirulina diets was significantly higher compared those from fish fed diet C. The yellow and blue coloration of yellow tail cichlid fed the diet containing Spirulina meal was enhanced and inclusion of dietary Spirulina meal was elevated carotenoids in skin. Results of the present study indicated that Spirulina meal has the potential to enhance the growth, reproductive performance and coloration on yellow tail cichlid.  相似文献

The European Non‐native Species in Aquaculture Risk Analysis Scheme (ENSARS) was used to assess one of the most popular aquaculture species in the world, the striped catfish Pangasianodon hypophthalmus (Sauvage, 1878), in two locations of southern Anatolia (Turkey). The overall mean risk score generated for P. hypophthalmus by the ENSARS Organism module (which assesses the risks of introduction, establishment, dispersal and impact) indicated that the species poses a medium risk under current climatic conditions. All other ENSARS modules rendered scores that indicated a moderately low risk under current climatic conditions. However, the risks of introducing novel diseases and the actual use (e.g. deliberate introduction to the natural habitats or food market) of the species both attracted scores indicating a medium risk. Confidence levels were medium or high for all modules except the Socio‐economic, with low confidence values also attributed to the risks for farming process of the organism, and its overall risk of spread into the wild during farming procedures and to marketing impacts. Recommendations are provided for further use of the ENSARS scheme, especially for a a priori assessment of potential aquaculture species in Turkey, where the sector has been remarkably developing in the last decades.  相似文献

The crucian carp Carassius carassius is a species of cyprinid fish native to south‐east England, but few studies exist on its growth and reproduction in England, and the species is threatened by introductions of its Asiatic congener, goldfish Carassius auratus. To increase knowledge of the crucian carp as a means of aiding its conservation, the present study assesses the growth (back‐calculated length at age, body condition) and reproduction (fecundity, egg size, length and age at maturity) of crucian carp in small ponds of Epping Forest (north‐east London, England).

Evaluation of growth and reproduction data with published data for populations from northern Europe (i.e. latitude ?50°) suggest that growth is very variable, even within the same area, and that data on reproductive indices, in particular length and age at maturity, are scarce. In England, the length at age trajectory of crucian carp living in sympatry with feral goldfish Carassius auratus did not differ significantly from allopatric crucian carp populations, but crucian carp body condition and relative fecundity (eggs per body weight) were highest, and mean age and standard length at maturity were lowest in sympatry with goldfish.
  • 3. These data suggest that somatic growth and reproductive output may be maximized in crucian carp when confronted by coexistence with feral goldfish. However, the potential impact of goldfish introductions on crucian carp growth and reproduction requires further study, involving a much larger number of crucian carp populations, both in allopatry and sympatry with feral goldfish populations.

Leukotrienes and lipoxins: structures, biosynthesis, and biological effects   总被引:94,自引:0,他引:94  
Arachidonic acid is released from membrane phospholipids upon cell stimulation (for example, by immune complexes and calcium ionophores) and converted to leukotrienes by a 5-lipoxygenase that also has leukotriene A4 synthetase activity. Leukotriene A4, an unstable epoxide, is hydrolyzed to leukotriene B4 or conjugated with glutathione to yield leukotriene C4 and its metabolites, leukotriene D4 and leukotriene E4. The leukotrienes participate in host defense reactions and pathophysiological conditions such as immediate hypersensitivity and inflammation. Recent studies also suggest a neuroendocrine role for leukotriene C4 in luteinizing hormone secretion. Lipoxins are formed by the action of 5- and 15-lipoxygenases on arachidonic acid. Lipoxin A causes contraction of guinea pig lung strips and dilation of the microvasculature. Both lipoxin A and B inhibit natural killer cell cytotoxicity. Thus, the multiple interaction of lipoxygenases generates compounds that can regulate specific cellular responses of importance in inflammation and immunity.  相似文献

A 12‐week feeding trial was conducted to estimate the effects of supplemented amino acids (lysine and methionine) with different dietary protein levels on growth, haematology and liver histology in meagre (Argyrosomus regius) at two different sizes: fingerling (15.2 g) and juvenile (44.4 g). Six practical diets were prepared, and three of these practical diets were formulated with 40 (P40), 44 (P44) and 48% (P48) crude protein without supplementation of amino acids. Other three diets having same protein levels were supplemented with lysine and methionine (P40AA, P44AA and P48AA) to bring these amino acids level up to that estimated to be at the least in the 48% protein diet. Final mean weight (FMW) and specific growth rate (SGR) of P48AA in fingerlings were higher than that of 40AA. According to FMW and SGR of juveniles, there was interaction found between dietary protein levels and the supplementation of amino acids. The FMW and SGR of meagre fed P44AA were higher than that of fish fed the other diets, except fish fed the P48. Economic profit index, packed cell volume and haemoglobin for juvenile meagre were significantly improved with the dietary protein level and amino acid supplementation. The liver tissues of the P48 group in both sizes displayed better structure compared with the other groups. In conclusion, dietary crude protein content of practical diets for juvenile meagre could be reduced from 48% to 44% by adding essential amino acids with an enhancement in growth, health and economic profit.  相似文献

TLR11 activation of dendritic cells by a protozoan profilin-like protein   总被引:1,自引:0,他引:1  
Mammalian Toll-like receptors (TLRs) play an important role in the innate recognition of pathogens by dendritic cells (DCs). Although TLRs are clearly involved in the detection of bacteria and viruses, relatively little is known about their function in the innate response to eukaryotic microorganisms. Here we identify a profilin-like molecule from the protozoan parasite Toxoplasma gondii that generates a potent interleukin-12 (IL-12) response in murine DCs that is dependent on myeloid differentiation factor 88. T. gondii profilin activates DCs through TLR11 and is the first chemically defined ligand for this TLR. Moreover, TLR11 is required in vivo for parasite-induced IL-12 production and optimal resistance to infection, thereby establishing a role for the receptor in host recognition of protozoan pathogens.  相似文献
